Project description:Neurogenic hypertension stems from an imbalance in autonomic function that shifts the central cardiovascular control circuits towards a state of dysfunction. Using the female spontaneously hypertensive rat (SHR) and the normotensive Wistar Kyoto (WKY) rat model, we compared the transcriptomic changes in three autonomic nuclei in the brainstem, the nucleus of the solitary tract (NTS), caudal ventrolateral medulla (CVLM), and rostral ventrolateral medulla (RVLM) in a time series at 8, 10, 12, 16, and 24 weeks of age, spanning the pre-hypertensive stage through extended chronic hypertension.

Project description:Genome-wide gene expression was obtained in three auditory brainstem nuclei (defined below), at two different ages in mice, postnatal day (P)3 and P14. The primary aim was to identify genes which are differentially expressed between the medial nucleus of the trapezoid body (MNTB) and the superior olive (LSO), at both age groups. Tissue samples from the ventral cochlear nucleus (VCN), the medial nucleus of the trapezoid body (MNTB) and the lateral superior olive (LSO) were collected from 200 μm thick fresh transverse mouse brainstem slices prepared with a vibratome tissue slicer. Individual brainstem nuclei were dissected out with fine forceps under optical control with a binocular. For each nucleus (VCN, MNTB, and LSO), the tissue from six male C57Bl6 mice at postnatal day (P) 3, and from three male C57Bl6 mice at P14 was pooled. The RNA was isolated with the RNeasy Micro kit (Qiagen); RNA samples were amplified using a WT ovation Pico RNA amplification (Nugen, CA, USA). The cDNA was hybridized onto mouse Affymetrix microarrays 430 2.0 (Affymetrix, Santa Clara, USA).

Project description:We here test the concept that disease states may result not from a single cause but from small changes in a network that are collectively significant. We recently showed that development of hypertension (HTN) in the spontaneously hypertensive rat (SHR) model of human essential hypertension is accompanied by changes in microRNA expression levels in the brainstem tracking the development of HTN 1,2. This led to the hypothesis that preventing the change in microRNA levels could prevent the development of HTN. We propose that hypertension emerges from a network that has been pushed out of a normotensive equilibrium into a compensatory, pathological state. We show that small perturbations in the gene regulatory networks in the brainstem by selectively blocking two microRNAs highlighted in our previous results, miR-135a and miR-376a, is sufficient to prevent development of hypertension in the SHR model with a 38% reduction in blood pressure after one week persisting to 29% reduction after two weeks with no change in blood pressure in WKY controls at either timepoint. This effect appears driven by only modest, yet sometimes significant, changes in the expression of rate-limiting genes including IL1a, IL1b, Agtr1a, and Dbh among others. Many of these genes are direct targets of these miRNAs, suggesting that the combination of genes that are targeted in the network is responsible for the effect. The demonstration that hypertension is an emergent property of an underlying regulatory network suggests that a new treatment paradigm altogether is needed.

Project description:Brainstem nuclei dysfunction is implicated in sudden unexpected death in epilepsy (SUDEP). In animal models, deficient serotonergic activity is associated with seizure-induced respiratory arrest. In humans, glia are decreased in the ventrolateral medullary pre-Botzinger complex that modulates respiratory rhythm, as well as in the medial raphe that modulates respiration and arousal. Finally, SUDEP cases have decreased midbrain volume.
To understand the potential role of brainstem nuclei in SUDEP, we evaluated molecular signaling pathways using localized proteomics in microdissected midbrain dorsal raphe and medullary raphe serotonergic nuclei, as well as the ventrolateral medulla in brain tissue from epilepsy patients who died of SUDEP and other causes in diverse epilepsy syndromes, and non-epilepsy control cases

Project description:Leigh Syndrome is characterized by symmetrical brain lesions and neuroinflammation in select nuclei, predominantly brainstem and/or basal ganglia. Accordingly, Ndufs4-deficient mice present overt lesions in brainstem, olfactory bulb, and basal ganglia. To define the contribution of Ndufs4 deficiency in excitatory neurons to the overall neuroinflammatory phenotype, we performed Gene expression analysis in the brainstem of mice with specific deletion of Ndufs4 in Vglut2-expressing neurons (Vglut2:Ndufs4cKO mice). This analysis allowed us to further define the inflammatory phenotype present in the brainstem of Vglut2:Ndufs4cKO mice.

Project description:Hypertension is a multifactor disease that possibly involves alterations in gene expression in hypertensive relative to normotensive subjects that are largely unknown. In this study we used high-density oligoarrays to compare gene expression profiles in cultured neurons and glia from pons and medulla oblongata of newborn spontaneously hypertensive (SHR) and normotensive Wistar Kyoto (WKY) rats, a widely documented animal model of hypertension. We found 358 genes differentially expressed between SHR and WKY brainstem cells that preferentially map to 24 metabolic/signaling pathways. Some of the pathways and regulated genes identified herein are obviously related to blood pressure regulation; however there are several genes differentially expressed in SHR not yet associated to hypertension or participating in blood pressure regulation. These constitute a rich resource for the identification and characterization of novel genes involved in hypertension development, or associated to phenotypical differences observed in SHR relative to WKI. In conclusion, this study describes for the first time the gene profiling pattern of brainstem cells from SHR and WKY rats, which opens up new possibilities and strategies of investigation and possible therapeutics to hypertension, as well as for the understanding of the brain contribution in this pathology.
