Expression data from CD73 positive adipose derived mesenchymal stem cells (CD73+ AD-MSCs) and CD73 negative adipose derived mesenchymal stem cells (CD73- AD-MSCs) of mouse

ABSTRACT: MSCs are a heterogeneous population and the specific population of MSCs may exhibit a selective ability for tissue repair. The aim of our research was to adapt the CD73+ subgroup of adipose derived MSCs (AD-MSCs) for the therapy of myocardial infarction (MI). Our results revealed that CD73+ AD-MSCs played more effective role in the acceleration function of cardiac recovery by promoting angiogenesis in a rat model of MI compared to mixed AD-MSCs and CD73- AD-MSCs. Microarray analysis shows differences between CD73+ and CD73- AD-MSCs when transcription profile of these two subgroups were compared, especially in VEGF pathway.

Project description:Mesenchymal Stem Cells (MSCs), and Adipose-Derived Mesenchymal Stem Cells (AD-MSCs) have been used for many years in regenerative medicine for clinical and surgical applications. Additionally, recent studies reported improved respiratory activity after intravenous administration of MSCs into patients affected by coronavirus disease 2019 (COVID-19) caused by the Coronavirus 2 (SARS-CoV-2) suggesting their role as anti-viral therapy. Severe COVID-19 patients usually progress to acute respiratory distress syndrome, sepsis, metabolic acidosis that is difficult to correct, coagulation dysfunction, multiple organ failure, and even death in a short period after onset. Currently, there is still a lack of clinically effective drugs for such patients. The high secretory activity, the immune-modulatory effect, and the homing ability make MSCs and in particular AD-MSCs both a potential tool for the anti-viral drug-delivery in the virus microenvironment and potential cellular therapy. AD-MSCs as the most important exponent of MSCs are expected to reduce the risk of complications and death of patients due to their strong anti-inflammatory and immune-modulatory capabilities, which can improve microenvironment, promote neovascularization and enhance tissue repair capabilities. In this literature review, the role of regenerative strategies through MSCs, AD-MSCs, and adipocyte-secreted exosomal microRNAs (A-SE-miRs) as a potential antiviral therapy was reported, comparing the results found with current research progress on drugs and vaccines in COVID-19 disease.

Csf1+ AD-MSCs promote stroke repair by activating the resident microglia

Project description:We have employed a single cell sequencing approach using 10x Genomics scRNAseg to study the mechanism of AD-MSCs for treating the injured brain tissue. This study establishes a rat model of hemorrhagic stroke, followed by intravenous infusion of allogeneic AD-MSCs 24 hours post-model induction, in order to observe the therapeutic effect and analyze the underlying mechanism of action. Based on single-cell transcriptomic sequencing technology, we demonstrated that the re-infusion of AD-MSCs 24 hours after stroke mainly promotes the repair of injured brain tissue by activating brain tissue resident microglia and inhibiting bone marine-derived monocytes. Importantly, this role depends on the cell subsets of AD-MSCs that are highly expressing Csf1+. This study offers novel insights into the mechanisms underlying MSC-based stroke treatment and supports the potential for stable and efficacious MSC therapies.

Aging-associated gene expression profiling in adipose tissue derived mesenchymal stem cells.

Project description:Background & Aims: Recently, energy metabolism in adipose tissue has been the subject of renewed interest, because significant energy expenditure can be induced in cells derived from white adipose tissue derived mesenchymal stem cells (Ad-MSCs) as beige adipocytes (BA), in addition to brown adipose tissue. Here we evaluated whether age-related changes in Ad-MSCs affect on impaired energy homeostasis and progressive obesity-related diseases. Materials and Methods: We isolated Ad-MSCs from subcutaneous murine fat tissues fed with control chow (C) or high-fat diet (60% fat; HF) for 12 or 24 weeks (younger or older groups, respectively). After two to three times passage, we performed the cellular characteristics analyses, which included differentiation property, flow cytometric and microarray analyses. Results: Ad-MSCs from older mice showed impaired differentiation capacity into BAs compared to the cells from younger mice. Global gene expression analysis by microarray revealed several genes involved with the browning of white adipose tissue were down-regulated according to aging. Additionally, HF feeding promoted aging-associated characteristic changes in Ad-MSCs. Conclusion: These results indicate the compensation potential of Ad-MSCs on energy homeostasis is affected by aging and dietary factors. A new approach targeting Ad-MSCs might represent a potential strategy for the prevention of obesity-related diseases.

Two-condition experiments: AD-MSCs vs CB-HSCs, and AD-MSCs transfected with the combination of siR-EID1 and shR-EID1 vs AD-MSCs transfected with shR-EID1.

Single Cell Transcriptomic Analysis of Clinical-used MSCs Derived From Adipose Tissue, Bone Marrow, Placenta Membrane and Umbilical Cord

Project description:Here, we use single cell transcriptomics to characterize a dynamic senescence process of in vitro expanded MSC products from adipose tissue (AD), bone marrow (BM), placenta membrane (PM) and umbilical cord (UC). We found that cultured MSCs underwent progressive cell aging. As compared to perinatal MSCs (derived from PM and UC), adult MSCs (derived from AD and BM) showed higher degree of senescence and impaired immunosuppressive function. Moreover, we identified the transcriptional network regulating the intra-population functional diversity, which provides us guidance to drive reprogramming of determined MSCs lineage with specific cellular functions for a particular therapeutic intent.

Effects of human adipose tissue-derived and umbilical cord tissue-derived mesenchymal stem cells in a dextran sulfate sodium-induced mouse model

Project description:Background: Mesenchymal stem cells (MSCs) can be acquired from medical waste. MSCs are easily expanded and have multiple functions, including anti-inflammatory effects. We evaluated the effects of human adipose tissue-derived MSCs (AD-MSCs) and umbilical cord tissue-derived MSCs (UC-MSC) in a dextran sulfate sodium (DSS)-induced mouse model. Methods: Human AD-MSCs and UC-MSCs (1 × 106 cells) were injected intravenously into a 7-day DSS-induced colitis model. The therapeutic effects of cell origin, injection timing, and supernatants obtained from MSC cultures were evaluated. We also analyzed mRNA expression in MSCs, tissues, and intestinal flora. Results: AD-MSCs and UC-MSCs had strong anti-inflammatory effects when injected on day 3 in a mouse model. On day 11, mRNA levels of inflammatory factors in colon tissues were significantly decreased after injection of MSCs on day 3. Supernatants from MSCs culture decreased mRNA levels of tumor necrosis factor (Tnf)-α, but had reduced therapeutic effects compared with MSC cell injection. RNA sequencing using colon tissues obtained the day after cell injection revealed changes in the TNF-α/nuclear factor-κB and T-cell receptor signaling pathways. Additional analyses showed that several factors, including chromosome 10 open reading frame 54, stanniocalcin-1, and TNF receptor superfamily member 11b were increased in MSCs after adding serum from DSS colitis mice. Furthermore, both AD-MSCs and UC-MSCs maintained the balance of intestinal flora. Conclusion: AD-MSCs and UC-MSCs showed therapeutic effects against inflammation after early cell injection while maintaining the intestinal flora. Although supernatants showed therapeutic effects, cell injection was more effective against inflammation.
