Project description:Muscle is a highly adaptable tissue capable of undergoing environmentally-induced changes and remarkable evolutionary adaptations. Muscle atrophy, a common feature in various myopathies as well as other conditions like cancer and diabetes, has been studied at the transcriptomic level, uncovering shared molecular factors. The lesser Egyptian jerboa, adapted for bipedal locomotion in a desert environment, experiences natural muscle loss in its foot. Cellular characterization of this process revealed similarities with human skeletal muscle atrophy, such as the loss of Desmin and upregulation of the ubiquitin proteasome pathway. By using RNA-sequencing, gene candidates associated with muscle loss in jerboa were identified, and genome-wide comparisons with established mouse models of muscle dystrophy/atrophy elucidated shared molecular signatures. These findings provide valuable insights into the genes and pathways involved in natural muscle loss and contribute to understanding the evolutionary relationship between natural and pathological muscle loss.

Project description:In this study, we uncover the molecular mechanism that PPARD transactive-deficient pigs have large-eared feature. Evidently from this work, PPARD plays a vital role on inhibiting cartilage growth by regulating the transcription of critical genes for chondrogenesis in ligand dependent manner; the activation of sPPARD speeds up apoptosis in CSPCs, terminal differentiation of chondroblasts and matrix degradation in auricular cartilage. The transactive-deficient PPARD has not the capability to slow down cartilage growth and consequently causes that ear size of carried animals increases at a high growth rate. As the role of PPARD in cartilage is similar in pigs, mice and humans, and since cartilage development is well-conserved in animals, our data helps to better understanding of cartilage development and treatment of cartilage-related diseases.

Project description:Osteoarthritis (OA) of spine (facet joints; FJ) is one of the major causes of severe low back pain and disability worldwide. The degeneration of facet cartilage is a hallmark of FJ OA. However, endogenous mechanisms that initiate degeneration of facet cartilage are unknown and there are no disease-modifying therapies to stop FJ OA. In this study we performed microRNA array analysis to identify differentially expressed microRNAs in facet cartilage from patients with FJ OA compared to facet cartilage from patients with lumbar disc herniation (LDH).

Project description:Hox genes control domain specific cartilage pattern formation along the proximodistal axis. Hoxa11 and Hoxd11 (Hox11) show overlapping expression in the mesenchyme of the zeugopod and regulate cartilage growth and differentiation. Hoxa13 and Hoxd13 (Hox13) are expressed in the autopodal mesenchyme and are crucial for the autopodal cartilage patterning. Since both Hox11 and Hox13 are involved in the limb cartilage development, they may share the target genes. To identify common target genes among the Hoxa11 and Hoxa13, we performed ChIP-Seq analysis using specific antibody.

Project description:Cartilage originates from mesenchymal cell condensations that differentiate into chondrocytes of transient growth plate cartilage or permanent cartilage of the articular joint surface and trachea. MicroRNAs fine-tune the activation of entire signaling networks and thereby modulate complex cellular responses, but so far only limited data are available on miRNAs that regulate cartilage development. Here we characterize an miRNA which promotes the biosynthesis of a key component in the RAF/MEK/ERK pathway in cartilage. Specifically, by transcriptome profiling we identified miR-322 to be upregulated during chondrocyte differentiation. Among the various miR-322 target genes in the RAF/MEK/ERK pathway, only Mek1 was identified as a regulated target in chondrocytes. Surprisingly, an increased concentration of miR-322 stabilizes Mek1-mRNA to arise protein levels and dampen ERK1/2 phosphorylation, while cartilage-specific inactivation in mice linked the loss of miR-322 to decreased MEK1 levels and increased RAF/MEK/ERK pathway activation. Such mice died perinatally due to tracheal growth restriction and respiratory failure. Hence, a single miRNA can stimulate the production of an inhibitory component of a central signaling pathway to impair cartilage development.

Project description:To identify genes that maintain the homeostasis of the articular cartilage, we compared gene expression profiles of adult articular cartilage chondrocytes with that of growth plate cartilage chondrocytes in adult (10-week-old) Sprague Dawley (SD) rats. Furthermore, to identify genes that have a potency to regenerate the articular cartilage, we compared gene expression profiles of superficial layer chondrocytes of infant epiphyseal cartilage which form articular cartilage with that of the deep layer chondrocytes which form growth plate cartilage in infant (6-day-old) SD rats.
