ClariomS Array to determine the effect of a vaspin-derived peptide on gene expression in fully differentiated primary subcutaneous mouse adipocytes

Project description:Screening for potential target proteases of vaspin revealed rapid and specific cleavage within the vaspin N-terminus, releasing short peptides with cell penetrating activity. For microarray analysis to determine the effect of a vaspin-derived peptide on gene expression in adipocytes, fully differentiated primary mouse adipocytes from subcutaneous AT were treated with 100 nM of peptide for 24 h before harvesting, RNA isolation and microarray analysis. Solvent (water) treated cells served as controls.

Effect of deletion of carnitine palmitoyltransferase2 on interscapular BAT gene expression upon adrenergic stimulation in thermoneutral acclimatized mice

Project description:Ambient temperature affects energy intake and expenditure to maintain homeostasis in a continuously fluctuating environment. Here, mice with an adipose-specific defect in fatty acid oxidation (Cpt2A-/-) were subjected to varying temperature to determine the role of adipose bioenergetics to environmental adaptation. Cpt2A-/- brown adipose tissue (BAT) failed to induce thermogenic genes such as Ucp1 and Pgc1Î± in response to adrenergic stimulation, which is exacerbated by increasing temperature. Thermoneutrality induced a mitochondrial DNA stress in Cpt2A-/- BAT that resulted in a loss of classical interscapular BAT, but did not affect body weight gain or glucose tolerance in response to a high-fat diet. In this dataset, we include the expression data obtained from dissected mouse interscapular brown adipose tissue from mice acclimatized to thermoneutrality (30C) with and without beta3adrenergic stimulation with and without the deletion of carnitine palmitoyltransferase 2 (i.e., adipose unable to beta-oxidize long chain fatty acids in mitochondria). WildType and Cpt2A KnockOut mice were treated either with or without beta3adrenergic stimulation, thus four classes. Three biologic replicates were compared per class, thus twelve mice.

Transcriptome of human HT1080 cells treated with purified Salmonella Typhi toxin

Project description:Aim of the experiment: To examine changes in gene expression in HT1080 fibroblasts following treatment with purified recombinant wild-type typhoid toxin (TOX) from Salmonella Typhi relative to a mutant H160Q variant lacking DNase activity (TOX-H160Q). Experimental workflow: HT1080 were seeded one day before intoxication into T75 tissue culture flasks for a 30% confluency in DMEM supplemented with 10% FBS and PenStrep. The next day, TOX (5 ng/ml) or negative control TOX-H160Q (5 ng/ml) was incubated for 2h with cells followed by three washes with PBS and a 48h chase in DMEM supplemented with 10% FBS and PenStrep. After 48 h, cells were collected by trypsinisation. RNA was isolated and samples analysed using human Clariom S assay (ThermoFisher Scientific, 902927). Analysis was performed with Transcriptome Analysis Console 4.0 software (Applied Biosystems, Thermo Fisher Scientific).

miRNA mimic transfected SC and MIPC transcriptome analysis

Project description:In the present study, we analyzed two populations of myogenic cells isolated from mouse skeletal muscle, that is, myoblasts derived from satellite cells (SCs) and muscle interstitial progenitor cells (MIPCs). We found that SDF-1 treatment of myoblasts led to down-regulation of miR10a, miR151, miR425, and miR5100. Therefore, to follow the impact of these miRNAs on myogenic cells, SC-derived myoblasts and MIPCs were transfected with selected miRNA mimics in the concentration of 50 mol/L and changes in their transcriptome were analyzed.
