Project description:BackgroundCholesterol oxidase has numerous biomedical and industrial applications. In the current study, a new bacterial strain was isolated from sewage and was selected for its high potency for cholesterol degradation (%) and production of high cholesterol oxidase activity (U/OD600).ResultsBased on the sequence of 16S rRNA gene, the bacterium was identified as Bacillus subtilis. The fermentation conditions affecting cholesterol degradation (%) and the activity of cholesterol oxidase (U/OD600) of B. subtilis were optimized through fractional factorial design (FFD) and response surface methodology (RSM). According to this sequential optimization approach, 80.152% cholesterol degradation was achieved by setting the concentrations of cholesterol, inoculum size, and magnesium sulphate at 0.05 g/l, 6%, and 0.05 g/l, respectively. Moreover, 85.461 U of cholesterol oxidase/OD600 were attained by adjusting the fermentation conditions at initial pH, 6; volume of the fermentation medium, 15 ml/flask; and concentration of cholesterol, 0.05 g/l. The optimization process improved cholesterol degradation (%) and the activity of cholesterol oxidase (U/OD600) by 139% and 154%, respectively. No cholesterol was detected in the spectroscopic analysis of the optimized fermented medium via gas chromatography-mass spectroscopy (GC-MS).ConclusionThe current study provides principal information for the development of efficient production of cholesterol oxidase by B. subtilis that could be used in various applications.

Project description:BackgroundPolyhydroxybutyrate (PHB) is a biopolymer formed by some microbes in response to excess carbon sources or essential nutrient depletion. PHBs are entirely biodegradable into CO2 and H2O under aerobic and anaerobic conditions. It has several applications in various fields such as medicine, pharmacy, agriculture, and food packaging due to its biocompatibility and nontoxicity nature.ResultIn the present study, PHB-producing bacterium was isolated from the Dirout channel at Assiut Governorate. This isolate was characterized phenotypically and genetically as Bacillus cereus SH-02 (OM992297). According to one-way ANOVA test, the maximum PHB content was observed after 72 h of incubation at 35 °C using glucose and peptone as carbon and nitrogen source. Response surface methodology (RSM) was used to study the interactive effects of glucose concentration, peptone concentration, and pH on PHB production. This result proved that all variables have a significant effect on PHB production either independently or in the interaction with each other. The optimized medium conditions with the constraint to maximize PHB content and concentration were 22.315 g/L glucose, and 15.625 g/L peptone at pH 7.048. The maximum PHB content and concentration were 3100.799 mg/L and 28.799% which was close to the actual value (3051 mg/l and 28.7%). The polymer was identified as PHB using FTIR, NMR, and mass spectrometry. FT-IR analysis showed a strong band at 1724 cm- 1 which attributed to the ester group's carbonyl while NMR analysis has different peaks at 169.15, 67.6, 40.77, and 19.75 ppm that were corresponding to carbonyl, methine, methylene, and methyl resonance. Mass spectroscopy exhibited molecular weight for methyl 3- hydroxybutyric acid.ConclusionPHB-producing strain was identified as Bacillus cereus SH-02 (OM992297). Under optimum conditions from RSM analysis, the maximum PHB content and concentration of this strain can reach (3100.799 mg/L and 28.799%); respectively. FTIR, NMR, and Mass spectrometry were used to confirm the polymer as PHB. Our results demonstrated that optimization using RSM is one of the strategies used for reducing the production cost. RSM can determine the optimal factors to produce the polymer in a better way and in a larger quantity without consuming time.

Project description:Talaromyces albobiverticillius 30548 is a marine-derived pigment producing filamentous fungus, isolated from the La Réunion island, in the Indian Ocean. The objective of this study was to examine and optimize the submerged fermentation (SmF) process parameters such as initial pH (4-9), temperature (21-27 °C), agitation speed (100-200 rpm), and fermentation time (0-336 h), for maximum production of pigments (orange and red) and biomass, using the Box-Behnken Experimental Design and Response Surface Modeling (BBED and RSM). This methodology allowed consideration of multifactorial interactions between a set of parameters. Experiments were carried out based on the BBED using 250 mL shake flasks, with a 100 mL working volume of potato dextrose broth (PDB). From the experimental data, mathematical models were developed to predict the pigments and biomass yields. The individual and interactive effects of the process variables on the responses were also investigated (RSM). The optimal conditions for maximum production of pigments and biomass were derived by the numerical optimization method, as follows-initial pH of 6.4, temperature of 24 °C, agitation speed of 164 rpm, and fermentation time of 149 h, respectively.

