[E-MEXP-3786] IGF-I-induced chronic gliosis and retinal stress lead to neurodegeneration in an animal model of retinopathy

Project description:Transcription profiling by array of mouse male retinas to investigate IGF-I-induced chronic gliosis and retinal stress IGF-I exert multiple effects in different retinal cell populations in both physiological and pathological conditions. Transgenic mice overexpressing IGF-I in the retina showed impaired electroretinographic responses at 6-7 months of age that worsen with age. This retinal neuronal dysfunction was correlated with the loss of rod photoreceptors, bipolar, ganglion and amacrines cells. Neuronal alterations were preceded by the overexpression of retinal stress markers, acute phase proteins and gliosis-related genes. IGF-I overexpression leads to chronic gliosis and microgliosis in TgIGF-I retinas, with mild oxidative stress, impaired recycling of glutamate and defective potassium buffering. These impaired supportive functions can contribute to neurodegeneration in TgIGF-I retinas, together with the increased production of pro-inflammatory cytokines, potential mediators of neuronal death.

Mice with iron deficiency anemia (IDA)-low hepcidin show a pro-inflammatory response that is blunted in iron deficienct-high hepcidin Tmprss6 null mice. We investigated the transcriptional response associated with chronic hepcidin overexpression by comparing whole genome transcription profiling of the liver of Tmprss6 KO mice and IDA animals, irrespective of iron deficiency.

Comparison of proteomic responses to individual and combinatorial copper and iron deficiencies in Arabidopsis thaliana rosettes.

Project description:Purpose: plants exposed to multiple simultaneous adverse growth conditions trigger molecular responses that differ from the sum of those to individual stressors. Copper and iron are fundamental elements required for proper photosynthesis, energy production, DNA metabolism and hormone sensing, among all. Therefore, copper and iron deprivation limits plant yield. In natural environments, simultaneous deficiency to copper and iron can occur. As part of a multiple high-throughput study to identify combinatorial responses to both copper and iron deficiency, proteomic profiling of Arabidopsis thaliana rosette leaves exposed to copper and/or iron deficiencies have been conducted.

Defining effects of in utero arsenic exposure on immune cells in lungs of adult mice exposed to Influenza A with single cell RNA sequencing

Project description:Our recent studies have shown that adult mice exposed to inorganic arsenic exclusively in utero (100 ppb via maternal drinking water) exhibit greater lung damage and inflammation following infection with IAV (Influenza A virus, H1N1) than control mice not exposed in utero. We employed an unbiased single cell RNA sequencing approach to conduct a high resolution molecular analysis of recruited immune cells and their transcriptional activities in the lungs of arsenic-exposed and control mice at day 3 after IAV infection. Transcriptional profiling of single immune cells from 6 mice (3 As, 3 Con, all exposed to IAV) revealed evidence of an enhanced inflammatory response mediated by dysregulation of innate immune functions of monocyte derived macrophages, neutrophils and alveolar macrophages.
