Project description:Within a cell, proteins are in a dynamic state of turnover and are continuously synthesized and degraded. As an energetically expensive cellular process, protein turnover can have two opposing effects on maintaining a healthy proteome during the lifespan of an organism. Rapid protein turnover can replace old and damaged proteins with newly synthesized proteins. However, the high energetic demands of this process can potentially generate damaging reactive oxygen species that comprise the long-term health of the proteome. Thus, the relationship between aging, protein turnover kinetics and energetic demands of an organism remain unclear. Here, we used a proteomic approach to measure global rates of protein turnover within cultured fibroblasts isolated from a number of species with a wide range of lifespans. We show that organismal lifespan is negatively correlated with global rates of turnover. By further comparing cells from mice and naked mole rats (a short-lived and long-lived rodent species, respectively) we show that the latter has slower rates of turnover, lower levels of ATP production and reduced cellular ROS levels. Despite its slower rate of protein turnover, naked mole rat cells are able to tolerate protein misfolding stress more effectively than mouse cells. We suggest that in lieu of rapid constitutive protein turnover, long-lived species such as the naked mole rat have may have evolved more energetically efficient mechanisms for selective clearance of damaged proteins.

Project description:Mammals display wide range of variation in their lifespan. Investigating the molecular networks that distinguish long- from short-lived species has proven useful to identify determinants of longevity. Here, we compared the liver of long-lived naked mole-rats (NMRs) and the phylogenetically closely related, shorter-lived, guinea pigs using an integrated omic approach. We found that NMRs livers display a unique expression pattern of mitochondrial proteins that result in distinct metabolic features of their mitochondria. For instance, we observed a generally reduced respiration rate associated with lower protein levels of respiratory chain components, particularly complex I, and increased capacity to utilize fatty acids. Interestingly, we show that the same molecular networks are affected during aging in both NMR and humans, supporting a direct link to the extraordinary longevity of both species. Finally, we identified a novel longevity pathway and validated it experimentally in the nematode C. elegans.

Project description:The conserved Rap1 protein is part of the shelterin complex that plays critical roles in chromosome end protection and telomere length homeostasis. Previous studies addressed how fission yeast Rap1 contributes to telomere length maintenance, but the mechanism by which the protein inhibits end fusions has remained elusive. Here, we use a genetic screen in combination with high throughput sequencing to identify several amino acid positions in Rap1 that have a key role in end protection. Interestingly, mutations at these sites render cells susceptible to genome instability in a conditional manner with longer telomeres being prone to undergoing end fusions, while short telomeres are sufficiently protected. The protection of long telomeres requires their nuclear envelope attachment mediated by the Rap1-Bqt4 interaction. Our data demonstrates that longer telomeres pose an additional challenge for the maintenance of genome integrity and provides an explanation for a species-specific upper limit in telomere length.

Project description:Memory T cells provide immunity against pathogen reinvasion, but mechanisms of their long-term maintenance is unclear. Here we show that mice with the deletion of the transcription factor Foxo1 in activated CD8+ T cells had defective secondary but not primary responses to Listeria monocytogenes infection. Compared to short-lived effector T cells, memory precursor effector T cells expressed higher amounts of Foxo1 that promoted their generation and maintenance. Gene expression profiling and chromatin immunoprecipitation sequencing experiments revealed the chemokine receptor CCR7 and the transcription factor TCF1 as novel Foxo1-bound target genes with critical functions in memory T cell trafficking and transcriptional regulation. These findings demonstrate that Foxo1 is selectively incorporated into the genetic program that regulates memory but not effector CD8+ T cell responses to infection. CD8+ T cells were isolated from wild-type or Foxo1tagBirA mice in which Foxo1 is endogenously biotinylated. Foxo1 binding targets in CD8+ cells were identified by using Foxo1 antibody- and Streptavidin- ChIP-Seq approaches.

Project description:Plasma cell differentiation is characterized by differentiation of progenitor cells into short-lived and long-lived compartments. Here we examine how gene expression and nuclear architecture are linked to instruct short-lived and long-lived plasma cell fate. We find that plasma cell commitment is concurrent with the nuclear repositioning of transcription start sites associated with activation or repression of genes including the Irf4 and Bcl6 loci, respectively. Intergenic regions associated with the Ebf1 and Prdm1 loci also reposition concomitant with altered patterns of gene expression. Plasma cell differentiation is also enriched for inter-chromosomal associations concurrent at a global scale with alterations in gene expression. Finally, we find that the onset of plasma cell development is associated with a gain in euchromatic strength for genes encoding for cell cycle regulation and DNA repair that characterize in part the long-lived plasma cell fate. We conclude that at the onset of plasma cell development different strategies are applied to modulate genes that dictate short-lived and long-lived plasma cell gene programs.

Project description:The objective of the present study is to investigate gene regulation in developing fetal brain in congenic or inbred mice strains that differ in longevity. Gene expression and alternative splice variants were analyzed in a genome-wide manner in the fetal brain of C57BL/6J mice (long-lived), and compared to that of a congenic strain (short-lived) or an inbred strain AKR/J (short-lived) on days 12, 15 and 17 of gestation. The analysis showed a contrasting gene expression pattern during fetal brain development in these mice. Genes related to brain development, aging and spliceosome were significantly differentially regulated in the fetal brain of the short-lived mice when brain developed from d15 to d17. A significantly reduced number of splice variants were observed on d15 compared to d12 or d17 but in a strain-dependent manner.
