Silica sulfuric acid: a reusable solid catalyst for one pot synthesis of densely substituted pyrrole-fused isocoumarins under solvent-free conditions.

ABSTRACT: A convenient and efficient methodology for the synthesis of densely substituted pyrrole-fused isocoumarins, which employs solid-supported silica sulfuric acid (SSA) as catalyst, has been developed. When the mixture of ninhydrin adducts of acetylacetone/ethyl acetoacetate and primary amines was heated on the solid surface of SSA under solvent-free conditions, the pyrrole-fused isocoumarins were formed in good yields. This synthetic method has several advantages such as the employment of solvent-free reaction conditions without the use of any toxic reagents and metal catalysts, the ease of product isolation, the use of a recyclable catalyst, the low cost, the easy availability of the starting materials, and the excellent yields of products.

Direct Hydroxymethylation of Furaldehydes with Aqueous Formaldehyde over a Reusable Sulfuric Functionalized Resin Catalyst.

Project description:Transformation of C5 furaldehydes to the corresponding C6 furaldehydes over a heterogeneous catalyst is a versatile but demanding reaction process. This report describes our discovery that Amberlyst-15, a conventional sulfuric functionalized resin, affords a 43.1% yield for 5-hydroxymethyl-2-furaldehyde (HMF) production with 57.5% selectivity via direct hydroxymethylation of 2-furaldehyde (furfural) with an aqueous formaldehyde reagent. This report is the first to describe an important achievement on the heterogeneous acid catalyst-assisted HMF production from furfural via direct hydroxymethylation in a batch reactor system. The Amberlyst-15 was also reusable and applicable for C5 furaldehyde upgrading of various types: furfuryl alcohol, furfurylamine, and furoic acid toward the corresponding C6 furaldehydes. Results show that a liquid flow system becomes an advanced tool for the continuous production of upgrading furaldehydes through a fixed Amberlyst-15 catalyst bed. Accordingly, the sulfuric functionalized resin-catalyzed direct hydroxymethylation of furaldehydes with aqueous formaldehyde in both batch and flow reactor systems is worthwhile and opens up new avenues for biomass transformations in particles, an important area of inedible biomass utilization.

Immobilized Sulfuric Acid on Silica Gel as Highly Efficient and Heterogeneous Catalyst for the One-Pot Synthesis of Novel α-Acyloxycarboxamides in Aqueous Media.

Project description:The application of immobilized sulfuric acid on silica gel (H2SO4-SiO2) as an efficient and easily reusable solid catalyst was explored in the synthesis of novel α-acyloxycarboxamide derivatives via a Passerini reaction of benzoic acid, aldehyde/ketone, and isocyanides. The Passerini adducts were obtained in high to excellent yields within 10 min in aqueous media under catalytic conditions. The key advantages of the process include a short reaction time, high yields, the catalyst's low cost, and the catalyst's reusability.

Synthesisand Biological Evaluationof New Substituted Hantzsch Thiazole Derivatives from Environmentally Benign One-Pot Synthesis Using Silica Supported Tungstosilisic Acid as Reusable Catalyst.

Project description:An efficient and green method has been developed for the synthesis of new substituted Hantzsch thiazole derivatives in 79%-90% yield, via the one-pot multi-component procedure, by the reaction of 3-(bromoacetyl)-4-hydroxy-6-methyl-2H-pyran-2-one, thiourea and substituted benzaldehydes in the presence of silica supported tungstosilisic acid, as a reusable catalyst, under conventional heating or under ultrasonic irradiation. The catalyst is recoverable by a simple filtration and can be reused in the subsequent reactions. Most of the thiazoles exhibited significant antibacterial activity compared toamoxicillin and ciprofloxacin as positive controls. In addition, the new compounds showed moderate to good antioxidant (DPPH) radical scavenging activity.

Zr(HSO4)4: Green, efficient and reusable catalyst for one-pot synthesis of 1,8-dioxooctahydroxanthene under solvent-free conditions.

Project description:Many methods have been used to synthesize xanthene derivatives using different catalysts. However, some of these methodologies have not been entirely satisfactory. Most of the mentioned methods have disadvantages such as low yields, prolonged reaction times, harsh reaction conditions and the requirement of expensive catalysis and use of toxic organic solvent. In this research, a green and highly efficient procedure for the one-pot synthesis of 1,8-dioxo-octahydro-xanthenes has been developed. Zr(HSO4)4 catalyst was used as an efficient and recoverable catalyst for synthesis of 1,8-dioxo-octahydro-xanthene derivatives via cyclocondensation of dimedone and aromatic aldehydes in solvent-free conditions. There are no examples of the use of Zr(HSO4)4 for the synthesis of 1,8-dioxo-octahydro-xanthene derivatives. The present method offers several advantages such as green, highly efficient, recoverable, reusable, simple work-up and simple purification of products. The structure of the synthesized products was confirmed by Fourier Transform Infrared (Ft-IR) and Proton nuclear magnetic resonance (1HNMR) analyzes. The antibacterial activity of the synthesized compounds was determined by agar disk diffusion method against gram-positive (S. aureus bacteria) and gram-negative (E. coli bacteria) microorganisms. Among the synthesized compounds (3a-3j), 3h compound showed the highest antibacterial effect by forming an inhibitory diameter zone of 15 mm around the disc containing 2000 mg of 3h-compound against gram-positive (S. aureus bacteria). 1. Catalyst-Free One-Pot Synthesis of Densely Substituted Pyrazole-Pyrazines as Anti-Colorectal Cancer Agents.

