Project description:The objective of the present study is to investigate the effect of Nox4 overexpression on human brain endothelial cells. Comparative gene expression analysis was performed using cultured human brain microvascular endothelial cells with adenovirus-mediated LacZ or Nox4 overexpression. Gene expression was analyzed using 3D-Gene® Human 25k oligonucleotide microarrays and computational gene expression analysis tools. Nox4 overexpression resulted in significant changes in numerous genes related with (1) signaling pathways mediated by cytokines and chemokines, (2) stress responses, such as NFB activation, and (3) leukocyte chemotaxis and migration.

Brain microvascular endothelial cells: INSR KD

Project description:The insulin receptor (INSR) mediates insulin signaling to modulate cellular functions. Although INSR is expressed at the blood-brain barrier (BBB), its role in the modulation of BBB function is poorly understood. Therefore, in this study, we aimed to analyze the effect of INSR knockdown on the expression levels of functional proteins at the BBB. We established the INSR-knockdown cell line (shINSR) using human cerebral microvascular endothelial cells (hCMEC/D3). The cellular proteome was analyzed using quantitative proteomics. Protein expression levels were quantified by targeted proteomics.

mRNAseq of Huntington's disease and control patient iPSC-derived brain microvascular endothelial cells

Project description:Purpose: The goals of this study are to compare the transcriptomic profile (mRNA-seq) of HD and control patient iPSC-derived brain microvascular endotheial cells to identify alterations in gene expression. Methods: RNA were isolated from HD and control iPSC-derived brain microvascular endothelial cells. mRNAseq using Illumina TruSeq mRNA PolyA+ v2 lib prep and HiSeq 2500. Statistical difference in mRNA levels were calculated with subsequent GO and pathway analysis. Results: mRNAseq and statistical analysis revealed differentially expressed genes between HD and control iPSC-derived brain microvascular endothelial cells. Conclusions: Our study shows differentially expressed genes between HD and control iPSC-derived brain microvascular endothelial cells, and reveals gene networks that are relevant to the mechanism of HD pathogenesis.

Cytotoxic and anti-Toxoplasma gondii activities of monensin on human brain microvascular endothelial cells

Project description:The aim of the present work was to investigate the effect of monensin on the in vitro growth of T. gondii tachyzoites and on the host cells (human brain microvascular endothelial cells - hBMECs). The hypotheses were that (1) inhibition of the WNT signalling pathway by monensin can reduce the growth of T. gondii infecting human brain microvascular endothelial cells (hBMECs) and (2) by suppression of the growth of T. gondii using monensin, impairment of the BBB integrity can be restored (3) inhibition of WNT pathway by monensin can be detected by microarray experiment.
