Project description:Serpine1 mRNA is abundantly induced during EMT. Paradoxically, this mRNA is enriched in the RISC complex, blocking its translation and acting as an endogenous RNA competitor for several miRNAs. Cells that overexpress Serpine1 mRNA have a greater capacity for migration, invasiveness, and resistance to anoikis. These cells show an altered proteomic profile, highlighting the increased levels of expression of the splicing factor TRA2b. This splicing factor is posttranscriptionally regulated during EMT by upregulation of Serpine1 mRNA and the resulting sequestration of miR-130b-5p by this mRNA, which contains three binding sites for miR-130b-5p. Through transcriptomic analysis, we confirmed that Serpin 1 mRNA expression modulates the expression of numerous genes, most of which (>60%) are also modulated by TRA2b overexpression.

Project description:EPB41L5 belongs to the band 4.1 superfamily. We investigate here the involvement of EPB41L5 in epithelial-mesenchymal transition (EMT) during mouse gastrulation. EPB41L5 expression is induced during TGFbeta-stimulated EMT, whereas silencing of EPB41L5 by siRNA inhibits this transition. In EPB41L5 mutants, cell-cell adhesion is enhanced, and EMT is greatly impaired during gastrulation. Moreover, cell attachment, spreading, and mobility are greatly reduced by EPB41L5 deficiency. Gene transcription regulation during EMT occurs normally at the mRNA level; EPB41L5 siRNA does not affect either the decrease in E-cadherin or the increase in integrin expression. However, at the protein level, the decrease in E-cadherin and increase in integrin are inhibited in both EPB41L5 siRNA-treated NMuMG cells and mutant mesoderm. We find that EPB41L5 binds p120ctn through its N-terminal FERM domain, inhibiting p120ctn-E-cadherin binding. EPB41L5 overexpression causes E-cadherin relocalization into Rab5-positive vesicles in epithelial cells. At the same time, EPB41L5 binds to paxillin through its C terminus, enhancing integrin/paxillin association, thereby stimulating focal adhesion formation.

SM004 - SARS infection of C57BL6, TIMP1 and Serpine1 knock-out mice.

Project description:This purpose of this experiment was to investigate the transcriptional differences between C57BL6, TIMP1 and Serpine1 knock-out mice infected with SARS MA15 virus. Overview of Experiment: Groups of 20 week old C57BL6, TIMP1 and Serpine1 (PAI1) knock-out mice were infected with SARS MA15 virus. Infections were done at 10^4 PFU or time-matched mock infected. Time points were 4 and 7 d.p.i. There were 2-4 animals/dose/time point. Lung samples were collected for virus load, transcriptional and proteomics analysis. Weight loss and animal survival were also monitored.

Identification of SERPINE1 as a Regulator of Glioblastoma Cell Dispersal via Analyzing Dynamic Transcriptome of Dispersing Cells

Project description:With a model mimicking GBM tumor cell dispersal, transcriptome changes between core (immotile) and dispersive (motile) cells were analyzed. Many genes are differentially expressed between these populations. This study focused on the genes that are significantly upregulated in dispersive cells. Besides gene sets related with the cell cycle and cell survival, epithelial to mesenchymal transition gene set is upregulated in dispersive cells. In this gene set, this study identified SERPINE1 gene as an important regulator of GBM cell dispersal.
