Project description:Regulatory T cells (Tregs) are proposed to restrain anti-tumor T cell responses either directly or by suppression of antigen-presenting cells and can act in a tissue-specific manner. Treg abundance is typically reported as a measure of suppression, without consideration of Treg functional quality. Here, we find that Tregs suppress type 1 conventional dendritic cells (DC1) and thereby induce dysfunctional CD8+ T cell responses against lung cancer. This immunoregulatory effect is spatially coordinated within tissue-specific lymph node microniches and requires antigen-specific contact between DC1 and Tregs. Suppressive clonally expanded TH1-like effector Tregs were differentially induced in the lung lymph node in response to tissue-specific levels of interferon-gamma. In cancer patients, TH1-like Tregs but not CD8+/Treg ratios correlate with poor responses to checkpoint blockade immunotherapy. Thus, Tregs that adopt the interferon-gamma-dependent TH1-like effector state have increased potential to restrain tumor-reactive T cell responses and represent a critical barrier to productive anti-tumor immunity.

Project description:Local environmental factors influence CD8+ T cell priming in lymph nodes (LNs). Here, we sought to understand how factors unique to the tumor-draining mediastinal LN (mLN) impact CD8+ T cell responses toward lung cancer. Type 1 conventional dendritic cells (DC1s) showed a mLN-specific failure to induce robust cytotoxic T cells responses. Using regulatory T (Treg) cell depletion strategies, we found that Treg cells suppressed DC1s in a spatially coordinated manner within tissue-specific microniches within the mLN. Treg cell suppression required MHC II-dependent contact between DC1s and Treg cells. Elevated levels of IFN-γ drove differentiation Treg cells into Th1-like effector Treg cells in the mLN. In patients with cancer, Treg cell Th1 polarization, but not CD8+/Treg cell ratios, correlated with poor responses to checkpoint blockade immunotherapy. Thus, IFN-γ in the mLN skews Treg cells to be Th1-like effector Treg cells, driving their close interaction with DC1s and subsequent suppression of cytotoxic T cell responses.

Even though DC1 development was restored in these mice, they were still unable to reject a transplanted immunogenic fibrosarcoma, indicating that there are some functions of Batf3-dependent dendritic cells that are not restored by the Irf8VENUS transgene.

Abstract:
Cytotoxic T lymphocytes (CTL) responses are required to fight many diseases such
as viral infections and tumors. At the same time, they can cause disease when
induced inappropriately. Which factors regulate CTL and decide whether they
should remain silent or react is open to debate. The phenomenon called
cross-priming has received attention in this respect. That is, CTL expansion
occurs if antigen is recognized on the surface of professional antigen
presenting cells (APCs). This is in contrast to direct presentation where
antigen is seen on the surface of the target cells (e.g. infected cells or tumor
cells). Here we introduce a mathematical model, which takes the phenomenon of
cross-priming into account. We propose a new mechanism of regulation which is
implicit in the dynamics of the CTL: According to the model, the ability of a
CTL response to become established depends on the ratio of cross-presentation to
direct presentation of the antigen. If this ratio is relatively high, CTL
responses are likely to become established. If this ratio is relatively low,
tolerance is the likely outcome. The behavior of the model includes a parameter
region where the outcome depends on the initial conditions. We discuss our
results with respect to the idea of self/non-self discrimination and the danger
signal hypothesis. We apply the model to study the role of CTL in cancer
initiation, cancer evolution/progression, and therapeutic vaccination against
cancers.

Etv6 is an ETS family transcription factor that controls hematopoietic stem and progenitor cell (HSPC) function. We report that like HSPC, cDCs express Etv6 but not its antagonist ETS1, whereas interferon-producing plasmacytoid dendritic cells (pDCs) express both factors. Deletion of Etv6 in the bone marrow reduced the generation of DC1-like cells in vitro and the differentiation of DC1 in vivo, including the loss of signature marker CD8a. Global expression and chromatin profiling of Etv6-deficient DCs revealed impaired lineage identity of DC1, including the reduction of cDC signature and upregulation of pDC signature. Accordingly, DC-specific Etv6 deletion impaired CD8+ T cell cross-priming and tumor-specific CD8+ T cell responses. These results identify Etv6 as a regulator of DC1 differentiation and functional fitness that indirectly facilitates T cell cross-priming and antitumor immunity.

Project description:While conventional dendritic cells (DC)1 and DC2 are found in tumors, DC1 were shown to control tumor response to checkpoint blockade in preclinical models and are associated with better overall survival in cancer patients, reflecting their specialized ability to prime CD8+ T cell responses. Paradoxically, DC1 can also be found in tumors that resist checkpoint blockade, suggesting that, like many tumor infiltrating T cells, DC1 functionality may be altered in tumors. To address this question, we performed scRNAseq analyses to characterize DC gene architecture in mouse non-small cell lung cancer (NSCLC) lesions. To understand the relationship of the resulting transcriptional signatures with antigen uptake and migration, we profiled lung DCs in CCR7-/- mice, or DCs that had taken up (i) GFP from GFP-expressing tumors that had been seeded in the lung, and (ii) fluorescent microbeads that had been applied intranasally.

Project description:In addition to helper and regulatory potential, CD4+T cells also acquire cytotoxic activity marked by granzyme B (GzmB) expression and the ability to promote rejection of established tumors. Here we examined the molecular and cellular mechanisms underpinning the differentiation of cytotoxic CD4+T cells following immunotherapy. CD4 transfer into lymphodepleted animals or regulatory T cell (Treg)depletion promoted GzmB expression by tumor-infiltrating CD4+which was prevented by IL-2 neutralization. Transcriptional analysis revealed a polyfunctional helper and cytotoxic phenotype characterized bythe expression of the transcription factors T-bet and Blimp-1. Whilst T-bet ablation restrictedIFN-gproduction, lossof Blimp-1preventedGzmB expressionin response to IL-2, suggesting these are two independent programs required forpolyfunctionality of tumor-reactive CD4+T cells. The data underscores the role of Treg, IL-2 and Blimp-1 controlling the differentiation of cytotoxic T cells and offers a pathway to enhancement of anti-tumor activity through their manipulation.
