Project description:Purpose: NGS was used to determine if a distinct transcriptomic profile is observed among the experimental mice fed four different dietary components. Methods: We carried out RNA-Seq analysis of ileum tissue from 6 weeks male mice ad libitum fed for 10 weeks a high liquid sugar (23% (w/v)) or/and high fat (60% Kcal from fat) diet. The combined effect of sugar drink and high fat diet (HF-Sugar) was compared with sugar drink only (NF-Sugar), or high fat diet only (HF), or control diet that was plain water and normal fat diet (NF). Results: RNA-Seq revealed sample-specific clusters that included genes responding to each experimental diet. We found only addition of sugar drink to high fat group (HF-Sugar) not NF-Sugar and HF, there was a significant enrichment in biological functions relating to Inflammatory/Immune Responses, especially including dendritic cell (DC) and T cell related signaling pathway. Conclusions: Taken together, our data demonstrate that sugar drink synergistically promotes and exacerbates inflammatory responses driven by the high fat diet.

Project description:Adipose tissue plays an important role in storing excess nutrients and preventing ectopic lipid accumulation in other organs. Obesity leads to excess lipid storage in adipocytes, resulting in the generation of stress signals and the derangement of metabolic functions. SIRT1 is an important regulatory sensor of nutrient availability in many metabolic tissues. Here we report that SIRT1 functions in adipose tissue to protect from the development of inflammation and obesity under normal feeding conditions, and the progression to metabolic dysfunction under dietary stress. Genetic ablation of SIRT1 from adipose tissue leads to gene expression changes that highly overlap with changes induced by high fat diet in wild type mice, suggesting that dietary stress signals inhibit the activity of SIRT1. Indeed, we show that high fat diet induces the cleavage of SIRT1 in adipose tissue by the inflammation-activated caspase-1, providing a link between dietary stress and predisposition to metabolic dysfunction.

Project description:The main goal of swine production is to convert feedstuffs into edible meat whose major component is skeletal muscle. The overall objective of this project is to study the effect of dietary lysine on the gene expression profile of skeletal muscle in late stage finishing pigs. The hypothesis for this is that adequate or excess level of dietary lysine will change the expression levels of numerous genes, and these changes are in favor of muscle protein accretion and are associated with various blood metabolites and growth performance parameters. Three experimental (lysine-deficient, lysine-adequate, lysine-excess) diets were respectively fed to 3 groups of pigs (initial body weight, ~95 kg/pig; 3 pigs/group) for a total of 5 weeks.

Project description:It is known that exogenous factors influences sperm quality and function, such as toxins, exercise, diet and smoking. Still, the moleuclar mechanisms of this is not understood. Here, we set out to determine how a dietary insult of different sugar levels in the diet affects Drosophila sperm proteome, by dissecting out seminal vesicles containing mature sperm from adult flies. These flies have been on a dietary intervention of two days on a sugar diet of 3, 30 or 300 g/L of sugar. Whole seminal vesicles were then used for mass spectometry to increase our understanding of how an acute dietary intervention may change sperm proteomics.

Project description:Nutrient-dependent gene regulation critically contributes to homeostatic control of animal physiology in changing nutrient landscape. In Drosophila, dietary sugars activate transcription factors (TFs), such as Mondo-Mlx, Sugarbabe and Cabut, which control metabolic gene expression to mediate physiological adaptation to high sugar diet. TFs that correspondingly control sugar responsive metabolic genes under conditions of low dietary sugar remain, however, poorly understood. We have used de novo motif prediction to uncover a significant over-representation of GATA-like motifs on the promoters of sugar-responsive genes in Drosophila larvae. GATA TF Grain was found to contribute to the regulation of sugar-responsive genes, and consequently to central carbon and lipid metabolism, primarily on low sugar diet. Grain targets include known sugar responsive TFs, cabut and smad on X (smox). Moreover, Grain promotes the expression of genes involved in de novo lipogenesis. Grain chromatin binding sites significantly converge with those of Sugarbabe. Grain and Sugarbabe both activate lipogenic genes, but display functional predominance on low and high sugar conditions, respectively. Collectively, our data provides evidence for a metazoan GATA transcription factor in nutrient-responsive metabolic regulation in vivo.

Project description:Iron is an essential nutrient yet toxic in excess. Our understanding of mammalian mechanisms of dietary iron absorption exceeds our understanding of mechanisms of iron excretion. Here we demonstrate that biliary excretion of excess iron in mice requires the metal transporter SLC39A14 and that excess Fe is excreted into bile as ferritin and heme. The identification of a molecular determinant of iron excretion and the biochemical form of bile iron could lead to development of novel therapeutics for human diseases of iron excess
