KaratzaKaralis2020 - CYP mediated losartan metabolism

ABSTRACT: A four-compartment pharmacokinetic model that describes the disposition of losartan and E-3174 (Karatza and Karalis, 2020). The main objective of this model is to describe the secondary maxima observed in the plasma concentration-time profiles of losartan and E-3174. This model incorporates a sinusoidal equation which illustrates open-close cycles of gastric pyloric valve.

Project description:We report label-free quantification of xenobiotic metabolizing enzymes (XME), transporters, redox enzymes, proteases, nucleases and tight junction proteins in 22 human brain microvessel samples. More than 3500 proteins were identified and quantified. These data can be used in physiologically based pharmacokinetic models to predict drug disposition in the brain and permitting dose adjustment (precision dosing) in special populations of patients, such as those with dementia.

Project description:This model was developed with the aim of constructing an equilibrium model of the pharmacokinetic behaviour of a drug exhibiting target-mediated drug disposition (TMDD). TMDD involves the inclusion of drug-target interactions within a pharmacokinetic description, something which is usually considered negligible and subsequently excluded. Two approaches were used, one of which involved a quasi-equilibrium method to describe the kinetics of TMDD.

Project description:The primary objective of this study was to determine the effectiveness of bortezomib alone or in combination with irinotecan in patients with advanced gastric and gastroesphageal cancer. A secondary objecitve was to determine whether treatment was associated with changes in gene expression in the tumor and normal adjascent tissue. Tumor biopsies and biopsies of normal adjascent tissue were obtained before therapy and 24 hours after therapy. Differences in gene expression were evaluated between tumor and normal tissue (N=8 patients), and between post-treatment and pretreatment specimens for bortezomib alone (N=2 patients) and bortezomib plus irinotecan (N=10 patients).

Quantitative LC-MS/MS Proteomics of human small intestine

Project description:The objective was to determine the expression of a wide range proteins pertinent to metabolism and disposition of chemicals and nutrients in the intestinal epithelium. Ileum and jejunum biopsy specimens were obtained from 16 patients undergoing gastrointestinal elective surgery. Mucosal fractions were prepared and analysed using targeted and global proteomic approaches. A total of 29 enzymes, 32 transporters, 6 tight junction proteins, 2 adhesion proteins, alkaline phosphatase, thioredoxin, 5 markers and 1 regulatory protein were quantified; 60 for the first time.

Resistance to ATR Inhibitors Mediated by Loss of the Nonsense-Mediated Decay Factor UPF2

Project description:Over one million cases of gastric cancer are diagnosed each year and metastatic disease continues to have a poor prognosis. A significant proportion of gastric tumors have defects in the DNA damage response pathway creating therapeutic opportunities through synthetic lethal approaches. In particular, several small molecule inhibitors of Ataxia-Telangiectasia Mutated and Rad3-related protein kinase (ATR), a key regulator of the DNA Damage response, are now in clinical development as targeted agents for gastric cancer. Here, we sought to discover genetic determinants of response and resistance to ATRi in gastric cancer cells. We performed a large-scale CRISPR interference screen to discover determinants of resistance to ATRi in gastric cancer. Top hits were validated and further evaluated to define mechanisms of resistance. We identified components of the nonsense-mediated decay pathway and the UPF2 gene, in particular, as mediators of sensitivity to ATRi. We show loss of UPF2 causes resistance to ATRi through the abrogation of ATRi mediated cell cycle arrest and find evidence that UPF2 regulates R-loops. Our results uncover a novel role for NMD factors in R-loop formation and resistance to ATRi in gastric cancer cells, potentially through R-loop formation.

Macrophages preserve endothelial cell specialization in the adrenal gland to modulate aldosterone secretion and blood pressure (scRNA-Seq)

Project description:The adrenal cortex is characterized by a distinct architecture as well as a high density of specialized sinusoidal blood vessels. The preservation of this particular endothelial cell phenotype is likely vital for proper adrenal gland function. The aldosterone-producing zona glomerulosa harbors macrophages in close association with sinusoidal capillaries. However, the function of this macrophage-endothelial cell-juxtaposition in steady-state conditions is unknown. We show that macrophages preserve capillary specialization in the adrenal gland and modulate aldosterone secretion. By combining macrophage-specific deletion of the angiogenic cytokine Vascular Endothelial Growth Factor A (VEGF-A), single-cell transcriptomics and functional phenotyping, we provide evidence that loss of VEGF-A in myeloid cells, including adrenal gland macrophages depletes a specialized subset of PLVAP+ fenestrated endothelial cells in the zona glomerulosa of mice, along with increased deposition of basement membrane collagen IV and in
