Project description:Allyl and vinyl chlorides represent important structural motifs in organic chemistry. Herein is described the chemoselective and regioselective reaction of aryl- and ?-substituted phenylallenes with the hypervalent iodine (HVI) reagent 1-chloro-1,2-benziodoxol-3-one. The reaction typically results in vicinal dichlorides, except with proton-containing ?-alkyl substituents, which instead give chlorinated dienes as the major product. Experimental evidence suggests that a radical mechanism is involved.

Enantio- and Diastereoselective Copper-Catalyzed Allylboration of Alkynes with Allylic gem-Dichlorides.

Project description:Allylic gem-dichlorides are shown to be efficient substrates for catalytic asymmetric allylboration of alkynes. The method employs a chiral NHC-Cu catalyst capable of generating in a single step chiral skipped dienes bearing a Z-alkenyl chloride, a trisubstituted E-alkenyl boronate and a bis-allylic stereocenter with excellent levels of chemo-, regio- enantio- and diastereoselectivity. This high degree of functionalization makes these products versatile building blocks as illustrated with the synthesis of several optically active compounds. DFT calculations support the key presence of a metal cation bridge ligand-substrate interaction and account for the stereoselectivity outcome.

A general synthesis of dendralenes.

Project description:The first general synthetic approach to substituted [3]- and higher dendralenes is reported. Fifty-one mono- through to penta-substituted dendralenes carrying alkyl-, cycloalkyl-, alkenyl-, alkynyl-, aryl- and heteroaryl-substitutents are accessed, and the first (E)/(Z)-stereoselective syntheses of dendralenes are reported (twenty-eight examples). The approach involves twofold Pd(0)-catalyzed Negishi couplings of 1,1-dibromoalkenes with alkenylzinc reagents, and exploits both substrate- and catalyst-controlled aspects of chemo-, regio- and stereoselectivity in the two C(sp2)-C(sp2) bond forming steps. The value of the new hydrocarbons in rapid structural complexity generation is demonstrated through their deployment in unprecedented diene- and triene-transmissive pericyclic reaction sequences.

A simple, modular synthesis of substituted pyridines.

Project description:A simple, modular method to prepare highly substituted pyridines is disclosed. The method employs a cascade reaction comprising (1) a novel N-iminative, Cu-catalyzed cross-coupling of alkenylboronic acids at the N-O bond of alpha,beta-unsaturated ketoxime O-pentafluorobenzoates, (2) electrocyclization of the resulting 3-azatriene, and (3) air oxidation affording highly substituted pyridines in moderate to excellent isolated yields (43-91%). Starting materials are readily available, and functional group tolerance is very good.

Simple sulfinate synthesis enables C-H trifluoromethylcyclopropanation.

Project description:A simple method to convert readily available carboxylic acids into sulfinate salts by employing an interrupted Barton decarboxylation reaction is reported. A medicinally oriented panel of ten new sulfinate reagents was created using this method, including a key trifluoromethylcyclopropanation reagent, TFCS-Na. The reactivity of six of these salts towards C-H functionalization was field-tested using several different classes of heterocycles.

A simple synthesis of nitriles from aldoximes.

Project description:Easily synthesized aldoximes have been converted to the corresponding nitriles under very mild conditions by a simple reaction with 1H-benzotriazol-1-yloxytris(dimethylamino)phosphonium hexafluorophosphate (BOP) and DBU in CH(2)Cl(2), THF, or DMF. As an alternative reagent that eliminates the formation of hexamethylphosphoramide as a byproduct, use of 1H-benzotriazol-1-yl-4-methylbenzenesulfonate (Bt-OTs) and DBU was investigated. Reactions with this reagent also proceeded smoothly and in good yields, although in one case N-sulfonylation was observed. An attempt to gain mechanistic insight into the BOP-mediated reaction has been made using (31)P{(1)H} NMR. However, no phosphorus-bearing intermediate could be readily observed. Finally, the method has been applied to the synthesis of an antiviral 4'-cyano adenosine analogue from a commercial precursor using a single saccharide protecting group.

A simple and general strategy for generating frequency-anticorrelated photon pairs.

Project description:Currently, two-photon excitation microscopy is the method of choice for imaging living cells within thick specimen. A remaining problem for this technique is the damage caused by the high photon flux in the excitation region. To reduce the required flux, a promising solution is to use highly frequency-anticorrelated photon pairs, which are known to induce two-photon transitions much more efficiently. It is still an open question what the best scheme is for generating such photon pairs. Here we propose one simple general strategy for this task. As an example, we show explicitly that this general strategy can be realized faithfully within the widely applicable coherently pumped Jaynes-Cummings model. It is shown quantitatively that this strategy can generate highly frequency-anticorrelated photon pairs which can dramatically enhance two-photon excitation efficiency. We believe the proposed strategy can guide new designs for generating frequency-anticorrelated photon pairs.

Toward a general synthesis of chlorins.

Project description:Recently, we described a new synthesis of C,D-ring symmetric chlorins 11, involving 2 + 2 condensation of bis-formyl-dihydrodipyrrins 9 with symmetrically substituted dipyrromethane diacids 10 (Method I). However, while versatile in many aspects, Method I was unsuited to the broader goal of synthesizing fully non-symmetric chlorins of general structure 15, which requires regioselective control over the reacting centers in the A,B- and C,D-ring components. In this paper, we describe four new 2 + 2 strategies that accomplish this differentiation (Methods II-V). Of these, Method V, which combines operational simplicity with moderate to high product yields, proved to be the most effective route, exploiting reactivity differences between the two formyl groups of A,B-rings 9 to impart excellent regioselectivity. Methods II-IV are also useful alternatives to Method V, although in some cases, the appropriately functionalized precursors are less readily available. All four approaches generate single regioisomers of diversely substituted chlorins, and in every case, the 2 + 2 condensation is accomplished in a simple, one-flask procedure without need for additives such as oxidizing agents or metals. Taken together, these methodologies provide expanded access to an array of chlorins for SAR studies that may advance the effectiveness of PDT and other applications.
