ABSTRACT: The title compound, C(41)H(70)O(12)·CH(4)O, was prepared by microbial transformation. Within the steroid skeleton of the mol-ecule, three six-membered rings exhibit a chair conformation, while the five -membered ring adopts an envelope conformation. The two pyranosyl rings also adopt chair conformations. The mol-ecules are held together by an extensive O-H?O hydrogen-bonding network.

Project description:Studies have shown that a major metabolite of the red ginseng ginsenoside Rb1, called 20-O-β-D-glucopyranosyl-20(S)-protopanaxadiol (GPD), exhibits anticancer properties. However, the chemotherapeutic effects and molecular mechanisms behind GPD action in human melanoma have not been previously investigated. Here we report the anticancer activity of GPD and its mechanism of action in melanoma cells. GPD, but not its parent compound Rb1, inhibited melanoma cell proliferation in a dose-dependent manner. Further investigation revealed that GPD treatment achieved this inhibition through the induction of autophagy and apoptosis, while Rb1 failed to show significant effect at the same concentrations. The inhibitory effect of GPD appears to be mediated through the induction of AMPK and the subsequent attenuation of mTOR phosphorylation. In addition, GPD activated c-Jun by inducing JNK phosphorylation. Our findings suggest that GPD suppresses melanoma growth by inducing autophagic cell death and apoptosis via AMPK/JNK pathway activation. GPD therefore has the potential to be developed as a chemotherapeutic agent for the treatment of human melanoma.

Three O atoms of the perchlorate anion are disordered over two positions with refined occupancy factors of 0.649?(7):0.351?(7).

Project description:The title compound, C(8)H(12)O(7)P(2)·CH(4)O, is a monoesterified bis-phospho-nate (or 1-hydroxy-methyl-ene-1,1-bis-phospho-nic acid). These synthetic compounds are widely used in medicine to inhibit bone resorption in diseases like osteoporosis, and are characterized by a stable P-C-P group and are thus analogs of inorganic pyrophosphate. By masking one or several ionizable groups, introduced as phosphono-ester, it was anti-cipated the formation of prodrugs with higher lipophilicity that could facilitate the drug delivery and metabolization. Mol-ecules are paired by inter-molecular hydrogen bonds involving the phospho-nic groups. In addition, dimers are connected side-by-side, building infinite ribbons along the a-axis direction; these ribbons are cross-linked perpendicularly along the b-axis direction via a methanol solvent mol-ecule (disordered over two sites with occupancy factors ca 0.6 and 0.4), forming an extended inter-molecular hydrogen-bonded network. The H atoms of the methyl group in the main molecule are disordered equally over two positions.

Project description:The title complex, [Re(C(7)H(2)Br(3)O(2))(CO)(3)(H(2)O)]·CH(3)OH, crystallized as a neutral Re(I) compound and one methanol solvent mol-ecule in the asymmetric unit. The metal centre is coordinated facially by three carbonyl groups. The bidentate tribromo-tropolanate ligand and a water mol-ecule complete the distorted octahedral coordination around the central metal. Inter-molecular Br⋯O [3.226 (5) Å] and Br⋯Br [3.590 (2) Å] contacts are observed between adjacent mol-ecules. These contacts, together with an array of O-H⋯O, O-H⋯Br and C-H⋯O hydrogen bonds, complete a three-dimensional polymeric network formed between the methanol solvent and the complex.

The most notable feature of the crystal packing is the formation of supra-molecular chains along the b direction mediated by O-H?N hydrogen bonds occurring between the hydr-oxy and quinoline N atoms; the methanol mol-ecules are linked to these chains via O-H?N(amine) hydrogen bonds. C-H?O inter-actions also occur.

Chloridotris[tris-(4-fluoro-phen-yl)phosphine]rhodium(I) methanol solvate.

Project description:In the title compound, [RhCl{P(p-FC(6)H(4))(3)}(3)]·CH(3)OH, the Rh atom adopts a distorted square-planar geometry. Rh, Cl and one P atom lie on a mirror plane, as does the solvent molecule. There are two inter-molecular hydrogen bonds, one between the methanol O atom and an aryl H atom (2.51 Å), and one between the Cl atom and the hydr-oxy H atom of methanol [2.34 (3) Å]. The complex precipitates in trace amounts from a reaction between RhCl(cod)(thp) [cod is 1,5-cyclo-octa-diene and thp is tris-(hydroxy-meth-yl)phos-phine] and P(p-FC(6)H(4))(3) under argon in CD(3)OD. Two C(6)H(4)-F units are disordered over two positions; for one the site occupancy factors are ca. 0.53 and 0.47, for the other the values are ca. 0.64 and 0.36. The methyl H atoms of the solvent molecule are disordered across the mirror plane.
