Project description:Total RNA in mesenteric LNs from chow- or high fat diet-fed Ldlr–/– mice were isolated using an RNeasy mini kit (Qiagen) with on-column DNaseI digestion. The DNA array was conducted using a Mouse Oligo chip 24k (TORAY Industries Inc.) and a 3D-Gene® Scanner (TORAY Industries Inc.) by Kamakura Techno-Science, Inc. (TORAY Industries Inc.).
Project description:To determine the different gene signatures between B lymphocytes from tumor draining lymph node (DLN) and normal lymph node (NLN), we have employed gene microarray as a discovery platform to identify gene signatures of tumor-educated B cells in DLN from tumor-bearing mice, taking NLN from normal mice as a control. We subcutaneously inoculated Balb/c mice with breast cancer cell line 4T1. Two weeks later, DLN was harvest and B cells were purified as descript in “treatment protocol”. From gene microarray, we found that B cells in DLN showed quite different transcript profiles from that in NLN.

Project description:Tingible body macrophages (TBMs) exist in lymph node B cell follicles and are responsible for clearing apoptotic B cells. Our data suggests that TBMs are of Cd169-lineage. We isolated cells from Cd169Cre/+.Tdtomatofl/fl reporter mice and transcriptionally profiled them to identify putative TBMs and chacterise their development and function relative to other LN cells of the Cd169 lineage.
