Project description:AimsPrevalence of prediabetes and type 2 diabetes mellitus(T2DM) are increasing worldwide. Key lncRNAs were detected to provide a reference for searching potential biomarkers of prediabetes and T2DM in hypertriglyceridemia patients.MethodsThe study included 18 hypertriglyceridemia patients: 6 newly diagnosed type 2 diabetes patients, 6 samples with prediabetes and 6 samples with normal blood glucose. Weighted gene co-expression network analysis (WGCNA) was conducted to construct co-expression network and obtain modules related to blood glucose, thus detecting key lncRNAs.ResultsThe green, yellow and yellow module was significantly related to blood glucose in T2DM versus normal controls, T2DM versus prediabetes, prediabetes versus normal controls, respectively. ENST00000503273, ENST00000462720, ENST00000480633 and ENST00000485392 were detected as key lncRNAs for the above three groups, respectively.ConclusionsFor hypertriglyceridemia patients with different blood glucose levels, ENST00000503273, ENST00000462720 and ENST00000480633 could be potential biomarkers of T2DM.

Identified lncRNAs functional modules and genes in prediabetes with hypertriglyceridemia by weighted gene co-expression network analysis.

Project description:BackgroundHypertriglyceridemia (HTG) is one of the most important comorbidities in abnormal glucose patients. The aim of this study was to identify lncRNAs functional modules and hub genes related to triglyceride (TG) in prediabetes.MethodsThe study included 12 prediabetic patients: 6 participants with HTG and 6 participants with normal triglyceride (NTG). Whole peripheral blood RNA sequencing was performed for these samples to establish a lncRNA library. WGCNA, KEGG pathways analysis and the PPI network were used to construct co-expression network, to obtain modules related to blood glucose, and to detect key lncRNAs. Meanwhile, GEO database and qRT-PCR were used to validate above key lncRNAs.ResultsWe found out that the TCONS_00334653 and PVT1, whose target mRNA are MYC and HIST1H2BM, were downregulating in the prediabetes with HTG. Moreover, both of TCONS_00334653 and PVT1 were validated in the GEO database and qRT-PCR.ConclusionsTherefore, the TCONS_00334653 and PVT1 were detected the key lncRNAs for the prediabetes with HTG, which might be a potential therapeutic or diagnostic target for the treatment of prediabetes with HTG according to the results of validation in the GEO database, qRT-PCR and ROC curves.

MicroRNA Expression Profiles Analysis in Mouse Pancreatitis Tissue With or Without Hypertriglyceridemia

Project description:The crucial roles of miRNAs have been implicated in various pathological processes, including pancreatitis. However, roles of miRNAs in hypertriglyceridemia-associated pancreatitis have not been reported. Pancreatitis tissue from mice with or without hypertriglyceridemia were analyzed for microRNA profiling. Using qPCR verification, we further identified miR-153 as the most upregulated miRNA in mice with hypertriglyceridemia. This work provide potential therapeutic targets for the treatment of hypertriglyceridemia-associated pancreatitis.

Prediabetes is associated with musculoskeletal dysfunction independent of total body adiposity

Project description:It has been proposed that there is a direct pathophysiological link between metabolic dysfunction and musculoskeletal degeneration in people with obesity but direct evidence for this proposition is lacking. We performed a comprehensive assessment of musculoskeletal status in people with obesity and prediabetes and a control group of sex-, age- and adiposity-matched participants with normoglycemia. We find muscle mass and bone mineral density scores, muscle growth markers, myofiber size, myofiber capillarization, and muscle type 2 macrophage content are lower, and intramyocellular lipid, but not inter- or intra-muscular adipose tissue, content are higher in people with prediabetes compared with the control group. Moreover, muscle strength during repeated maximum voluntary contractions declines faster in people with prediabetes. These findings demonstrate a close interrelationship between metabolic and musculoskeletal dysfunction in people with obesity and have direct clinical implications, because greater muscle fatigability in people with prediabetes may present a barrier to physical activity.

Next generation sequencing of long non-coding RNAs in diabetes mellitus rats after sleeve gastrectomy

Project description:Sleeve gastrectomy (SG) could improve diabetes mellitus dramatically. However, the mechanisms remain largely undetermined. With the advancement of in-depth bioinformatic analysis, non-coding RNAs especially long non-coding RNAs(lncRNAs), have been implicated in many biological processes. To explore whether lncRNAs mediate the amelioration of diabetes mellitus condition after SG, we subjected liver samples of diabetic rats after SG and shame operations for RNA sequencing. Further, interaction network between lncRNAs and mRNAs/microRNAs(miRNAs) were constructed to infer the function of lncRNAs in diabetic remission, followed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses. Through RNA sequencing, we identified more than 9000 lncRNAs with a dramatically differentially expressed profile of 147 lncRNAs. Some of these specific lncRNAs were only expressed in diabetic rats after SG, which may be promising biological markers of diabetic remission and SG prognosis. Through co-analysis between lncRNAs and mRNAs, 20 potential target genes of lncRNAs were screened out. Further bioinformatic analysis indicated the differentially expressed lncRNAs harbored various miRNAs binding sites and three representative lncRNA-miRNA networks of lnc50 and lnc136 were constructed. The KEGG analysis revealed that differential expressed lncRNAs were mainly related to mitogen-activated protein kinase (MAPK) pathway. These results demonstrated that SG dramatically altered expression profile of lncRNAs on diabetes mellitus, and altered lncRNAs expression may contribute to the improvement on diabetic remission through MAPK pathway by interaction with mRNA/miRNA. Our findings provide valuable resources for molecular mechanism of diabetic remission after SG and is benefit for development of lncRNAs-based diagnostic and therapeutic interventions of diabetes mellitus.

Cardiac miRNA expression changes induced by prediabetes

Small RNA sequencing analysis was applied to explore the expression changes resulted high-fat chow (supplemented with 40% lard) diet for 21 weeks and a single low dose of streptozotocin (20 mg/kg) treatment at week 4. Long-Evans rats were used in the present study. Left ventricular samples(n=6) of both prediabetic and control groups were analyzed.

Nicotinamide mononucleotide increases muscle insulin sensitivity in women with prediabetes

Project description:Studies in rodents have shown obesity and aging impair tissue nicotinamide adenine dinucleotide (NAD+) biosynthesis, which contributes to metabolic dysfunction. The availability of nicotinamide mononucleotide (NMN) is an important rate-limiting factor in mammalian NAD+ biosynthesis. We conducted a 10-week, randomized, placebo-controlled, double-blind trial to evaluate the effect of NMN supplementation on metabolic function in 25 postmenopausal women with prediabetes who were overweight/obese. Insulin-stimulated glucose disposal, assessed by using the hyperinsulinemic-euglycemic-clamp procedure, increased by 25±7% (P<0.01) in the NMN group, which was accompanied by an increase in insulin-stimulated phosphorylation of muscle AKT (P<0.01), whereas neither outcome changed after placebo treatment. Body composition (fat mass, fat-free mass, intra-abdominal fat, intrahepatic triglyceride content) and muscle mitochondrial respiratory capacity did not change after treatment with placebo or NMN. These results demonstrate NMN improves muscle insulin sensitivity in women with prediabetes who are overweight/obese, independent of changes in body composition or mitochondrial function.
