Project description:The innate immune system can recognize tumor cells and coordinate with the adaptive immune system to initiate an anti-tumor response within the tumor microenvironment. To investigate the role of NK cells and their interaction with other immune cells in the anti-tumor immunity in Reg-1ΔNK. We subcutaneously injected B16F10 cells into Reg-1fl/fl and Reg-1ΔNK, followed by scRNA-seq analysis of the whole tumor cell suspension and sorted TIL-NK cells.

Project description:Purpose: For comparing the transcript changes, we conducted mRNA-sequencing of splenic NK cells from Ncr1Cre-Mettl3fl/fl (cKO) and Mettl3fl/fl (WT) mice. Method: Firstly, The splenic NK cells (CD45.2+CD3-NK1.1+NKp46+) are purified via Fluorescence activated Cell Sorting (FACS), then frozen in -80 °C ultra-low temperature refrigerator, followed by High-throughput sequencing, in three replicates, using Illumina Hiseq 1500 platform. Result: Using standard data process workflow, we found the differentailly expressed genes in NK cells from Ncr1Cre-Mettl3fl/fl (cKO) mice, compared with those from Mettl3fl/fl (WT) mice.

CITE-seq of in vitro control and memory-like human NK cells

Project description:Peripheral blood NK cells were cultured into either control or cytokine-induced memory-like NK cells as described in PMID: 32614951. After differentiation, cells were labeled with hashtag antibodies and cite-seq antibodies and control and memory-like NK cells were combined and loaded onto the 10x Genomics Chromium Instrument a the McDonnell Genome Institute at Washington University School of Medicine. The following samples were analyzed to develop a classification tool for unbiased identification of memory-like NK cells in a bulk single-cell RNA-seq population.