Project description:In the present investigation, a newly isolated organic solvent-tolerant and alkaliphilic bacterial strain was reported from a hydrocarbon (gasoline and diesel) contaminated soil collected from the petrol station, Shirpur (India). The strain was identified as Bacillus circulans MTCC 7942, based on phenotype, biochemical, and phylogenetic analysis of 16S rRNA gene sequence. The capability of Bacillus circulans to secrete an extracellular, thermostable, alkaline protease and grow in the presence of organic solvents was explored. Bacillus circulans produced maximum alkaline protease (412?U/mL) in optimized medium (g/L): soybean meal, 15; starch, 10; KH2PO4, 1; MgSO4·7H2O, 0.05; CaCl2, 1; Na2CO3, 8; pH 10.0 at 37°C and 100?rpm. The competence of strain to grow in various organic solvents-n-octane, dodecane, n-decane, N,N-dimethylformamide, n-hexane, and dimethyl sulfoxide, establishes its potential as solvent-stable protease source for the possible applications in nonaqueous reactions and fine chemical synthesis.

Project description:The present work established the optimization and production of biodegradable thermoplastic polyhydroxybutyrate (PHB) from Amazonian microalga Stigeoclonium sp. B23. The optimization was performed in eight different growth media conditions of Stigeoclonium sp. B23, supplemented with sodium acetate and sodium bicarbonate and total deprivation of sodium nitrate. B23 was stained with Nile Red, and PHB was extracted and quantified by correlating the amount of fluorescence and biopolymer concentration through spectrofluorimetry and spectrophotometry, respectively. Our results detected the production of PHB in Stigeoclonium sp. B23 and in all modified media. Treatment with increased acetate and bicarbonate and without nitrate gave the highest concentration of PHB, while the treatment with only acetate gave the lowest among supplemented media. Our results showed a great potential of Stigeoclonium sp. B23, the first Amazonian microalga reported on PHB production. The microalga was isolated from a poorly explored and investigated region and proved to be productive when compared to other cyanobacterial and bacterial species. Additionally, microalga biomass changes due to the nutritional conditions and, reversely, biopolymer is well-synthetized. This great potential could lead to the pursuit of new Amazonian microalgae species in the search for alternative polyesters.

Project description:Microwave pyrolysis offers rapid and low-cost technology to upgrade agro-forestry residues to high-value products. I-optimal experimental design was used to determine the optimal combination of microwave power and exposure time to maximize biochar yield from corn cob. A validation experiment at optimal conditions of 600 W and 6.9 min produced an average yield of 56.98% on a dry and ash-free basis, agrees with the predicted value (3.43% error) and confirms the adequacy of the model yield equation. Characterization of biochar product revealed an organized mesoporous structure with a carbon content of 62.68%, surface area of 3.05 m2/g, pore volume of 0.003 cm3/g, capacitance range of 27.14-53.99 μF/g, energy density range of 6.0 × 10-7 - 1.2 × 10-6 Wh/kg, and power density range of 9.4 × 10-4 - 2.49 × 10-3 W/kg. The biochar produced would require further process to be considered for various industrial applications.

Project description:The study optimized the hydrolysis conditions for the production of fish collagen peptides from skin of Malabar grouper (Epinephelus malabaricus) using response surface methodology. The hydrolysis was done with enzymes pepsin, papain and protease from bovine pancreas. Effects of process parameters viz: pH, temperature, enzyme substrate ratio and hydrolysis time of the three different enzymes on degree of hydrolysis were investigated. The optimum response of degree of hydrolysis was estimated to be 10, 20 and 28% respectively for pepsin, papain and protease. The functional properties of the product developed were analysed which showed changes in the properties from proteins to peptides. SDS-PAGE combined with MALDI TOF method was successfully applied to determine the molecular weight distribution of the hydrolysate. The electrophoretic pattern indicated that the molecular weights of peptides formed due to hydrolysis were nearly 2 kDa. MALDI TOF spectral analysis showed the developed hydrolysate contains peptides having molecular weight in the range below 2 kDa.