Project description:The first catalyst-free post-Ugi cascade methodology was developed for expeditious access to structurally diverse and complex pyrazole-pyrazines in one-pot. This novel cascade reaction features an intramolecular N2-arylation of pyrazoles with allenes at the C-β position of triple bond. Screening in the colorectal cancer cell lines HCT116 and SW620 validated the feasibility of the methodology for generating bioactive compounds. The lead compound 7h which is active against HCT116 and SW620 with IC50 of 1.3 and 1.8 µM, respectively, can be synthesized and purified in a gram process synthetic scale in 7 hours. The mechanical studies indicated that compound 7h can induce cell cycle arrest in the G2/M phase and inhibit proliferation and viability in human colon cancer cells. Overall, compound 7h is represented as a promising starting point for the development of new anti-colorectal cancer drugs.

Silica: An efficient catalyst for one-pot regioselective synthesis of dithioethers.

Project description:The development of a silica-promoted highly selective synthesis of 1,2 or 1,3-dithioethers via solvent-free one-pot tandem reactions of an allyl bromide with excess thiol at room temperature is described. The choice of silica gel, either pre-calcined or moistened with water, exhibited notable regioselectivity in the formation of dithioethers. Plausible mechanistic routes were explored and postulated.

Polymer ionic liquid network: a highly effective reusable catalyst for one-pot synthesis of heterocyclic compounds.

Project description:Significant efforts have been devoted to developing immobilized chiral catalysts with high activity, selectivity, and stability. In this present study, a new heterogeneous proline catalyst system was prepared based on strong noncovalent interactions between polymer ionic liquid (PIL) and l-proline. First, pyridine PILs, which can complex with l-proline monomers through noncovalent interactions, were synthesized using reversible addition-fragmentation chain transfer (RAFT) polymerization. The polymer network-supported chiral catalysts were obtained following further free radical polymerization. Different structures were formed in response to different ratios of PIL and chiral monomer, as well as different PIL anions, in the reactions. The new formed layer structures and synergic effects of PIL resulted in heterogeneous catalysts with high catalytic activity and enantioselectivity, thus endowing them with better catalytic performance for the one-pot synthesis of heterocyclic compounds compared to homogeneous catalytic systems. These catalytic systems were able to be reused and recycled five times with no discernible loss in catalytic activity and enantioselectivity. l-Proline was efficiently loaded onto the polymer network simply based on supramolecular interactions, providing a novel method of synthesizing high performance supported catalysts for organic reactions.

One-pot multicomponent nitro-Mannich reaction using a heterogeneous catalyst under solvent-free conditions.

Project description:An environmentally-friendly, one-pot multicomponent reaction of various aldehydes, amines and nitroalkanes for the synthesis of ?-nitroamines is here described. Amberlyst A-21 supported CuI was found to be a highly efficient novel heterogeneous catalyst for the three-component nitro-Mannich reaction between aldehydes, amines and nitroalkanes. The developed protocol is performed in a solvent-free medium to produce a variety of ?-nitroamines in good to excellent yields within short reaction times. The catalyst can be easily prepared and recovered. It has been tested up to eight times with only a minor activity loss.

Pyrrole and Fused Pyrrole Compounds with Bioactivity against Inflammatory Mediators.

Project description:A new series of pyrrolopyridines and pyrrolopyridopyrimidines have been synthesized from aminocyanopyrroles. The synthesized compounds have been characterized by FTIR, ¹H-NMR and mass spectroscopy. The final compounds have been screened for in vitro pro-inflammatory cytokine inhibitory and in vivo anti-inflammatory activity. The biological results revealed that among all tested compounds some fused pyrroles, namely the pyrrolopyridines 3i and 3l, show promising activity. A docking study of the active synthesized molecules confirmed the biological results and revealed a new binding pose in the COX-2 binding site.

Project description:In the present research article, we have developed solid heterogenous silica supported lanthanum trifluoroacetate and trichloroacetate as green Lewis acid catalysts. These catalysts were synthesized by a novel, simple, cheap, clean, and environment friendly method. The physicochemical properties of the prepared catalysts were well studied and characterized by sophisticated spectroscopic techniques such as FTIR, TGA, XRD, EDX, SEM, TEM and BET analysis. The catalyst was utilized in the synthesis of arylimidazole derivatives via green protocols under solvent-free conditions at 70 °C with a higher yield, mild reaction conditions and a short reaction time. The catalyst works superiorly in water as well as in various organic solvents as a reusable and easily recoverable catalyst.
