Project description:Oilseed rape is both an important oleaginous crop and agriculture sightseeing crop whereas has relatively scanty flower color. As natural flavonoids, Anthocyanin are responsible for the attractive red, purple, and blue colors of various tissues in higher plants, especially for the ornamental plants flower. One Brassica napus-Orychophragmus violaceus disomic addition line (M4) obtained previously exhibits red petals whichresult from anthocyanin biosynthesis. Transcriptome analysis of M4, B. napus (H3), natural individuals of O. violaceus with purple petals (OvP) and white petals (OvW) revealed that most of structural genes for the anthocyanin synthesis were up-regulated in both M4 and OvP, especially key gene ANS in the last step. Reads assembling and sequence alignment showed that the regulatory DEG PAP2 in M4 was from the transcript of O. violaceus. OvPAP2 was transformed into Arabidopsis thaliana and B. napus driven by the CaMV35S promoter and the rape petal-specific prompter XY355. Transgenic A. thaliana plants showed different levels of purple pigments in most of the organs, including the petals, and transgenic B. napus flowers exhibited restricted accumulation of anthocyanins in stamens when driven by CaMV35S promoter, but generated both red petals and anthers driven by the XY355 promoter. These results provided a platform for expounding the anthocyanin biosynthesis pathway in B. napus petals and give a successful case for flower color modification of the agriculture sightseeing rape.

Project description:Animal pigment patterns play important roles in behavior and, in many species, red coloration serves as an honest signal of individual quality in mate choice. Among Danio fishes, some species develop erythrophores, pigment cells that contain red ketocarotenoids, whereas other species, like zebrafish (D. rerio) only have yellow xanthophores. Here, we use pearl danio (D. albolineatus) to assess the developmental origin of erythrophores and their mechanisms of differentiation. We show that erythrophores in the fin of D. albolineatus share a common progenitor with xanthophores and maintain plasticity in cell fate even after differentiation. We further identify the predominant ketocarotenoids that confer red coloration to erythrophores and use reverse genetics to pinpoint genes required for the differentiation and maintenance of these cells. Our analyses are a first step towards defining the mechanisms underlying the development of erythrophore-mediated red coloration in Danio and reveal striking parallels with the mechanism of red coloration in birds.

Project description:In order to reveal the candidate genes related to yellow plumage in Chinese chicken, a hybrid population of Huiyang Bearded chicken and White Leghorn chicken, and transcriptome data were generated from the yellow and white feather follicle of F3 population in two periods (7 and 11weeks) respectively, using RNA-seq. 127 common different expressed genes were obtained(DEGs) between two periods, these DEGs were mainly enriched in the Gene Ontology classes ‘developmental pigmentation’, ‘melanin biosynthetic process’, ‘melanosome organization’, ‘melanosome membrane’ and ‘melanosome’all related to the pigmentation process. And involved genes were considered as pigment genes that play important roles in melanogenesis. The results reveal key functional genes and possible molecular mechanisms for the elucidation of yellow plumage formation in Chinese indigenous chickens.

Project description:Gliomas represent 80% of all malignant cerebral tumors and are classified within different malignity grade. Glioblastoma, the most aggressive group, represent more than half of all gliomas but remain a heterogeneous group. Indeed, patient survival can reach from several months to a few years after surgery and chemotherapy. To study gliomas, MALDI mass spectrometry imaging is an interesting technique, allowing the analysis of tumor heterogeneity. In this study, MALDI-MSI is coupled with spatially-resolved microproteomic within the objective to identify subgroups of glioblastomas patients in order to help diagnosis and prognostic. Molecular images are generated from thin tissue section in order to determine the spatial localization of digested peptides. Thanks to unsupervised statistical analysis, we then generated hierarchical clustering of homogeneous molecular regions. According to these regions, spatially resolved proteomic gave access to large scale protein identification and relative quantification. Hierarchical clustering reveal three molecular region within all the tumors sample : region red which represent 33% of surface of all sample, region yellow (25.7%) and region blue (40.8%). Consequently, 3 groups of patient can be determined: group A composed mostly of region red (13/50 patients), group B composed mostly of region yellow (9/50 patients) and group C composed mostly of region blue (24/50 patients). Spatially resolved proteomic was performed and the 147 extractions were analyzed by nano-LC MS/MS. Two-way ANOVA test reveal panel of proteins specifically overexpressed in each region. Protein overexpressed in region red are associated with neurogenesis; protein overexpressed in region yellow are associated with an activation of the immune system and protein overexpressed in region blue are involved with viral processes. To conclude, the combination of MALDI-MSI and microproteomic provides new information on these tumors, allowing us to differentiate three group of patient. In the future, these data will permit to build a more precise classification, a better medical care for patients and the identification of potential new therapeutic targets.
