Project description:The microbial loop is the conventional model by which nutrients and minerals are recycled in aquatic eco-systems. Biochemical pathways in different organisms become metabolically inter-connected such that nutrients are utilized, processed, released and re-utilized by others. The result is that unrelated individuals end up impacting each others’ fitness directly through their metabolic activities. This study focused on the impact of programmed cell death (PCD) on a population’s growth as well as its role in the exchange of carbon between two naturally co-occurring halophilic organisms. Flow cytometric, biochemical, 14C radioisotope tracing assays, and global transcriptomic analyses show that organic algal photosynthate released by Dunalliela salina cells undergoing PCD complements the nutritional needs of other non-PCD D. salina cells. This occurs in vitro in a carbon limited environment and enhances the growth of the population. In addition, a co-occurring heterotroph Halobacterium salinarum re-mineralizes the carbon providing elemental nutrients for the mixoheterotrophic chlorophyte. The significance of this is uncertain and the archaeon can also subsist entirely on the lysate of apoptotic algae. PCD is now well established in unicellular organisms; however its ecological relevance has been difficult to decipher. In this study we found that PCD in D. salina causes the release of organic nutrients such as glycerol, which can be used by others in the population as well as a co-occurring halophilic archaeon. H. salinarum also re-mineralizes the dissolved material promoting algal growth. PCD in D. salina was the mechanism for the flow of dissolved photosynthate between unrelated organisms. Ironically, programmed death plays a central role in an organism’s own population growth and in the exchange of nutrients in the microbial loop.

We compare the transcriptional changes induced in the human epithelial cell line, Caco-2 after co-culture with Lactobacillus rhamnosus GG or a consortium of Lactobacillus rhamnosus GG and Bacteroides caccae.

Project description:Bioelectrochemical systems employing mixed microbial communities as biocatalysts are gaining importance as potential renewable energy, bioremediation, or biosensing devices. While we are beginning to understand how individual microorganism species interact with an electrode as electron donor, not much is known about the interactions between different microbial species in a community. Here, we compare the bioelectrochemical performance of Shewanella oneidensis in a pure-culture and in a co-culture with the homolactic acid fermenter Lactococcus lactis. While S. oneidensis alone can only use lactate as electron donor for current production, the co-culture is able to convert glucose into current with a similar coulombic efficiency of approximately 17%, respectively. With (electro)-chemical analysis and transcription profiling, we found that the BES performance and S. oneidensis physiology were not significantly different whether grown as a pure- or co-culture. These co-culture experiments represent a first step in understanding microbial interactions in BES communities with the goal to design complex microbial communities, which specifically convert target substrates into electricity. Further, for the first time, we elucidated S. oneidensis gene expression with an electrode as the only electron acceptor. The expression pattern confirms many previous studies regarding the enzymatic requirements for electrode respiration, and it generates new hypotheses on the functions of proteins, which are so far not known to be involved in electrode respiration.

Halobacterium NRC-1 was grown to mid-logarithmic phase (OD600 ~0.4 – 0.8) in MM2. Cells were harvested and washed in MM1 and incubated in MM1 (control) or in Dunaliella harvested supernatant. Cells were harvested at time zero, 5, 10, 20, 40, 80, 160 minutes. RNA from two biological replicate time courses were prepared, averages of these replicates are reported in the published study. The zero time point was harvested immediately after incubation Halobacteria-NRC1 in MM1 or in Dunaliella salina's supernatant.

Project description:In hypersaline brines, biodegradation of recalcitrant plant polymers can be inhibited by salt-induced microbial stress and/or caused by inadequate metabolic capabilities of extremely halophilic microbes. Therefore, woody materials can be well-preserved even in NaCl brines that are less biologically hostile than most other brines. Here, we considered whether the nanohaloarchaea, that live alongside (the related) haloarchaea, ever partake in the degradation of xylan, a major hemicellulose component of wood. Samples were taken from natural evaporitic brines and anthropogenic solar salterns located in various parts of Europe and Asia. We recently demonstrated that nanohaloarchaeon Ca. Nanohalobium constans lives as an ectosymbiont associated with the chitinolytic haloarchaeon Halomicrobium. Here, we describe an extremely halophilic xylan-degrading consortium with three members, where nanohaloarchaea act as ectosymbionts of Haloferax lucertensis, which in turn acts as a scavenger of xylan-degradation products, produced by a primary xylan hydrolytic Halorhabdus species. The two corresponding binary associations of nanohaloarchaea, Candidatus Nanohalococcus occultus SVXNc and Candidatus Nanohalovita haloferacivicina BNXNv and their hosts were obtained, stably cultivated and characterized. In contrast to the previously described association of chitinolytic haloarchaeon Halomicrobium and its amylolytic symbiont Ca. Nanohalobium, the host haloarchaea within the xylan-degrading consortium could metabolize α-glucans (glycogen and starch), and, thus, obtained no obvious trophic benefit from ectosymbionts. The current study has broadened the range of culturable ectosymbiontic nanohaloarchaea and demonstrates that they are an important ecophysiological component of polysaccharide-degrading halophilic microbial communities and can be readily isolated in binary co-cultures by using the appropriate enrichment strategy.

The BES was either operated with S. oneidensis alone, fed with lactate, or it was operated with S. oneidensis and L. lactis with glucose as primary substrate. The basic medium was a modified M4 medium containing 0.5 g/L yeast extract, 0.5 g/L trypton and 5 g/L glycerol phosphate, besides the commen M4 incredients. S. oneidensis oxidizes lactate to acetate and electrons in a BES - the latter generate a current at a graphite anode. The anode biofilm was harvested after about 4 weeks of continuous BES operation and subjected to total RNA extraction.
