ABSTRACT: 2-Thiohydantoin derivatives are produced by heating a mixture of thiourea and an alpha-amino acid. The method described offers the advantages of simplicity, low cost, easy work-up and scalability.

A simple, modular synthesis of substituted pyridines.

Project description:A simple, modular method to prepare highly substituted pyridines is disclosed. The method employs a cascade reaction comprising (1) a novel N-iminative, Cu-catalyzed cross-coupling of alkenylboronic acids at the N-O bond of alpha,beta-unsaturated ketoxime O-pentafluorobenzoates, (2) electrocyclization of the resulting 3-azatriene, and (3) air oxidation affording highly substituted pyridines in moderate to excellent isolated yields (43-91%). Starting materials are readily available, and functional group tolerance is very good.

Simple sulfinate synthesis enables C-H trifluoromethylcyclopropanation.

Project description:A simple method to convert readily available carboxylic acids into sulfinate salts by employing an interrupted Barton decarboxylation reaction is reported. A medicinally oriented panel of ten new sulfinate reagents was created using this method, including a key trifluoromethylcyclopropanation reagent, TFCS-Na. The reactivity of six of these salts towards C-H functionalization was field-tested using several different classes of heterocycles.

A simple synthesis of nitriles from aldoximes.

Project description:Easily synthesized aldoximes have been converted to the corresponding nitriles under very mild conditions by a simple reaction with 1H-benzotriazol-1-yloxytris(dimethylamino)phosphonium hexafluorophosphate (BOP) and DBU in CH(2)Cl(2), THF, or DMF. As an alternative reagent that eliminates the formation of hexamethylphosphoramide as a byproduct, use of 1H-benzotriazol-1-yl-4-methylbenzenesulfonate (Bt-OTs) and DBU was investigated. Reactions with this reagent also proceeded smoothly and in good yields, although in one case N-sulfonylation was observed. An attempt to gain mechanistic insight into the BOP-mediated reaction has been made using (31)P{(1)H} NMR. However, no phosphorus-bearing intermediate could be readily observed. Finally, the method has been applied to the synthesis of an antiviral 4'-cyano adenosine analogue from a commercial precursor using a single saccharide protecting group.

Simple synthesis of nanosheets of rGO and nitrogenated rGO.

Project description:A green and facile approach has been developed for the large-scale synthesis of nanosheets of reduced graphene oxide (rGO) and nitrogenated reduced graphene oxide (N-rGO). This has been achieved by direct thermal decomposition of sucrose and glycine at 475 °C in ca. 7 minutes, respectively. The present protocols for synthesizing rGO and N-rGO are simple and environmentally friendly as we do not use any harmful reagents, metal catalysts and solvents. Along with that, this method offers an inexpensive route with high yields to prepare rGO with a high nitrogen content (20-25 atom %). To further improve the properties of the synthesized rGO sheets, hydrogen treatment has been carried out to reduce the oxygen functional groups. Cyclic voltammograms and charge-discharge experiments have been carried out to understand the supercapacitor behavior of rGO and hydrogen treated (H-rGO) samples.

Prebiotic synthesis of simple sugars by photoredox systems chemistry.

Project description:A recent synthesis of activated pyrimidine ribonucleotides under prebiotically plausible conditions relied on mixed oxygenous and nitrogenous systems chemistry. As it stands, this synthesis provides support for the involvement of RNA in the origin of life, but such support would be considerably strengthened if the sugar building blocks for the synthesis--glycolaldehyde and glyceraldehyde--could be shown to derive from one carbon feedstock molecules using similarly mixed oxygenous and nitrogenous systems chemistry. Here, we show that these sugars can be formed from hydrogen cyanide by ultraviolet irradiation in the presence of cyanometallates in a remarkable systems chemistry process. Using copper cyanide complexes, the process operates catalytically to disproportionate hydrogen cyanide, first generating the sugars and then sequestering them as simple derivatives.

Multistep synthesis of complex boronic acids from simple MIDA boronates.

Project description:Due to its sensitivity to most synthetic reagents, it is typically necessary to introduce the boronic acid functional group just prior to its utilization. Overcoming this important limitation, we herein report that air- and chromatographically stable MIDA boronates are compatible with a wide range of common reagents which enables the multistep synthesis of complex boronic acid building blocks from simple B-containing starting materials. X-ray and variable temperature NMR studies link the unique stability of MIDA boronates to a kinetic inaccessibility of the potentially reactive boron p-orbital and/or nitrogen lone pair. These findings were collectively harnessed to achieve a short and modular total synthesis of (+)-crocacin C via the iterative cross-coupling of a structurally complex, MIDA-protected haloboronic acid building block.

A Simple Route for Synthesis of 4-Phospho-D-Erythronate.

Project description:4-Phospho-D-erythronate is an intermediate in synthesis of pyridoxal 5'-phosphate in some bacteria and an inhibitor of ribose 5-phosphate isomerase. Previous synthetic schemes for the preparation of 4-phospho-D-erythronate required expensive precursors and typically gave low yields. We report a straightforward synthesis of 4-phospho-D-erythronate from the inexpensive precursor D-erythronolactone in 5 steps with a preparatively useful yield of 22%.
