Project description:We developed a thermogelling polymer with intrinsic anti-scarring properties on retinal cells in-vitro. To study its mechanism of action in-vivo, this thermogelling polymer was injected into the rabbit eye as a vitreous substitute and the retinal tissues were harvested for RNA-sequencing to understand transcriptomic changes induced by the polymer.

Project description:Human embryonic stem-cell derived retinal pigment epithelial cells were co-stimulated with TNF-α and TGF-β1 to model a retinal scarring complication, proliferative vitreoretinopathy. Thermogelling polymer prevented this scarring complication. To understand the polymer-cell interaction and ultimately the mechanism underlying it's anti-scarring activity, RNA-Seq was performed.

Dual-function thermogelling polymers as effective endotamponade for retinal detachment repair and reassembly of vitreous-like body in-situ

Project description:Endo-tamponade agents are crucial surgical adjuncts for vitreo-retinal surgery. Current clinical agents provide internal tamponade through buoyancy forces requiring the need for prolonged facing down post-operative positioning. Other disadvantages include temporary loss of vision, raised intra-ocular pressure, cataract formation and a need for an additional removal surgery. Therein lies an unmet clinical need to develop alternative biomaterials to overcome these key challenges. We describe a biocompatible and biodegradable thermogelling polymer which provides an internal tamponade effect through surface tension and viscosity. Furthermore, we demonstrate its surgical efficacy in the repair of retinal detachment in a non-human primate model. By three-month post-surgery, the thermogel undergoes biodegradation, and is replaced by a vitreous-like body, thereby mimicking the biophysical needs of the natural vitreous body. In summary, we demonstrate the potential superiority of our thermogel compared to current vitreous tamponade gold standards; and present a significant advancement in the use of biomaterials for vitreo-retinal surgery.

Protein composition of the subretinal fluid suggest selective diffusion of vitreous proteins in retinal detachment

Project description:Purpose: To study the proteome of the subretinal fluid (SRF) from rhegmatogenous retinal detachment (RRD) in search for novel markers for improved diagnosis and prognosis of RRD. Methods: Human undiluted SRF obtained during vitrectomy for primary RRD using a 41G needle (n = 24) was analyzed and compared to vitreous humor from 2-days post-mortem eyes (n = 20). Sample preparation underwent nanoflow liquid chromatography-tandem mass spectrometry (LC-MS/MS). Label-free quantification (LFQ) using MaxQuant was used to determine differential expressed proteins between SRF and vitreous humor. The intensity-based absolute quantification (iBAQ) was used to rank proteins according to their molar fractions within groups. Identification of proteins beyond the quantitative level was performed using the Mascot search engine. Results: The protein concentration of the control vitreous humor was lower and more consistent (1.2 ± 0.4 mg) than that of the SRF (17.9 ± 22 mg). The iBAQ analysis showed high resemblance between SRF and vitreous humor, except for crystallins solely identified in vitreous humor. The LFQ analysis found 37 protein misregulations between SRF and vitreous humor of which the blood coagulation pathway was found to be enriched using the PANTHER Classification System. Combined, the iBAQ, LFQ and Mascot analysis found an overlap only in chitinase-3-like protein 1 and galectin-3-binding protein unique to the SRF. Conclusions: The proteome of the SRF was highly represented by proteins involved in proteolysis. Such proteins can possibly serve as targets in modulating the effects of SRF in RD.

Project description:There have been reports of unexplained visual loss following intra-ocular silicone oil (SiO) tamponade in retinal detachment patients, yet the underlying mechanism is unknown. In the present study, we identified differentially expressed proteins in the vitreous humor of four groups (each group, n=3): pars plana vitrectomy (PPV) for rhegmatogenous retinal detachment (RD group), SiO removal after successful retinal reattachment (SO group), cataract surgery after successful retinal reattachment with sterilized air tamponade (FA group), and PPV for epiretinal membrane (ERM group). DIA was combined with two-dimensional liquid chromatography-tandem mass spectrometry (LC-MS/MS) to find expression changes in the proteome of vitreous.

Vitreous samples from Reeler (n=9; RELN-/-) and WT (n=9; wild-type RELN+/+) mice at different postnatal (14, 21, 28) days, were used. Some potential candidate proteins were analyzed by chip array and confirmed by conventional analysis.

Vitreous of proliferative diabetic retinopathy patients

Project description:We identified lncRNA expression profiles in vitreous samples between proliferative diabetic retinopathy (PDR) patients and idiopathic macular hole (IMH) patients, and between PDR patients who had received preoperative anti-vascular endothelial growth factor (anti-VEGF) therapy and PDR patients who had received surgery alone. There had been the systemic expression differences in vitreous at the microarray level from PDR patients and IMH patients, and from PDR patients after anti-VEGF treatment and untreated PDR patients.
