Chemically Modified Plastic Tube for High Volume Removal and Collection of Circulating Tumor Cells.

ABSTRACT: In this preliminary effort, we use a commercially available and chemically modified tube to selectively capture circulating tumor cells (CTCs) from the blood stream by immobilizing human anti-EpCAM antibodies on the tube's interior surface. We describe the requisite and critical steps required to modify a tube into a cancer cell-capturing device. Using these simple modifications, we were able to capture or entrap about 85% of cancer cells from suspension and 44% of cancer cells from spiked whole blood. We also found that the percentage of cells captured was dependent on the tube's length and also the number of cancer cells present. It is our strong belief that with the utilization of appropriate tube lengths and procedures, we can ensure capture and removal of nearly the entire CTC population in whole blood. Importantly after a patient's entire blood volume has circulated through the tube, the tube can then be trypsinized to release the captured live CTCs for further analysis and testing.

Chemically Modified Polyvinyl Chloride for Removal of Thionine Dye (Lauth's Violet).

Project description:The chemical modification of hydrophobic polymer matrices is an alternative way to elchange their surface properties. The introduction of sulfonic groups in the polymer changes the surface properties such as adhesion, wettability, catalytic ability, and adsorption capacity. This work describes the production and application of chemically modified polyvinyl chloride (PVC) as adsorbent for dyes removal. Chemical modification of PVC was evaluated by infrared spectroscopy and elemental analysis, which indicated the presence of sulfonic groups on PVC. The chemically modified PVC (PVCDS) showed an ion exchange capacity of 1.03 mmol-1, and efficiently removed the thionine dye (Lauth's violet) from aqueous solutions, reaching equilibrium in 30 min. The adsorption kinetics was better adjusted for a pseudo second order model. This result indicates that the adsorption of thionine onto PVCDS occurs by chemisorption. Among the models for the state of equilibrium, SIPS and Langmuir exhibited the best fit to the experimental results and PVCDS showed high adsorption capacities (370 mg-1). Thus, it is assumed that the system presents homogeneous characteristics to the distribution of active sites. The modification promoted the formation of surface characteristics favorable to the dye adsorption by the polymer.

Chemically Dual-Modified Biochar for the Effective Removal of Cr(VI) in Solution.

Project description:Here, a dual-modification strategy using KMnO4 (potassium permanganate) and AlCl3·6H2O (aluminum chloride, hexahydrate) as co-modifiers to improve the Cr(VI) removal capacity of K2CO3 activated biochar is introduced. As a result, the dual-modified biochar with KMnO4 and AlCl3·6H2O has the calculated adsorption energy of -0.52 eV and -1.64 eV for HCrO4-, and -0.21 eV and -2.01 eV for Cr2O72-. The Al2O3 (aluminum oxide) and MnO (manganese oxide) embedded on the surface of dual-modified biochar bring more Cr(VI) absorption sites comparing to single-modified biochar, resulting in a maximum Cr(VI) saturated adsorption capacity of 152.86 mg g-1. The excellent removal performance is due to the synthetic effect of electrostatic attraction, reduction reaction, complexation reaction, and physical adsorption. The experimental results also indicated that the spontaneous adsorption process agreed well with the pseudo-second order and Langmuir models. This dual-modification strategy is not limited to the treatment of Cr(VI) with biochar, and may also be incorporated with the treatment of other heavy metals in aqueous environment.

High-performance liquid chromatography purification of chemically modified RNA aptamers.

Project description:2'-Fluoro modified RNAs are useful as potential therapeutics and as special substrates for studying RNA function. 2'-Fluoro modified RNAs generally need to be purified after they are prepared either enzymatically or by solid-phase synthesis. Here we introduce a protocol by which 2'-fluoro modified RNAs with 57 and 58 nucleotides can be resolved and purified using ion-pair, reverse-phase high-performance liquid chromatography (HPLC). Because the size of our RNA samples is in the range of many known RNA aptamers of therapeutic values, our protocol should be generally useful.

What is the allowed volume threshold for chest tube removal after lobectomy: A randomized controlled trial.

