Unprecedented rearrangement of 2-(2-aminoethyl)-1-aryl-3,4-dihydropyrazino[1,2-b]indazole-2-ium 6-oxides to 2,3-dihydro-1H-imidazo[1,2-b]indazoles.

ABSTRACT: Easily accessible 2-(2-aminoethyl)-1-aryl-3,4-dihydropyrazino[1,2-b]indazole-2-ium 6-oxides rearranged to 2,3-dihydro-1H-imidazo[1,2-b]indazoles under mild conditions. The rearrangement appeared to be general, tolerated a wide range of functional groups, and provided access to an as yet unexplored class of heterocycles. Herein we report the characterization of these heterocycles.

Mean planes fitted through all non-H atoms of each ring system have an r.m.s. deviation of 0.019?Å for the imidazole-based ring and 0.016?Å for the 2,6-dimethyl-phenyl ring. The angle between the two planes is 86.76?(2)°. In the crystal structure, N-H?O inter-actions form a one-dimensional chain, which propagates in the b-axis direction. C-H?O inter-actions are also found in the crystal packing.

Project description:A highly efficient novel traceless solid-phase synthesis of 3,4-dihydropyrazino[1,2-b]indazoles and their 6-oxides was developed by using commercially available building blocks, diamines, 2-nitrobenzenesulfonyl chlorides, and bromoketones/bromoacetates. Project description:A copper(I)-mediated one-pot synthesis of 2,3-dihydro-1H-indazole heterocycles has been developed. This synthetic route provides the desired indazoles in moderate to good yields (55%-72%) which are substantially better than those achievable with an alternative two-step reaction sequence. The reaction is tolerant of functionality on the aromatic ring.
