Project description:MSCs are a heterogeneous population and the specific population of MSCs may exhibit a selective ability for tissue repair. The aim of our research was to adapt the CD73+ subgroup of adipose derived MSCs (AD-MSCs) for the therapy of myocardial infarction (MI). Our results revealed that CD73+ AD-MSCs played more effective role in the acceleration function of cardiac recovery by promoting angiogenesis in a rat model of MI compared to mixed AD-MSCs and CD73- AD-MSCs. Microarray analysis shows differences between CD73+ and CD73- AD-MSCs when transcription profile of these two subgroups were compared, especially in VEGF pathway.

Aging-associated gene expression profiling in adipose tissue derived mesenchymal stem cells.

Project description:Background & Aims: Recently, energy metabolism in adipose tissue has been the subject of renewed interest, because significant energy expenditure can be induced in cells derived from white adipose tissue derived mesenchymal stem cells (Ad-MSCs) as beige adipocytes (BA), in addition to brown adipose tissue. Here we evaluated whether age-related changes in Ad-MSCs affect on impaired energy homeostasis and progressive obesity-related diseases. Materials and Methods: We isolated Ad-MSCs from subcutaneous murine fat tissues fed with control chow (C) or high-fat diet (60% fat; HF) for 12 or 24 weeks (younger or older groups, respectively). After two to three times passage, we performed the cellular characteristics analyses, which included differentiation property, flow cytometric and microarray analyses. Results: Ad-MSCs from older mice showed impaired differentiation capacity into BAs compared to the cells from younger mice. Global gene expression analysis by microarray revealed several genes involved with the browning of white adipose tissue were down-regulated according to aging. Additionally, HF feeding promoted aging-associated characteristic changes in Ad-MSCs. Conclusion: These results indicate the compensation potential of Ad-MSCs on energy homeostasis is affected by aging and dietary factors. A new approach targeting Ad-MSCs might represent a potential strategy for the prevention of obesity-related diseases.

In OA, the interest for mesenchymal stem cell (MSC)-EV therapeutic applications has increased. We have assessed the immunomodulary properties of adipose-derived MSCs (AD-MSCs) microvesicles (MV) and exosomes (EX) in interleukin (IL)-1β stimulated OA chondrocytes and cartilage explants and characterized them by mass spectrometry in order to uncover novel mediators in (AD-MSC)-EV immunomodulation.

PGC-1 alpha Mediates Differentiation of Mesenchymal Stem Cells toward Brown Adipose Cells

Project description:Mesenchymal stem cells (MSCs) are a multipotent cell type that can differentiate into non-hematopoietic cells, such as adipocytes. Adipocyte tissue is central to regulate energy balance. PGC-1 alpha controls several aspects of mitochondrial biogenesis. However, roles of PGC-1 alpha in brown fat differentiation of MSCs remain uncertain. To investigate roles of PGC-1 alpha in brown fat differentiation immortalized human MSCs were used for all experiments. The changes in genetic profiling between MSCs and PGC-1 alpha-expressing MSCs were analyzed by microarray analysis. The genetic profiling of PGC-1 alpha-expressing MSCs shows the significant increase of genes related to mitochondrial functions and lipid metabolism compared to that of MSCs. When expressed in MSCs, PGC-1 alpha activates a robust mitochondrial biogenesis and respiration. The expression of thermogenic markers, such as cytochrome C and complex II, was significantly increased in MSCs with treatment of adenovirus expressing PGC-1 alpha. Our microarray results also indicate that genetic pattern of PGC-1 alpha-expressing MSCs is very closed to that of adipose tissues. Bone marrow-derived MSCs were infected with Ad-GFP, or Ad-PGC-1? at a multiplicity of infection (m.o.i.) of 500 overnight.

Graphene Oxide Substrate Promotes Neurotrophic Factor Secretion and Survival of Human Schwann-Like Adipose Mesenchymal Stromal Cells - RNAseq

Project description:Mesenchymal stromal cells from adipose tissue (AD-MSCs) exhibit favourable clinical traits for autologous transplantation and can develop a ‘Schwann-like’ phenotype (sAD-MSCs) to improve peripheral nerve regeneration, where severe injuries yield insufficient recovery. However, sAD-MSCs regress without biochemical stimulation and detach from conduits under unfavourable transplant conditions, negating their paracrine effects. Graphene-derived materials support AD-MSC attachment, regulating cell adhesion and function through physiochemistry and topography. We report graphene oxide (GO) as a suitable substrate for human sAD-MSCs incubation towards severe peripheral nerve injuries, through evaluating transcriptome changes, neurotrophic factor expression over a 7-day period, and cell viability in apoptotic conditions. Transcriptome changes from GO incubation across four patients were minor compared to biological variance.

Two-condition experiments: AD-MSCs vs CB-HSCs, and AD-MSCs transfected with the combination of siR-EID1 and shR-EID1 vs AD-MSCs transfected with shR-EID1.