Project description:IntroductionThe management of chest tubes and the volume threshold for chest tube removal after pulmonary resection remain controversial. Several studies have reported the volume threshold for chest tube removal following pulmonary resection to range from 200 to 450 mL/24 h.MethodsA prospective randomized single-blind clinical study was performed with data collected from patients who had undergone lobectomy and lymph node dissection at our hospital between June 2014 and April 2018. The patients were randomly assigned to the High group (removal of chest tube when drainage was <450 mL/24 h) or Low group (removal of chest tube when drainage was <200 mL/24 h) at postoperative day (POD) 2. The primary end point was drainage time. The secondary end point were complications and rate of thoracentesis.ResultsSeventy patients met the inclusion criteria and were randomized, with 35 patients assigned to the High group and 35 patients to the Low group. The average duration of chest tube placement was 2.05 days in the High group and 2.31 days in the Low group. The duration of chest tube placement in the High group was significantly shorter than that in the Low group (p = 0.02). There were no major postoperative complications in either group. Thoracentesis was not necessary in either group.ConclusionPleural effusion of 450 mL/day is tolerable as the volume threshold for the removal of a chest tube after pulmonary resection.

Destruction of mesoscopic chemically modulated domains in single phase high entropy alloy via plastic deformation.

Project description:Chemically modulated mesoscopic domains in a fcc single phase CrMnFeCoNi equi-atomic high entropy alloy (HEA) are detected by small angle diffraction performed at a synchrotron radiation facility, whereas the mesoscopic domains cannot be detected by conventional X-ray diffraction and 2D mappings of energy dispersive X-ray spectroscopy by scanning electron microscopy and scanning transmission electron microscopy. The mesoscopic domains are deformed and shrieked, and finally destructed by plastic deformation, which is supported by the comprehensive observations/measurements, such as electrical resistivity, Vickers hardness, electron backscattering diffraction, and hard X-ray photoemission spectroscopy. The destruction of the mesoscopic domains causes the decrease in electrical resistivity via plastic deformation, so called K-effect, which is completely opposite to the normal trend of metals. We confirmed that the presence and the size of local chemical ordering or short-range order domains in the single phased HEA, and furthermore, Cr and Mn are related to form the domains.

Chemically and biologically synthesized CPP-modified gelonin for enhanced anti-tumor activity.

Project description:The ineffectiveness of small molecule drugs against cancer has generated significant interest in more potent macromolecular agents. Gelonin, a plant-derived toxin that inhibits protein translation, has attracted much attention in this regard. Due to its inability to internalize into cells, however, gelonin exerts only limited tumoricidal effect. To overcome this cell membrane barrier, we modified gelonin, via both chemical conjugation and genetic recombination methods, with low molecular weight protamine (LMWP), a cell-penetrating peptide (CPP) which was shown to efficiently ferry various cargoes into cells. Results confirmed that gelonin-LMWP chemical conjugate (cG-L) and recombinant gelonin-LMWP chimera (rG-L) possessed N-glycosidase activity equivalent to that of unmodified recombinant gelonin (rGel); however, unlike rGel, both gelonin-LMWPs were able to internalize into cells. Cytotoxicity studies further demonstrated that cG-L and rG-L exhibited significantly improved tumoricidal effects, with IC50 values being 120-fold lower than that of rGel. Moreover, when tested against a CT26 s.c. xenograft tumor mouse model, significant inhibition of tumor growth was observed with rG-L doses as low as 2 μg/tumor, while no detectable therapeutic effects were seen with rGel at 10-fold higher doses. Overall, this study demonstrated the potential of utilizing CPP-modified gelonin as a highly potent anticancer drug to overcome limitations of current chemotherapeutic agents.

Cd(ii) removal by Fe(ii) surface chemically modified layered double hydroxide-graphene oxide: performance and mechanism.

Project description:Cd(ii) adsorption onto Fe(ii) modified Layered double hydroxide-graphene oxide (LDH-GO@Fe(ii)) was investigated using batch experiments. With the modification of Fe(ii), LDH-GO maintained its structure, while Fe(ii) species formed non-crystalline iron oxide clusters on the surface of the LDH/GO. A kinetics study indicated that adsorption obeyed a pseudo-second-order rate law. The equilibrium data were fitted well with the Langmuir isotherm model. The maximum adsorption capacity of LDH-GO@Fe(ii)10 was 28.98 mg g-1, higher those that of pure LDH-GO and LDH-GO@Fe(ii)50. The increased sorption capacities could be explained by the increased specific surface area. Modification with Fe(ii) would lead to the generation of amorphous Fe oxides and Fe could occupy the binding sites for Cd(ii), thus excess Fe in the structure will restrain the adsorption of Cd(ii). The XRD and XPS patterns revealed the formation of Cd(OH)2 after adsorption. Batch experiments indicated that precipitation and surface complexation were the main pathways for Cd(ii) removal.

