Project description:Auxin treatment of grape (Vitis vinifera L.) berries delays ripening by inducing changes in gene expression and cell wall metabolism and could combat some deleterious climate change effects. Auxins are inhibitors of grape berry ripening and their application may be useful to delay harvest to counter effects of climate change. However, little is known about how this delay occurs. The expression of 1892 genes was significantly changed compared to the control during a 48 h time-course where the auxin 1-naphthaleneacetic acid (NAA) was applied to pre-veraison grape berries. Principal component analysis showed that the control and auxin-treated samples were most different at 3 h post-treatment when approximately three times more genes were induced than repressed by NAA. There was considerable cross-talk between hormone pathways, particularly between those of auxin and ethylene. Decreased expression of genes encoding putative cell wall catabolic enzymes (including those involved with pectin) and increased expression of putative cellulose synthases indicated that auxins may preserve cell wall structure. This was confirmed by immunochemical labelling of berry sections using antibodies that detect homogalacturonan (LM19) and methyl-esterified homogalacturonan (LM20) and by labelling with the CMB3a cellulose-binding module. Comparison of the auxin-induced changes in gene expression with the pattern of these genes during berry ripening showed that the effect on transcription is a mix of changes that may specifically alter the progress of berry development in a targeted manner and others that could be considered as non-specific changes. Several lines of evidence suggest that cell wall changes and associated berry softening are the first steps in ripening and that delaying cell expansion can delay ripening providing a possible mechanism for the observed auxin effects.

Project description:SuperSAGE is a method of digital gene expression profiling that allows isolation of 26bp tag fragments from expressed transcripts. Because its tag size is larger than that of conventional SAGE, SuperSAGE allowed a secure tag-to-gene annotation using BLAST search against grape genome databases.Transcript profiles in nine samples of grape berry tissues under different light conditions were obtained by SuperSAGE analysis and used for screening the genes which have co-ordinated transcript profiles with the change in the flavonoid composition in the samples analyzed. Candidate genes related to flavonoid biosynthesis and regulation were identified. Nine different grape samples, i.e., flowers, grape berries of Cabernet Sauvignon at 2, 7, 9 weeks after flowering (WAF), berry skins at 17 days after flowering (DAF) shaded after flowering, and berry skins at 17DAF shaded from flowering to 14DAF and then light exposed, were analyzed.

Project description:Basal leaf removal has been widely practiced to increase cluster sunlight exposure, control berry rot and eventually improve grape berry quality. Early leaf removal performed in cool region appeared to benefit the accumulation of norisoprenoids and monoterpenes. However, effects of this canopy management and leaf moving on volatile profiles and transcriptome of grape berry in warm region characterized with strong sunshine and arid climate were paid less attention. To cope with multiple possible effects caused by global warming, researches in warm region are urgently needed to provide a guide for adjusting the sunlight exposure treatment to adapt the climate change.In Manas county of Xinjiang province, sunlight exposure treatments performed in this study resulted in decreased β-carotene and lutein, which mainly responsible for the reduced norisoprenoids in ripening grapes. Substrate competition due to the up-regulation of VvTPS-a and VvNCED3 may contribute to the decreased concentration of monoterpenes in exposed berries. The notable increased C6 alcohols in the grape berries treated by leaf removal applied at veraison (LR-V), was mainly attribute to the enhanced substrate availability of linoleic acid and linolenic acid together with the higher expression of VvLOXO and VvADH1. Free C6 aldehyde was reduced by half leaf removal applied at veraison (HLR-V), leaf moving at veraison (LM-V) and leaf removal applied at berry pepper-corn size (LR-PS). Moreover, transcriptomic analysis indicated that both LR-V and LM-V treated grape berry implemented multiple stress-mitigation strategies to acclimate the improved sunlight exposure. Interestingly, photosynthesis-related genes in grape berry were primarily up-regulated by LR-V as a consequence of carbohydrate source removal rather than improved sunlight exposure. Weighted gene co-expression network analysis (WGCNA) suggested that the genes encoding malate synthase and 3-oxoacyl-(acyl carrier protein) reductase show a significant correlation with the accumulation of C6 alcohol.