Project description:The identification of novel bacterial strains with a high production potential of polyhydroxybutyrate (PHB) to substitute the bioplastics with non-biodegradable plastics and reducing environmental pollution is really effective. The present study was done with the purpose of PHB bioplastic production using a novel bacterial strain. Twenty-one different bacterial isolates were obtained from petrochemical wastewater, which among them, 10 isolates were PHB positive. The presence of PHB granules was detected in the isolates using Sudan Black B staining. The most excellent PHB-accumulating bacterium with a maximum yield of PHB (61.53%) was selected and identified as Bacillus cereus saba.zh, based on morphological, biochemical, and molecular techniques. 16S rRNA nucleotide sequence of the bacterium was assigned accession number: MT975245 in the NCBI database. The phylogenetic tree data showed that the closest type strain to the Bacillus cereus saba.zh is the Bacillus cereus SDB4 (91%). The three genes (phaA, phaB, and phaC) responsible for the PHB biosynthesis were amplified using the specific oligonucleotide primers by PCR technique. The highest PHB yield was achieved when the culture medium was supplemented with 3% sugarcane molasses as a carbon source, ammonium sulfate as the nitrogen source, at pH 7, and temperature of 30 °C. The characterization of the extracted polymer by FTIR and 1H NMR spectroscopy proves the presence of methyl, methylene, methine, hydroxyl, and ester carbonyl groups and confirmed the structure of biopolymer as PHB. The novel strain Bacillus cereus saba.zh has good potential as an appropriate candidate for low-cost industrial production of bioplastic.

Project description:For many years, reduction of fuel consumption has been a major aim in terms of both costs and environmental concerns. One option is to reduce the weight of fuel consumers. For this purpose, the use of a lightweight material based on rigid foams is a relevant choice. This paper deals with a new high temperature epoxy expanded material as substitution of phenolic resin, classified as potentially mutagenic by European directive Reach. The optimization of thermoset foam depends on two major parameters, the reticulation process and the expansion of the foaming agent. Controlling these two phenomena can lead to a fully expanded and cured material. The rheological behavior of epoxy resin is studied and gel time is determined at various temperatures. The expansion of foaming agent is investigated by thermomechanical analysis. Results are correlated and compared with samples foamed in the same temperature conditions. The ideal foaming/gelation temperature is then determined. The second part of this research concerns the optimization of curing cycle of a high temperature trifunctional epoxy resin. A two-step curing cycle was defined by considering the influence of different curing schedules on the glass transition temperature of the material. The final foamed material has a glass transition temperature of 270 °C.

Project description:Straw mulching is an effective conservation tillage tool that utilizes waste resources and reduces environmental pollution. To determine the optimal levels of quality, placement and quantity of straw mulching, we performed a field experiment that used the Box-Behnken design combined with response surface methodology. The treatments designed for walnut saplings (Juglans regia) considered three independent variables: quality, placement, and quantity of straw mulching. Tree height of walnut saplings (THW) and net photosynthesis rate (NPR) were used as the response variables in a full, quadratic polynomial model. Analysis of variance (ANOVA) results showed that the selected models were significant (P < 0.05), expressing ideal relationships between the independent and dependent variables (R2 ≥ 0.9225). The optimum conditions for the THW and NPR responses were determined to be a straw mulching quality which mixed rice and rapeseed straws, a straw mulching placement which covered the entire soil surface of experimental plots, and a straw mulching quantity applied as 3 kg/m2 (i.e., the low level). This optimized scheme of straw mulching combinations offers an alternate choice for optimizing the growth and potential yield of walnut saplings, but practical field experiments should also be carried out to obtain more site-specific results.