Endonasal Skull Base Tumor Removal Using Concentric Tube Continuum Robots: A Phantom Study.

Project description:Objectives The purpose of this study is to experimentally evaluate the use of concentric tube continuum robots in endonasal skull base tumor removal. This new type of surgical robot offers many advantages over existing straight and rigid surgical tools including added dexterity, the ability to scale movements, and the ability to rotate the end effector while leaving the robot fixed in space. In this study, a concentric tube continuum robot was used to remove simulated pituitary tumors from a skull phantom. Design The robot was teleoperated by experienced skull base surgeons to remove a phantom pituitary tumor within a skull. Percentage resection was measured by weight. Resection duration was timed. Setting Academic research laboratory. Main Outcome Measures Percentage removal of tumor material and procedure duration. Results Average removal percentage of 79.8 ± 5.9% and average time to complete procedure of 12.5 ± 4.1 minutes (n = 20). Conclusions The robotic system presented here for use in endonasal skull base surgery shows promise in improving the dexterity, tool motion, and end effector capabilities currently available with straight and rigid tools while remaining an effective tool for resecting the tumor.

Preparation, Characterization, and Chemically Modified Date Palm Fiber Waste Biomass for Enhanced Phenol Removal from an Aqueous Environment.

Project description:The date palm tree is extensively cultivated in Middle Eastern countries such as Saudi Arabia, generating a large amount of waste in the form of leaves, seeds, and fibrous materials. This study examined the feasibility of using raw date palm fiber (RDPF) and NaOH chemically modified date palm fiber (NaOH-CMDPF) obtained from discarded agricultural waste for the removal of phenol in an aqueous environment. The adsorbent characterization was performed by using different techniques, i.e., particle size analysis; elemental analyzer (CHN); and BET, FTIR, and FESEM-EDX analysis. The FTIR analysis revealed the presence of various functional groups on the surface of the RDPF and NaOH-CMDPF. The results showed that chemical modification by NaOH increased the phenol adsorption capacity that was well-fitted by the Langmuir isotherm. Higher removal was obtained with NaOH-CMDPF (86%) than with the RDPF (81%). The RDPF and NaOH-CMDPF sorbents' maximum (Qm) adsorption capacities were more than 45.62 mg/g and 89.67 mg/g and were comparable to the sorption capacities of various other types of agricultural waste biomass reported in the literature. The kinetic studies confirmed that the adsorption of phenol followed the pseudo-second-order kinetic process. The present study concluded that the RDPF and NaOH-CMDPF were eco-friendly and cost-effective in promoting sustainable management and the reuse of the Kingdom's lignocellulosic fiber waste material.

Preanalytics of urine sediment examination: effect of relative centrifugal force, tube type, volume of sample and supernatant removal.

Project description:Introduction:Laboratories often modify procedures recommended by the European Urinalysis Guidelines for urine sediment analysis. The aim of this study was to compare the recommended protocol with our routine laboratory procedure and to evaluate the possible impact of modifications in the relative centrifugal force, type of tube, method of supernatant aspiration and urine volume on patient's results. Material and methods:Firstly, relative centrifugal force was investigated using 20 pairs of samples examined after centrifugation at 400xg and 1358xg. In phase two, 110 samples were examined, paired as: round bottom vs conical tube (N = 46), decanting vs suction of supernatant (N = 100) and 10 mL vs 5 mL of urine sample (N = 101). Results:The number of erythrocytes, leukocytes and squamous epithelial cells was significantly lower after centrifugation at 400xg (P = 0.001, 0.002 and 0.004, respectively). The number of leukocytes was significantly lower in conical tubes (P = 0.010), after the suction of supernatant (P = 0.045) and in 5 mL urine (P < 0.001). The number of squamous epithelial cells was significantly lower after the suction of supernatant (P < 0.001) and in 5 mL urine (P < 0.001). The number of erythrocytes (P < 0.001), total non-hyaline casts (P < 0.001) and the frequency of granular casts (P = 0.039) was significantly lower in 5 mL urine. Conclusion:Lower results of leukocytes, erythrocytes, squamous cells and non-hyaline casts were recorded in recommended procedures (centrifugation at 400xg, suction of supernatant, conical tube, 5 mL of sample) than in routine procedure (centrifugation at 1358xg, decanting of supernatant, round bottom tube, 10 mL) used in our laboratory.
