Fibrinogen chain positive circulating exosomes are a colorectal cancer potential biomarker

Project description:Exosomes are small vesicles contain cellular nucleic acids, proteins, lipids and other substances secreted by living cells. The special structure of exosomes makes it possible to protect the inside contents exist in blood stably. Recent researches showed that exosomes are blame for tumor metastatic dissemination, recurrence and malignance, which makes them strong competitors in diagnosis of cancer. The proteomics of exosomes in circulatory system as well as the phosphorylation state of them through different stages of colorectal cancer (CRC) process remain rarely reported. Here we isolate and identify proteins and phosphorylated protein in exosomes from human plasma in healthy donors and different stages of CRC patients by prolonged ultracentrifugation and liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) analysis. In this study, a total of 462 quantitative proteins were identified, meanwhile, the strategy allowed us to identify 177 quantifiable proteins along with 315 sites. Proteins with notable different levels are verified and fibrinogen chain positive (FGA+) circulating exosomes (crExos) from patients and from mice could identify CRC with excellent sensitivity and specificity while perform very well in early diagnosis of adenomas. According to our findings, FGA+ crExos could serve as a potential non-invasive diagnostic and screening tool to detect early stages of CRC to facilitate possible curative surgical therapy.

Proteomic profiling reveals the molecular changes of insomnia patients

Project description:Insomnia is an economic burden and public health problem. This study aimed to explore potential biological pathways and protein networks for insomnia characterized by wakefulness after sleep. Proteomics analysis was performed in the insomnia group with wakefulness and the control group. The differentially expressed proteins (DEPs) were enriched, then hub proteins were identified by protein-protein interaction (PPI) network and verified by parallel reaction monitoring (PRM). Compared with the control group, the sleep time and efficiency of insomnia patients were decreased, awakening time and numbers after sleep onset were significantly increased (P < 0.001). The results of proteomic sequencing found 68 DEPs in serum under 1.2-fold changed standard. These DEPs were significantly enriched in humoral immune response, complement and coagulation cascades, cholesterol metabolism. Through PPI network, we identified 10 proteins with the highest connectivity as hub proteins. Among them, differential expression of 9 proteins was verified by PRM.We identified the hub proteins and molecular mechanisms of insomnia patients characterized by wakefulness after sleep. It provided potential molecular targets for the clinical diagnosis and treatment of these patients, and indicated the immune and metabolic systems may be closely related to insomnia characterized by wakefulness after sleep.

Proteomic Profiling Identifies Novel Diagnostic Biomarkers and Molecular subtypes for Mucinous Tubular and Spindle Cell Carcinoma of the Kidney

Project description:Mucinous tubular and spindle cell carcinoma (MTSCC) is a relatively rare renal epithelial neoplasm resembling type 1 papillary renal cell carcinoma (PRCC) morphologically and immunohistochemically. The accurate diagnosis of MTSCC remains a challenge. Here, by using proteomic profiling, we characterized MTSCC and PRCC to identify diagnostic biomarkers. We found that MTSCC tumor proteome was significantly enriched in B cell-mediated immunity when compared with the proteome of adjacent normal tissues of MTSCC or tumors of PRCC. Importantly, we identified MZB1, VCAN, and SOSTDC1 as diagnostic biomarkers to distinguish MTSCC from PRCC, with an AUC of 0.9839 when combined. MZB1 was inversely correlated with tumor clinical stage and may play an anti-tumor role by activating the complement system. Finally, unsupervised clustering revealed two molecular subtypes of MTSCC, displaying different morphology, expression signatures of oxidative phosphorylation, and aggravation. In summary, our analyses identified a three-protein diagnostic panel and molecular subtypes for MTSCC.

Protein Profile Changes in the Frontotemporal Lobes in Human Severe Traumatic Brain Injury

Project description:Severe traumatic brain injury (sTBI) is a serious public health issue with high morbidity and mortality rates. Previous proteomic studies on sTBI have mainly focused on human cerebrospinal fluid and serum, as well as on brain protein changes in murine models. However, human proteomic data in sTBI brain is still needed. We used proteomics and bioinformatics strategies to investigate variations in protein expression in human brains after sTBI, using samples from the Department of Neurosurgery, Affiliated Hospital of Hebei University (Hebei, P.R. China). Our proteomics data identified 4031 proteins, of which 162 proteins were overexpressed and 5 proteins were downregulated. The biological pathways that showed significant changes in protein expression according to bioinformatics analysis were glial cell differentiation, complement activation, apolipoprotein catalysis in statin pathway, and the blood coagulation cascade. Western blot verification of protein changes in a subset of the available tissue samples showed results that were consistent with the proteomics data. This study is one of the first to investigate the whole proteome of human sTBI brains, and provides a characteristic signature and overall landscape of the sTBI brain proteome.
