A Solvent-free Approach to Glycosyl Amides: Towards the Synthesis of ?-N-Galactosyl Ceramides.

ABSTRACT: A new, simple and efficient method for the synthesis of both ?- and ?-glycosyl amides using solvent-free conditions is described. This method involves the coupling of glycosyl amines with the p-nitrophenol esters of lipids as a key step.

New CD1d agonists: synthesis and biological activity of 6″-triazole-substituted α-galactosyl ceramides.

Project description:Huisgen [3+2] dipolar cycloaddition of 6″-azido-6″-deoxy-α-galactosyl ceramide 11 with a range of alkynes (or a benzyne precursor) yielded a series of triazole-containing α-galactosyl ceramide (α-GalCer) analogues in high yield. These α-GalCer analogues and the precursor azide 11 were tested for their ability to activate iNKT cells and stimulate IL-2 cytokine secretion in vitro, and IFN-γ and IL-4 cytokine secretion in vivo. Some of these analogues, specifically 11, 12b, 12f and 13, were more potent IL-2 stimulators than the prototypical CD1d agonist, α-GalCer 1. In terms of any cytokine bias, most of the triazole-containing analogues exhibited a small Th2 cytokine-biasing response relative to that shown by α-GalCer 1. In contrast, the cycloaddition precursor, namely azide 11, provided a small Th1 cytokine-biasing response.

Project description:This study described an efficient and practical approach for amide synthesis. The reaction was conducted under metal- and solvent-free conditions at a mild temperature (40?°C) in air, and readily available formamides were used as an amino source. This reaction can be easily upgraded to the gram level with an excellent yield.

Project description:A library of dimeric CD1d ligands, containing two ?-galactosyl ceramide (?-GalCer) units linked by spacers of varying lengths has been synthesised. The key dimerisation reactions were carried out via copper-catalysed click reactions between a 6"-azido-6"-deoxy-?-galactosyl ceramide derivative and various diynes. Each ?-GalCer dimer was tested for its ability to stimulate iNKT cells.

Project description:Chlorination with equimolar POCl? can be efficiently achieved not only for hydroxypyrimidines, but also for many other substrates such as 2-hydroxy-pyridines, -quinoxalines, or even -amides. The procedure is solvent-free and involves heating in a sealed reactor at high temperatures using one equivalent of pyridine as base. It is suitable for large scale (multigram) batch preparations.

Solvent-Free Synthesis and Safener Activity of Sulfonylurea Benzothiazolines.

Project description:A series of novel sulfonylurea benzothiazolines was designed by splicing active groups and bioisosterism. A solvent-free synthetic route was developed for the sulfonylurea benzothiazoline derivatives via the cyclization and carbamylation. All compounds were characterized by IR, ¹H-NMR, 13C-NMR, HRMS. The biological activity tests indicated the compounds could protect maize against the injury caused by chlorsulfuron to some extent. The molecular docking result showed that the new compound competed with chlorsulfuron to bind with the herbicide target enzyme active site to attain detoxification.

Multicomponent approach to the synthesis of oxidized amides through nitrile hydrozirconation.

Project description:"Oxidized" amides, as represented by acyl aminals and acyl hemiaminals, are integral subunits of several natural products that exhibit useful biological activity. In this paper a multicomponent approach to these groups from acylimine intermediates is demonstrated. The acylimines are accessed through a sequence of nitrile hydrozirconation and acylation, making this highly versatile amide synthesis useful for a range of applications in target- and diversity-oriented synthesis.

Synthesis of a versatile building block for the preparation of 6-N-derivatized ?-galactosyl ceramides: rapid access to biologically active glycolipids.

Project description:A concise route to the 6-azido-6-deoxy-?-galactosyl-phytosphingosine derivative 9 is reported. Orthogonal protection of the two amino groups allows elaboration of 9 into a range of 6-N-derivatized ?-galactosyl ceramides by late-stage introduction of the acyl chain of the ceramide and the 6-N-group in the sugar headgroup. Biologically active glycolipids 6 and 8 have been synthesized to illustrate the applicability of the approach.

Development of solvent-free synthesis of hydrogen-bonded supramolecular polyurethanes.

Project description:In this work we describe the solvent free synthesis of supramolecular polyurethanes held together by heterocomplementary triple hydrogen bonding. We perform a systematic evaluation on the base catalyzed synthesis of small molecule ureas and carbamates from a range of isocyanates, amines and alcohols in solution and in the solid state using ball milling. These optimized procedures are then shown to be applicable to the synthesis of supramolecular polyurethanes using solvent-free methods.

New Methylamine-Iodine-Mediated Solvent-Free Approach of Hybrid Perovskite Synthesis via the Redox Conversion of Metallic Lead Films.

Project description:The fast progress of lab-scale perovskite solar cells makes the problem of upscaling of perovskite thin-film deposition more and more acute; therefore, the development of new methods for perovskite deposition is highly desired. In this work, we proposed a new solution-free preparation approach for hybrid perovskite films based on the in situ generation of methylammonium iodide from methylamine and iodine vapors in the presence of an organic reducing agent conjugated with a redox process of metallic lead conversion with iodine vapor. At first, we demonstrated that either metallic lead or lead iodide powders can readily react with the solution of methylamine and iodine in the presence of isopropyl alcohol acting as a reducing agent, resulting in a phase-pure polycrystalline CH3NH3PbI3 perovskite. A possible mechanism of underlying chemical processes has been proposed. In order to convert the predeposited thin films of metallic lead to CH3NH3PbI3 perovskite, a protocol of sequential treatment by iodine and methylamine vapors was proposed. Finally, we revealed the optimal conditions of processing, which were proven to be facile and robust for the sake of the better control of perovskite grain morphology.

Project description:Synthesis of ?-C-D-galactosyl D- and L-alanines is carried out via a highly stereoselective Grignard reaction of glycosyl iodides, Sharpless dihydroxylation and S(N)2 displacement of the corresponding mesylate or tosylate. Alternatively, attempted triflation of the intermediate alcohols triggers a stereoselective debenzylative cyclization leading to interesting bicyclic trans-fused compounds.
