ABSTRACT: The reaction of cis-[Pt(NO(3))(2)(NH(3))(2)] and sodium glycolate yielded the title compound, [Pt(C(2)H(2)O(3))(NH(3))(2)]. The Pt(II) atom, coordinated by two N atoms of ammine and two O atoms of the carboxyl-ate and oxido groups of the glycolate ligand, is in a square-planar environment. In the crystal structure, mol-ecules are connected by inter-molecular N-H?O hydrogen bonds, forming a three-dimensional network.

Project description:We have identified unique chemical and biological properties of a cationic monofunctional platinum(II) complex, cis-diammine(pyridine)chloroplatinum(II), cis-[Pt(NH(3))(2)(py)Cl](+) or cDPCP, a coordination compound previously identified to have significant anticancer activity in a mouse tumor model. This compound is an excellent substrate for organic cation transporters 1 and 2, also designated SLC22A1 and SLC22A2, respectively. These transporters are abundantly expressed in human colorectal cancers, where they mediate uptake of oxaliplatin, cis-[Pt(DACH)(oxalate)] (DACH = trans-R,R-1,2-diaminocyclohexane), an FDA-approved first-line therapy for colorectal cancer. Unlike oxaliplatin, however, cDPCP binds DNA monofunctionally, as revealed by an x-ray crystal structure of cis-{Pt(NH(3))(2)(py)}(2+) bound to the N7 atom of a single guanosine residue in a DNA dodecamer duplex. Although the quaternary structure resembles that of B-form DNA, there is a base-pair step to the 5' side of the Pt adduct with abnormally large shift and slide values, features characteristic of cisplatin intrastrand cross-links. cDPCP effectively blocks transcription from DNA templates carrying adducts of the complex, unlike DNA lesions of other monofunctional platinum(II) compounds like {Pt(dien)}(2+). cDPCP-DNA adducts are removed by the nucleotide excision repair apparatus, albeit much less efficiently than bifunctional platinum-DNA intrastrand cross-links. These exceptional characteristics indicate that cDPCP and related complexes merit consideration as therapeutic options for treating colorectal and other cancers bearing appropriate cation transporters.

Enhancement of cisplatin [cis-diammine dichloroplatinum (II)] cytotoxicity by O6-benzylguanine involves endoplasmic reticulum stress.

Project description:O6-Benzylguanine (BG) enhances cisplatin [cis-diammine dichloroplatinum (II)]-induced cytotoxicity and apoptosis in head and neck cancer cell lines by an unknown mechanism. We investigated the effect of cisplatin with and without BG on two targets of damage: DNA and the endoplasmic reticulum (ER). We chose three cancer cell lines to ascertain the mechanism of BG-enhanced cytotoxicity: SQ20b head and neck and SKOV-3x ovarian cancer cell lines, where BG enhanced cisplatin cytotoxicity, and A549 nonsmall cell lung cancer line, where BG did not enhance cisplatin cytotoxicity. All three lines had an increase in DNA damage when BG was added to cisplatin treatment, as evidenced by increased platination and phosphorylated histone H2AX formation. The increase in cisplatin-induced DNA damage after treatment with BG plus cisplatin is not sufficient to increase cytotoxicity or apoptosis in A549 cells. We evaluated the effect of cisplatin on the ER and observed increased caspase 12 cleavage in SQ20b and SKOV-3x cells, but not in A549 cells, after treatment with BG plus cisplatin versus cisplatin alone. Growth arrest and DNA damage inducible (GADD) 153, an ER stress-response gene, is up-regulated after treatment with BG plus cisplatin compared with cisplatin alone in SQ20b and SKOV-3x cells, but not in A549 cells. ER stress-induced apoptosis is an integral part of the mechanism by which BG enhances cisplatin. Inhibition of ER stress in the SQ20b cell line by salubrinal, an inhibitor of eIF2alpha dephosphorylation, or GADD153 small interfering RNA, abrogated BG-enhancement of cisplatin cytotoxicity and apoptosis through caspase 3 and 12 cleavage. These data indicate GADD153 up-regulation plays an important role in BG-enhanced cisplatin cytotoxicity and apoptosis.

Project description:In the title complex, cis-[PtCl(2)(C(11)H(9)N)(2)], the Pt(II) ion is situated in a slightly distorted square-planar environment coordinated by two N atoms from two 2-phenyl-pyridine ligands and two Cl atoms. The two pyridyl planes are inclined with dihedral angles of 59.1?(2) and 61.84?(19)° with respect to the PtCl(2)N(2) plane. In the crystal, the complex mol-ecules display inter- and intra-molecular ?-? stacking inter-actions, with centroid-centroid distances of 3.806?(5)-3.845?(5)?Å, which form a one-dimensional column structure along the a axis.

Project description:In the crystal structure of the title complex, [Pt(C(5)H(4)NOS)(2)], the Pt atom is coordinated by two O atoms and two S atoms in a cis configuration, forming a distorted square-planar coordination geometry. The mol-ecule exhibits pseudo-C(2v) symmetry and is essentially planar, with a maximum deviation from planarity of 0.0124 (2) Å. The dihedral angle between the two pyridine rings is 5.85 (2)°.

Project description:In the title complex, [PtBr(2)(C(11)H(9)N)(2)], the Pt(II) ion has a distorted cis-Br(2)N(2) square-planar coordination geometry defined by two N atoms from two 2-phenyl-pyridine (ppy) ligands and two Br(-) anions. The ppy ligands are not planar, the dihedral angles between the pyridine and benzene rings being 49.0 (3) and 47.3 (3)°. In the crystal, the complex mol-ecules are stacked in columns along the a axis. In the columns, there are numerous intra- and inter-molecular π-π inter-actions between the six-membered rings, the shortest ring centroid-centroid distance being 3.774 (6) Å.

Crystal structure of cis-diamminebis(nitrito-κN)platinum(II).

Project description:Single crystals of cis-[Pt(NO2)2(NH3)2], were obtained by means of hyper-saturation directly out of a plating electrolyte. The square-planar coordination environment of the divalent Pt(II) atom is formed by four N atoms belonging to two ammine and two monodentate nitrite ligands. The ligands adopt a cis configuration. The crystal structure contains stacks of close-packed mol-ecules which run parallel to [001]. There are nine crystallographically independent inter-molecular N-H⋯O hydrogen bonds, resulting in a hydrogen-bonded hxl-type framework in which each mol-ecule serves as an eight-connected node. Four of the nine distinct hydrogen bonds connect complexes which belong to the same close-packed column parallel to [001]. In contrast to the previously reported crystal structure of the trans isomer, the title structure does not display intra-molecular hydrogen bonding.

Project description:In the title compound, [PtCl(2)(C(9)H(7)N)(2)]·CH(3)NO(2), the Pt(II) cation is four-coordinated in an essentially square-planar environment by two N atoms from two quinoline ligands and two Cl(-) anions. One of the nearly planar quinoline ligands [maximum deviations = 0.042?(6) and 0.018?(7)?Å] is almost perpendicular to the PtCl(2)N(2) unit [maximum deviation = 0.024?(3)?Å], making a dihedral angle of 89.6?(1)°, whereas the other is slightly inclined to the central plane with a dihedral angle of 74.1?(1)°. The dihedral angle between the quinoline ligands is 88.3?(2)°. In the crystal, each solvent mol-ecule is linked to the metal complex by weak inter-molecular C-H?O hydrogen bonds.
