Exploration of genes expression with a DNA microarray Cancerochip.

ABSTRACT: Our goal was to search for new molecular markers of the mitochondrial implication in the thyroid tumorigenesis. Using a DNA microarray (Cancerochip), we explored a collection of thyroid samples representing 4 classes of thyroid tumors. 45 thyroid tumoral and paired control tissues representing 4 classes (benign follicular tumors [FTA], oncocytic variants of follicular tumors [OT], papillary thyroid carcinomas [PTC] and tumors of uncertain malignancy potential [TUMP]) of thyroid tumors were explored for the expression of 6866 genes.

Transcriptomics analysis and hormonal changes of female and male rats treated chronically with a low dose of acrylamide in their water supply

Project description:Acrylamide (AA) is known to produce tumors in animals in different tissues including the thyroid where typically the tumors are found in follicular cells in the thyroid gland. Using transcriptomic tools, we examined global gene expression changes in the thyroid glands of RccHan Wistar rats that were sub-chronically exposed to a low dose of AA (3 mg/Kg). A transcriptomic approach was used to investigate changes in gene expression and their association with physiological responses (changes in plasma hormone levels) in rats treated with acrylamide (AA), exposed from gestational day 6 to post natal day 21. We hypothesized that 3 mg AA/Kg bw/day exposure would produce changes in plasma hormone levels associated with key genes and biochemical pathways involved with molecular actions of AA.

Expression analysis of mouse thyroid tumors

Project description:Anaplastic thyroid carcinoma (ATC) is the most aggressive form of thyroid cancer, and often derives from pre-existing well-differentiated tumors. We have engineered the first mouse model of ATC by combining in the mouse thyroid follicular cells two molecular hallmarks of human ATC: activation of PI3K (via Pten deletion) and inactivation of p53. By 9 months of age, over 75% of the compound mutant mice develop aggressive, undifferentiated thyroid tumors that evolve from pre-existing follicular hyperplasia and carcinoma. These tumors display all the features of their human counterpart, including pleomorphism, epithelial-mesenchymal transition, aneuploidy, local invasion and distant metastases. We have performed expression profiling of thyroids from control, single mutants, compound mutants, follicular tumors from Pten-/- mice, and anaplastic tumors from Pten, p53-/- mice.

Project description:Although most thyroid tumours are benign, thyroid cancer represents the most common malignancy of the endocrine system, comprising mainly follicular and papillary thyroid carcinomas (FTC and PTC, respectively). Previous studies have shed some light on the molecular pathogenesis of thyroid cancer but there have not been any comprehensive mass spectrometry-based proteomic studies to reveal protein expression differences between thyroid tumours and the molecular alterations associated with tumour malignancy. We applied a label-free quantitative mass spectrometry analysis to compare normal thyroid tissue with the three most common tumours of the thyroid gland: follicular adenoma, follicular carcinoma and papillary carcinoma.
