Project description:The activation status of some DNA damage repair proteins is closely related to the efficacy of CDDP chemotherapy in MIBC. In this study, we used CRISPR/cas9 gene-editing technology to construct hMSH2-deficient bladder cancer cells and confirmed that loss of hMSH2 decreased the sensitivity of bladder cancer cells to cisplatin. Transcriptome analysis was performed to find the potential genes that could be regulated by hMSH2.

miR-200b is associated with cisplatin sensitivity in bladder cancer cells

Project description:We aimed to clarify the role of miR-200b in cisplatin (CDDP) sensitivity in bladder cancer (BCa). CDDP resistant T24 cells (T24RC) were transfected with a miR mimic negative control (NC) or a miR-200b mimic, after which cells were treated with or without CDDP. We found that ectopic miR-200b expression re-sensitized the T24RC cells to CDDP. Gene expression microarray analysis revealed that the combination of miR-200b and CDDP affected genes involved in CDDP sensitivity and cytotoxicity.

Project description:Circular RNAs (circRNAs) have been increasingly indicated to be important participants in the development and progression of various malignant tumors. In this study, we analyzed circRNA high-throughout sequencing from human bladder cancer tissues and paired adjacent normal tissues. Then we focused on the upregulated circRNAs and identified a novel circular RNA, hsa_circ_0008399 , as a new candidate circRNA derived from RBM2 gene. FISH assay has been proved that hsa_circ_0008399 mainly located in nucleus. Transcriptome analysis was performed to find the potential genes that could be regulated by hsa_circ_0008399.

Project description:Cdk4/6 inhibitors have shown to increase overall survival in hormone-positive breast tumors, but whether other solid tumors could respond to these inhibitors has not yet defined. Here we show that Palbociclib (a Cdk4/6 specific inhibitor in clinic use) + cisplatin (CDDP, a chemotherapeutic agent most active in locally or advanced bladder cancer patients) treatment exerts antiproliferative effects in vivo using a bladder cancer mouse model.
