Brain Region-Specific Gene expression in Exercise for a Mouse Model of Parkinson's Disease

Project description:Exercise plays an essential role in improving motor symptoms in Parkinson’s disease (PD), but the mechanism in the central nervous system remains obscure. We tested effects of 12-week treadmill exercise on a chronic 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced mouse model of PD and observed a significant improvement in motor ability. Here we performed RNA-sequencing on 4 brain regions from control animals, MPTP-induced PD mice and MPTP-induced PD mice treated with exercise for 12 weeks.

Seven brain regions examined: amygdala, basal ganglia, cerebellum, frontal cortex, hippocampus, cingulate cortex, olfactory bulb. Experiment Overall Design: Tissue from three animals was pooled on each array. Three biological replicates per strainxregion condition. Animals were 4-5 weeks of age.

Project description:Doxorubicin induces cardiomypathy in oncology patients. This study is aimed at identifying molecular mechanisms of doxorubicin cardiac toxicity in rats. Transcription profiling of heart tissue of rats treated with saline or doxorubicin 1, 2 or 3 mg/kg/week by iv once per week for 2, 4 or 6 weeks. Some animals were allowed to recovery from drug administration for 2 or 4 weeks. Note the recovery column refers to whether an animal was let without drug for some weeks before sacrifice.

Gene expression analysis of eight-cell stage rat embryos sired by paternally exposed males to saline or BEP

Project description:8-cell stage rat embryos were sired by males treated with saline or a chemotherapeutic cocktail, BEP, consisting of Bleomycin, Etoposide, and Cisplatin for 9 weeks. Animals were given an additional 9 weeks of recovery, without any treatment, prior to mating. qPCR gene expression profiling for 84 stem cell transcription factors. Total RNA extracted from 25-35 embryos at the eight-cell stage sired by control or BEP-treated males after a 9-wk recovery period (2 females mated per male, n= 4 males/treatment).

Influence of the Soluble Guanylate Cyclase Stimulator Riociguat on Myocardial Left Ventricular Proteome and microRNA expression after Experimental Aortic Stenosis in Mice

Project description:We analyzed global proteomic adaptations during heart failure (HF) progression in a mouse model, suffering from left ventricular pressure overload due to transverse aortic constriction (TAC) and in response to riociguat (RIO) treatment, to gain deeper insights in beneficial effects due to sGC stimulation on cardiac remodeling and HF. TAC and sham animals were treated with either riociguat (RIO; 3 mg/kg/day) or vehicle (VEH; Transcutol®/Cremophor®/water: 10%/20%/70%) for five weeks, starting three weeks post-op when cardiac hypertrophy was established, resulting in four treatment groups ( n=5-6 per group). At the end of the study, hearts were dissected and proteomes of the left ventricles (LV) were analyzed by mass spectrometry (MS).
