Ontogenesis of T cells in the thymus is a complex process whose molecular control is poorly understood. The present study investigated microRNAs involved in human thymocyte differentiation by comparing the microRNA expression profiles of thymocytes at the double-positive, single-positive CD4(+) and single-positive CD8(+) maturation stages. Molecular changes induced by melanoma cell conditioned medium (MCM) in HUVEC cells.

Project description:Malignant melanoma is a complex genetic disease and the most aggressive form of skin cancer. Melanoma progression and metastatic dissemination fundamentally relies on the process of angiogenesis. Melanomas produce an array of angiogenic modulators that mediate pathological angiogenesis. Such tumor-associated modulators arbitrate the enhanced proliferative, survival and migratory responses exhibited by endothelial cells, in the hypoxic tumor environment. The current study focuses on melanoma-induced survival of endothelial cells under hypoxic conditions. Melanoma conditioned media were capable of enabling prolonged endothelial cell survival under hypoxia, in contrast with the conditioned media derived from melanocytes, breast and pancreatic tumors. To identify the global changes in gene expression and further characterize the pro-survival pathway induced in endothelial cells, we performed microarray analysis on endothelial cells treated with melanoma conditioned medium under normoxic and hypoxic conditions. Huvec cells were grown in melanoma conditioned medium or DMEM 10% FCS for 12 h under hypoxic or normoxic conditions. In order to identify the transcripts modulated by melanoma CM, samples treated with MCM were compared to those grown in DMEM alone.

Expression data from mammary epithelial cells from BRCA1 mutation carriers and non BRCA1 mutation carriers

Project description:Female BRCA1 mutation carriers have a nearly 80% probability of developing breast cancer during their life-time. We hypothesized that the breast epithelium at risk in BRCA1 mutation carriers harbors mammary epithelial cells (MECs) with altered proliferation and differentiation properties. Microarray studies revealed that PMEC colonies from BRCA1 mutation carriers anticipate expression profiles found in BRCA1-related tumors, and that the EGFR pathway is upregulated in BRCA1 mutation carriers compared ton non BRCA1 mutation carriers. Keywords: Class comparison and pathway analysis 10 colonies were collected and RNA was isolated using the Absolutely RNA Nanoprep kit, Stratagene. The arrays included duplicates from four normal controls and from two BRCA1 mutation carriers and single arrays from another two BRCA1 mutation carriers.

Transcription profiling of Brucella abortus using an ORFeome-based Brucella whole-genome DNA microarray

Project description:A direct analysis of the global transcriptional profile of B. abortus cells grown in laboratory conditions was made. Thus, three independent biological replicas were made, and following the RNA isolation, samples were reverse transcribed in aRNA and labelled with Cy3 fluorescent dye. The use of a single dye labelling approach allowed us an estimation of the genes more abundantly expressed in Brucella under laboratory conditions. The evaluation experiment was composed of three slides yielding six measurements per gene (representing three biological replicates, since each gene is present twice on each slide) to confirm the reproducibility of the gene expression data. <br>The complete dataset was sorted by the normalized and corrected fluorescence intensity values.

Radiotherapy-induced miR expression influences breast cancer recurrence formation.

Project description:The act of surgery strongly stimulated breast cancer cell growth in mice. Local irradiation of wounded breast efficiently prevented wounded-induced breast cancer cell growth. These modifications were linked to miR modulation both in human specimens and in the mouse model, in vivo. The most significantly IORT-induced miR was miR-223. miR-223 expression controlled EGF production and, eventually, cancer cell proliferation and survival.