Project description:The abscisic acid (ABA) increase and auxin decline are both indicators of ripening initiation in grape berry, and norisoprenoid accumulation also start at around the onset of ripening. To investigate the transcriptional and posttranscriptional regulation of the ABA and synthetic auxin 1-naphthaleneacetic acid (NAA) on norisoprenoid production, we performed time series GC-MS and RNA-seq analyses on Cabernet Sauvignon grape berries from pre-veraison to ripening. Higher levels of free norisoprenoids were found in ABA-treated mature berries in two consecutive seasons, and both free and total norisoprenoids were significantly increased by NAA in one season. The expression pattern of known norisoprenoid-associated genes in all samples and the up-regulation of specific alternative splicing isoforms of VviDXS and VviCRTISO in NAA-treated berries were predicted to contribute to it. Combined weighted gene co-expression network analysis (WGCNA) and promoter motif prediction analysis suggested that GATA26 and GATA28 could be potential regulators of norisoprenoid accumulation. Finally, the network analysis uncovered the interaction between previously identified switch genes, hormone-related genes and norisoprenoid-associated genes.

Project description:The vacuole occupies a large portion of plant cell volume, it is especially true to fruit tissues. Berry flesh cell vacuole serves as storage organelle for water, sugars, acids, secondary metabolites and others, which largely determining berry quality (Fontes et al., 2011a, b; Shiratake and Martinoia, 2007, Conde et al., 2006). However, the molecular basis of these compartmentation processes is still poorly understood. As in many species, the major bottle neck to study these aspects in grapevine is to obtain highly purified vacuoles with a good yield (Fontes et al., 2010). Up to date, several vacuole or tonoplast proteome researches were applied on a few plants mainly on Arabidopsis thaliana, vacuoles or tonoplast were derived from mesophyll cells (Carter et al., 2004, Endler et al., 2006, Schulze, et al., 2012) or cell culture (Jaquinod et al 2006, Shimaoka et al 2004), cauliflower buds (Schmidt et al., 2007) and sugar beet taproots (Jung et al., 2015). Though the grape berry protoplasts and intact vacuoles were successfully isolated from Cabernet Sauvignon berry suspension-cultured cells (Fontes et al., 2010), the vacuoles isolated from grape berry or different development and ripening stages of grape berry mesocarp tissues were not achieved.

Project description:The Deficiens homologue 9-iaaM (DefH9-iaaM) gene is an ovule-specific auxin-synthesizing gene which is expressed specifically in placenta/ovules and promotes auxin-synthesis. It was introduced into the genome of two grape cultivars Thompson Seedless and Silcora (SIL) and both transgenic cultivars had an increased number of berries per bunch. This study investigates the down-stream metabolic changes of Silcora and Thompson seedless grape cultivars when genetically modified through the insertion of the DefH9-iaaM gene into their genome. The effect of the genetic modification upon the grape metabolome was evaluated by 1H-NMR and exploratory data analysis and chemometrics methods. Interval Partial Least Squares regression (iPLS) and metabolite heatmaps were employed for scrutinizing the changes in the transgenic metabolome as compared to the wild type metabolome. The result shows that the pleiotropic effects on the metabolome as a function of the gene modifications is relatively low, but that the insertion of transgene caused a decrement in the malic acid and proline and an increment in the p-coumaric acid concentration. The results further show that the concentration of malic acid was correlated with the number of inserted copies of transgene in Silcora cultivar, proving that the inserted genes, which catered for increased berry quality and production, are made on the expense of a greater decrement in malic acid concentration in the berries. NMR together with chemometrics is able to identify specific metabolites that were up- or down regulated in the genetically engineered plants, and thus able to highlight alterations in the down-stream metabolic pathways due to the up-stream genetic modifications.

Project description:Changes in gene expression during berry development during a grape growing season were analysed. The effect on gene expression of different viticultural practises during grape berry development was investigated in this study by comparing two pruning methods (spur versus machine). Grape berries were collected and pooled on a weekly basis to obtain a developmental series comprised of 17 developmental stages from flowering until harvest across the grape growing season for both spur and machine pruned vines. Project description:We conducted transcript profiling and metabolome profiling induced by UV irradiation in grape berry skin. Transcriptome analysis was carried out with genome-wide microarray and two hundred thirty eight genes were more than 5-fold up-regulated by UV irradiation. The enrichment analysis showed GO terms including stilbene synthase (STS) gene. Moreover, the principal component analysis (PCA) of metabolome analysis showed a compound, identified resveratrol, accumulated in grape berry skin specifically. Our result clearly shows that UV irradiation induced only accumulation of resveratrol and its analogues but did not induce accumulation of the other phenolic compounds.
