Transcription profiling of mouse visual cortex from P46 mouse non-deprived vs. deprived


ABSTRACT: Wild-type (C57Bl6) mice underwent monocular enucleation at P42 and were sacrificed 4 days later at P46. Bilateral (non-deprived and deprived) visual cortex was dissected from 1-mm thick coronal sections, and total RNA was extracted from the tissue lysate using Trizol (Gibco-BRL). Experiment Overall Design: Gene expression in pooled samples from 4 non-deprived (ipsilateral to monocular enucleation) and 4 deprived (contralateral to monocular enucleation) visual hemispheres was compared.

There are critical periods in development when sensory experience directs the maturation of synapses and circuits within neocortex. We report that the critical period in mouse visual cortex has a specific molecular logic of gene regulation. Four days of visual deprivation regulated one set of genes during the critical period, and different sets before or after. Dark rearing perturbed the regulation of these age-specific gene sets.
