Project description:In the title compound, C14H16ClNO2, the cyclo-hexyl ring has a chair conformation. The dihedral angle between the benzene ring and the mean plane of the four planar C atoms of the cyclo-hexyl ring is 45.2 (3)°. The two carbonyl groups are trans to one another, with an O=C-C=O torsion angle of -137.1 (3)°. In the crystal, mol-ecules are linked by N-H⋯O hydrogen bonds forming chains propagating along [001]. A region of disordered electron density, situated near the unit-cell corners, was treated using the SQUEEZE routine in PLATON [Spek (2009 ▶). Acta Cryst. D65, 148-155]. It gave a solvent-accessible void of ca 400 Å(3) for only 21 electrons. It is probably due to traces of the solvent of crystallization and was not taken into account during structure refinement.

The effect of hydrogen bonding on the conformations of 2-(1H-indol-3-yl)-2-oxoacetamide and 2-(1H-indol-3-yl)-N,N-dimethyl-2-oxoacetamide.

Project description:In the title compounds, C(10)H(8)N(2)O(2), (I), and C(12)H(12)N(2)O(2), (II), the two carbonyl groups are oriented with torsion angles of -149.3 (3) and -88.55 (15)°, respectively. The single-bond distances linking the two carbonyl groups are 1.528 (4) and 1.5298 (17) Å, respectively. In (I), the molecules are linked by an elaborate system of N-H···O hydrogen bonds, which form adjacent R(2)(2)(8) and R(4)(2)(8) ring motifs to generate a ladder-like construct. Adjacent ladders are further linked by N-H···O hydrogen bonds to build a three-dimensional network. The hydrogen bonding in (II) is far simpler, consisting of helical chains of N-H···O-linked molecules that follow the 2(1) screw of the b axis. It is the presence of an elaborate hydrogen-bonding system in the crystal structure of (I) that leads to the different torsion angle for the orientation of the two adjacent carbonyl groups from that in (II).

In the crystal, pairs of mol-ecules are connected via N-H⋯N hydrogen bonds into inversion dimers. The mol-ecules are characterized by a planar indole moiety [maximum deviation = 0.012 (1) Å], a chair conformation of the cyclo-hexane ring and an anti-periplanar conformation of the H atom on the cyclo-hexane and the adjacent cyano group.

In the mol-ecules, the dihedral angles between the benzene rings and indole mean planes are 24.5 (1), 22.5 (1), 8.8 (1) and 13.9 (1)°. In the crystal, inter-molecular N-H⋯O hydrogen bonds are present between the imino groups and the adjacent carbonyl groups. π-π stacking is also observed with a centroid-centroid distance between nearly parallel pyrrole rings of 3.745 (3) Å.

Novel N-Substituted 2-(2-(Adamantan-1-yl)-1H-Indol-3-yl)-2-Oxoacetamide Derivatives: Synthesis and Biological Evaluation.

Project description:In this study, a series of novel N-substituted 2-(2-(adamantan-1-yl)-1H-indol-3-yl)-2-oxoacetamide derivatives were synthesized, and evaluated for their cytotoxicity in human cell lines including Hela (cervical cancer), MCF7 (breast cancer ) and HepG2 (liver cancer). Several compounds were found to have potent anti-proliferative activity against those human cancer cell lines and compound 5r showed the most potent biological activity against HepG2 cells with an IC50 value of 10.56 ± 1.14 ??. In addition, bioassays showed that compound 5r induced time-dependent and dose-dependent cleavage of poly ADP-ribose polymerase (PARP), and also induced a dose-dependent increase in caspase-3 and caspase-8 activity, but had little effect on caspase-9 protease activity in HepG2 cells. These results provide evidence that 5r-induced apoptosis in HepG2 cell is caspase-8-dependent.

Project description:In the title mol-ecule, C(13)H(16)FNO, the amide (N-C=O) plane is oriented at an angle of 29.9 (2)° with respect to the aromatic ring. The cyclo-hexane ring adopts the usual chair conformation. In the crystal structure, inter-molecular N-H⋯O hydrogen bonds link the mol-ecules into chains along [100]. A weak C-H⋯F inter-action is also observed. The F atom is disordered over two positions with occupancy factors of 0.873 (3) and 0.127 (3).
