Transcription profiling by array of Burkholderia cenocepacia J2315 grown under low or atmospheric oxygen concentration conditions

Project description:B. cenocepacia J2315 was grown on LB medium to mid-stationary phase at OD 0.3 under full aeration and then transferred into a 50 ml centrifuge tube and placed upright into a CampyGen Compact (Oxoid, Basingstoke, UK) plastic pouch containing the gas generating paper sachet. The pouch was sealed immediately and then the cap of the centrifuge tube loosened to allow exchange of atmosphere between centrifuge tube and pouch. The culture was further incubated at 37 degrees centigrade and 150 rpm for 2-3 hours until it reached OD 0.5. The centrifuge tube was sealed before opening the pouch and then snap-cooled before harvest to minimise exposure to atmospheric oxygen and to ensure expression profiles reflect the lower oxygen concentration. The expression profile was compared to cells grown to OD 0.5 at atmospheric oxygen concentrations.

Project description:The purpose of this experiment was to identify genes responding differently to a 24 h low red to far red ratio (R:FR) treatment in plants grown at 16 and 22 degrees Experiment Overall Design: Microarrays were used as a gene discovery tool to identify genes showing differential regulation by low R:FR at 2 growth temperatures. Plants subject to a 24 h low R:FR light treatment were compared with control plants maintained in high R:FR. This experiment was carried out at two growth temperatures, 16oC and 22oC.

Cytosolic glutathione reductase (OsGR2) regulates plant development and salt stress tolerance in rice

Project description:Rice genome contains three genes that encode for glutathione reductase (GR) viz., OsGR1, 2 and 3. GR is an important component of the anti-oxidative machinery of plant cells. GR2 down-regulated plants were produced by RNAi mediated down-regulation of GR2 (GR2-Ri). GR2-Ri plants were significantly smaller and have significantly lower grain yield (grain number per panicle) compared to WT(Wild type), under control conditions. RNA-Seq analysis of panicles (differentiation stage) identified genes known to affect grain size to be differentially regulated in GR2-Ri compared to WT, respectively, under control conditions.
