ABSTRACT: Monohalo and dihalo 1,3-thiazole derivatives can be efficiently and selectively prepared under mild conditions from 2-amino-1,3-thiazoles. Halogenations proceed easily in the presence of copper(I) or copper(II) chlorides, bromides, or iodides directly in solution or with supported copper halides.

Design, development and applications of copper-catalyzed regioselective (4 + 2) annulations between diaryliodonium salts and alkynes.

Project description:Diaryliodonium salts have been extensively applied in organic synthesis as aryl cation equivalents. However, in the electrophilic reactions with alkenes or alkynes, only the electrophilic carbon of the diaryliodonium salts was involved while the other part of the aryl ring was not utilized. Herein, a reaction pattern of diaryliodonium was reported as oxa-1,4-dipoles to undergo (4 + 2) cycloaddition reactions with alkynes. Broad spectrum of the two reaction partners could be utilized in this protocol, enabling an operationally simple, high yielding, and regioselective synthetic approach to isocoumarins. Particularly, good to excellent regioselectivities were achieved for the sterically unbiased unsymmetrical diaryl acetylenes, which was challenging for other transition metal-catalyzed processes. The reaction could be scaled up with the ideal 1:1 stoichiometry and the isocoumarin type natural products Oospolactone and Thunberginol A could be obtained in one or three steps through this methodology.

An umpolung-enabled copper-catalysed regioselective hydroamination approach to α-amino acids.

Project description:A copper-catalysed regio- and stereoselective hydroamination of acrylates with hydrosilanes and hydroxylamines has been developed to afford the corresponding α-amino acids in good yields. The key to regioselectivity control is the use of hydroxylamine as an umpolung, electrophilic amination reagent. Additionally, a judicious choice of conditions involving the CsOPiv base and DTBM-dppbz ligand of remote steric hindrance enables the otherwise challenging C-N bond formation at the α position to the carbonyl. The point chirality at the β-position is successfully controlled by the Xyl-BINAP or DTBM-SEGPHOS chiral ligand with similarly remote steric bulkiness. The combination with the chiral auxiliary, (-)-8-phenylmenthol, also induces stereoselectivity at the α-position to form the optically active unnatural α-amino acids with two adjacent stereocentres.

Copper Salts/TBAB-Catalyzed Chemo- and Regioselective β-C(sp<sup>3</sup>)-H Acyloxylation of Aliphatic Amides.

Project description:An efficient Cu(II)-catalyzed and tetrabutylammonium bromide (TBAB)-promoted strategy for highly regioselective and chemoselective C(sp3)-H acyloxylation of aliphatic amides is described. Acyloxylation occurs selectively at the β position with a broad substrate scope of carboxylic acids and aliphatic amides and good functional group compatibility. Notably, the competing reaction of intramolecular dehydrogenative amidation and intermolecular acyloxylation could be efficiently controlled by the amount of copper salt and the addition of TBAB. The intramolecular dehydrogenative amidation product was obtained in high yield when the amount of copper salts was increased. However, when TBAB was used as an additive, a preference for acyloxylation over intramolecular amidation was observed and the acyloxylated products were obtained in good yield. Preliminary studies were carried out to gain insights into the mechanism.

Regioselective Iodination of Chlorinated Aromatic Compounds Using Silver Salts.

Project description:The iodination of chlorinated aromatic compounds using Ag(2)SO(4)/I(2), AgSbF(6)/I(2), AgBF(4)/I(2) and AgPF(6)/I(2) offers access to iodoarenes that are valuable intermediates in organic synthesis. Specifically, iodination of phenols, anisoles and anilines with a 3,5-dichloro substitution pattern preferentially yielded the ortho, para and para iodinated product, respectively. In the case of chlorobenzene and 3-chlorotoluene, AgSbF(6)/I(2), AgBF(4)/I(2) and AgPF(6)/I(2), but not Ag(2)SO(4)/I(2), selectively introduced the iodine in para position to the chlorine substituent.

Regioselective and Diastereoselective Aminoarylation of 1,3-Dienes.

Project description:The 1,4-functionalization of dienes is a synthetically useful strategy for incorporating molecular complexity into a class of simple substrates. We report the aminoarylation of acyclic and cyclic 1,3-dienes via the sequential [4+2] cycloaddition with a sulfurdiimide reagent and copper-catalyzed allylic substitution with Grignard reagents. The regioselective and diastereoselective aminoarylation of unsymmetrical dienes is also presented, which highlights the utility of this method for generating products with multiple functional groups and stereocenters.

Project description:In the title compound, [Cu(C(3)H(10)N(2))(2)(C(6)H(6)NO(3)S)(2)]·2H(2)O, the Cu(II) atom lies on an inversion center and is hexa-coordinated by four N atoms from two 1,3-diamino-propane ligands and two O atoms from two 4-amino-benzene-sulfonate ligands in a trans arrangement, displaying a distorted and axially elongated octa-hedral coordination geometry, with the O atoms at the axial positions. A three-dimensional network is formed in the crystal structure through O-H?O, N-H?O and N-H?N hydrogen bonds.
