Project description:We compared the transcriptomic response of polarized microspore stage tomato anthers to long-term mild heat (LTMH-) stress of wild-type and three lines that display increased pollen thermo-tolerance. Our results indicated distinct differences between the thermo-tolerant lines and wild-type, suggesting a dampened response to LTMH in the tolerant lines than in wild-type.

Project description:In order to better understand chemical hybridizing agent (CHA) SQ-1-induced pollen abortion in wheat better, comparative proteomic analyses were conducted. A total of 61 proteins showed statistically significant differences in abundance, among which, 18 proteins were highly abundant and 43 proteins were less abundant in CHA-SQ-1 treated plants. Here, 60 proteins were successfully identified using MALDI-TOF/TOF mass spectrometry. These proteins were found to be involved in pollen maturation. Interactions between these proteins were predicted using bioinformatics analysis. Gene ontology and pathway analyses revealed that the majority of the identified proteins were involved in diverse biological processes. These results provide information for the molecular events (carbohydrate metabolism and energy metabolism) underlying CHA-SQ-1-induced pollen abortion and may serve as an additional guide for practical hybrid breeding.

A RALF-like peptide EaF82 derived from Epipremnum aureum inhibits pollen development in Arabidopsis and tobacco

Project description:To investigate the function of previously cloned novel cysteine-rich peptide EaF82 which biochemical properties are similar to the Rapid Alkalinization Factor (RALF), we overexpressed EaF82 gene in Arabidopsis and found defects only in the pollen development. To further determine the affected genes and biological pathways, we compared the transcriptome profilings of two independent Arabidopsis EaF82 overexpressing lines (TA and TB) with that of the vector control line (C). The RNAseq analysis results revealed decreased expressions of five endogenous clade IV AtRALFs and genes responsible for cell wall modifications and pollen maturation, supporting the observed pollen abortion.

Project description:Purpose:The molecular mechanism of pollen abortion in interspecific hybrids of B.rapa and B.nigra was revealed by cytological and transcriptional analysis Methods:floral buds (contain two developmental progress,1.1-1.5 mm and 1.5-6.0 long floral buds) were collected, then Separated male organs were kept in liquid nitrogen immediately until use. Total RNA was extracted using the TRIzol reagent (Invitrogen, Waltham, MA, USA). DNase (Promega, USA) was used to remove potential DNA contamination. For the quantitative real-time polymerase chain reaction (qRT-PCR) analysis Results:In this study, there were extensive variations in gene expression patterns of this hybrid, and down-regulated expression of key genes for pollen development, such as meiosis of pollen mother cells, DNA damage repair and pollen sperm cell differentiation, may lead to pollen abortion. Styrene-propyl synthesis and wax biosynthesis pathway were blocked, which affected pollen wall formation. Conclusions:The wide variation of genes showed that the expression of A and B genomes were affected differently. B genome tended to downregulate the expression of key genes during meiosis and mitosis, while A genome tended to downregulate the expression of genes related to stress and pollen wall formation. The results showed that the development of male gametophyte was seriously affected after synthesis.

Melatonin Alleviates High Temperature-Induced Pollen Abortion in Solanum lycopersicum.

Project description:Melatonin is a pleiotropic signal molecule that plays critical roles in regulating plant growth and development, as well as providing physiological protections against various environmental stresses. Nonetheless, the mechanisms for melatonin-mediated pollen thermotolerance remain largely unknown. In this study, we report that irrigation treatment with melatonin (20 µM) effectively ameliorated high temperature-induced inactivation of pollen and inhibition of pollen germination in tomato (Solanum lycopersicum) plants. Melatonin alleviated reactive oxygen species production in tomato anthers under high temperature by the up-regulation of the transcription and activities of several antioxidant enzymes. Transmission electron micrograph results showed that high temperature-induced pollen abortion is associated with a premature degeneration of the tapetum cells and the formation of defective pollen grains with degenerated nuclei at the early uninuclear microspore stage, whilst melatonin protected degradation of organelles by enhancing the expression of heat shock protein genes to refold unfolded proteins and the expression of autophagy-related genes and formation of autophagosomes to degrade denatured proteins. These findings suggest a novel function of melatonin to protect pollen activity under high temperature and support the potential effects of melatonin on reproductive development of plants.

Expression data from Arabidopsis pollen and semi in vivo- and in vitro-grown pollen tubes.

Project description:Pollen tubes extend through pistil tissues and are guided to ovules where they release sperm for fertilization. Although pollen tubes can germinate and elongate in a synthetic medium, their trajectory is random and their growth rates are slower compared to growth in pistil tissues. Furthermore, interaction with the pistil renders pollen tubes competent to respond to guidance cues secreted by specialized cells within the ovule. The molecular basis for this potentiation of the pollen tube by the pistil remains uncharacterized. We used a surgical procedure to obtain large quantities of uncontaminated pollen tubes that grew through the pistil and defined their transcriptome by microarray analysis. We also characterized the transcriptome of in vitro-grown pollen tubes (for 0.5hours or 4hours) and dessicated mature pollen in Arabidopsis.
