Project description:Cancer stem-like cells are defined as small population of cancer cells which has, higher tumor-initiating ability, self-renewing ability and differentiation ability. In this experiment, transcription profile of cancer stem-like cells derived from human colon cancer line cell SW480 was investigated. Cancer stem-like cells were isolated as side population (SP) cells by Hoechst33342 dye staining from human colon cancer line cell SW480. Non-cancer stem-like cells were isolated as main population (MP) cells. RNAs were isolated from SP cells and MP cells derived from SW480 cells and array experiment was perfromed in dye swaped fashion.

Project description:Gallic acid (also known as 3,4,5-trihydroxybenzoic acid, GA), a naturally occurring phenolic acid, is a major constituent of tea and red wine. It has been demonstrated that GA possess anti-cancer activities. We found the epigenetic effect of GA in tobacco-associated human cancer cell lines. In this dataset, we include the expression array data from human lung cancer H1299 cell line with or without the treatment of 5azaC or GA. These data were used to obtain genes upregulated in both 5azaC- orGA-treated H1299 cells.

Project description:FoxA transcription factors are involved in development and tumorigenesis of the gastrointestinal tract. However, the downstream programs controlled by FoxA factors remain poorly understood. The goal of this study is to understand the transcriptional responses regulated by FoxA proteins in liver and colon cancer cells. Human liver cancer cell line HepG2 and colon cancer cell line LS174T infected with lentivirus expressing shRNAs targeting human FoxA1 and FoxA2.

Project description:Caffeic acid acetophenate exhibits anti-tumor activity. We employed proteomics to investigate the targets of caffeic acid acetophenate in human colon cancer cells. The experimental group consisted of SW480 human colon cancer cells treated with 10μM caffeic acid phenethyl for 24 hours, while the control group was treated with an equivalent volume of DMSO. Non-standard quantitative mass spectrometry was utilized to analyze differentially expressed proteins between the two groups and identify the specific target of caffeic acid acetophenate on colon cancer cells.

Proteomic characterization of transcription and splicing factors associated with a more invasive phenotype in colorectal cancer

Project description:To find new transcription and splicing factors associated with the metastatic phenotype we used a concatenated tandem array of consensus transcription factors response elements (catTFRE) on nuclear extracts of colon cancer cell lines. We used two different isogenic pairs of colorectal metastatic cells, KM12SM/KM12C and SW620/480. Proteins were pulled-down using biotinylated DNA sequences and streptavidin beads and analyzed by LC-MSMS.

Project description:Analysis of SW480 colon cancer cells overexpressing AMIGO2. Result provide insight into the role of AMIGO2 in colon cancer. Amphoterin-inducible genes and open reading frame 2 (AMIGO2) have been reported to play a role in promoting malignant transformation in several cancer types, including human colon cancer. However, the exact role of AMIGO2 in human colorectal cancer invasiveness remains unclear. We established an AMIGO2 overexpression line (SW480_A9) and a vector control line (SW480_E11) using a human colon cancer cell line (SW480) and analyzed differential expression of AMIGO2 on a microarray (Toray 3D-Gene).
