Project description:Osteoarthritis is a multifactorial disease in which not only physical tissue damage but also persistent inflammation is thought to be one of the main causes. The qualitative differences between mechanical and inflammatory cytokine stimulation in the production of OA-related molecules are not fully understood, and the biological events that occur in cells when the two stimuli are applied simultaneously remain unresolved. 　Articular cartilage tissue is formed by relatively sparse cells surrounded by a rich extracellular matrix composed mainly of collagen and proteoglycans, and three-dimensional culture using hydrogels such as collagen gel has been frequently used to reproduce chondrocytes that are three-dimensionally bound to the surrounding extracellular matrix and to evaluate physiological responses. 　In this study, we aimed to compare the responses of 3D cultured articular chondrocytes to mechanical stimuli and inflammatory cytokine stimuli, to analyze the responses to the two stimuli when they occur simultaneously, and to gain insight into the molecular mechanisms involved. Three-dimensional culture of human articular chondrocytes was performed using a stiff atelocollagen sponge and atelocollagen gel, and analysis focused on changes in gene expression due to cyclic compression stimulation.

Cyclic compressive loading induces a mature meniscal cell phenotype in mesenchymal stem cells with an atelocollagen-based scaffold

Project description:we investigated how cyclic compressive loading (CCL) could impact MSCs using three-dimensional cultures in atelocollagen-based meniscal substitute (ACMS). We extracted MSCs from the meniscus, synovium, and articular cartilage, cultured them in three-dimensional cultures, and submitted to CCL for seven days. We then compared the transcriptomes of MSCs treated with and without CCL. Our RNA-seq analysis revealed that CCL induced significant transcriptome changes, significantly affecting chondrocyte-related genes, including SOX9, TGFB1, and PRG4 upregulation. CCL-induced transcriptional differentiation from meniscus progenitors toward mature meniscal cells.

Project description:Objective: microRNAs (miRs) are small non-coding molecules that regulate post-transcriptional target gene expression and are involved in mechano-regulation of cellular activities in all physiological systems. It is unknown whether such epigenetic mechanisms are regulated in response to increasing magnitudes of load. This study investigated mechano-regulation of miRs in articular cartilage subjected to ‘physiological’ and ‘non-physiological’ compressive loads in vitro as a model system and validated findings in an in vivo model of abnormal joint loading. Design: Bovine full-depth articular cartilage explants were loaded to 2.5MPa (physiological) or 7MPa (non-physiological) (1Hz, 15 minutes) and mechanically-regulated miRs identified using Next Generation Sequencing and verified using quantitative PCR. Downstream targets were verified using miR-specific mimics or inhibitors in conjunction with 3’-UTR luciferase activity assays. Results: A subset of miRs were mechanically-regulated in ex vivo cartilage explants and in vivo joint cartilage. miR-221, miR-222, miR-21-5p, and miR-27a-5p were increased and miR-483 levels decreased with increasing load magnitude. Tissue Inhibitor of Metalloproteinase 3 (TIMP3) and Cytoplasmic Polyadenylation Element Binding Protein 3 (CPEB3) were identified as putative downstream targets. Conclusion: Our data confirms miR-221 and -222 mechano-regulation and demonstrates novel mechano-regulation of miR-21-5p and miR-27a-5p in ex vivo and in vivo cartilage loading models. TIMP3 and CPEB3 are putative miR targets in chondrocytes. Identification of specific miRs that are regulated by increasing load magnitude, and their potential to impact on tissue homeostasis, has direct relevance to other mechano-sensitive physiological systems and provides a mechanism by which load-induced tissue behaviour is regulated, both in health and pathology.

Differential gene expression of static and intermittent compressive force treated human periodontal ligament cells

Project description:Mechanical force has been shown to regulate periodontal ligament cells (PDLs) behaviors. However, different force types lead to distinct PDLs’ responses. Here, the differential gene expression profiling of PDLs subjected to static and intermittent compressive forces was examined using RNA sequencing technique. Results demonstrated that the static and intermittent compressive force treated PDLs exhibited the differential regulation genes. KEGG pathway enrichment analysis revealed that focal adhesion and transforming growth factor beta signaling pathway were commonly upregulated while calcium signaling pathway was downregulated in both static and intermittent compressive force-treated PDLs. Interestingly, Wnt signaling pathway was upregulated only in the PDLs that subjected to the intermittent compressive force.
