Genicunolide A, B and C: three new triterpenoids from Euphorbia geniculata.

ABSTRACT: Three new triterpenoids, designated as genicunolide A (1), B (2) and C (3), along with friedelin (4) and friedelinol (5), were isolated from the aerial parts of Euphorbia geniculata. They were characterized as 1?-acetoxy-3?-hydroxy-11?,12?-oxidotaraxer-14-ene, 1?,3?-diacetoxy-21?-hydroxy-11?,12?-oxidotaraxer-14-ene and 3?,9?,20?-trihydroxy-?-taraxast-5-ene, respectively, by spectral and chemical methods.

Project description:Three new triterpenoids, 3β-hydroxy-urs-30-p-Z-hydroxycinnamoyl-12-en-28-oic-acid (1), 3β-hydroxy-olean-30-p-Ehydroxycinnamoyl-12-en-28-oic-acid (2) and 3β,6α-dihydroxy-urs-14-en-12-one (3), together with seven known triterpenoids, were isolated from the roots of Rubia schumanniana. Their structures were established by means of spectroscopic analysis. All compounds were evaluated for cytotoxic activity, and compounds 2–6 showed cytotoxicity with the IC50 values of 10.75∼18.87 µg/mL. Triterpenoids from Euphorbia maculata and Their Anti-Inflammatory Effects.

Project description:Euphorbia maculata is a medicinal plant of the Euphorbiaceae family, which can produce anti-inflammatory and cancer chemopreventive agents of triterpenoids. The present study reports on the bioactive triterpenoids of this plant. Two new lanostane-type triterpenoids, named (3S,4S,7S,9R)-4-methyl-3,7-dihydroxy-7(8?9) abeo-lanost-24(28)-en-8-one (1) and 24-hydroperoxylanost-7,25-dien-3?-ol (2), together with 15 known triterpene derivatives, were isolated from Euphorbia maculata. The structures of the new compounds were determined on the basis of extensive spectroscopic data (UV, MS, ¹H and 13C-NMR, and 2D NMR) analysis. All tetracyclic triterpenoids (1?11) were evaluated for their anti-inflammatory effects in the test of TPA-induced inflammation (1 ?g/ear) in mice. The triterpenes exhibited significant anti-inflammatory activities.

Inhibition of 11b-HSD1 by tetracyclic triterpenoids from Euphorbia kansui.

Project description:The roots of Euphorbia kansui are considered an important traditional folk medicine. In this study the ethanol extracts of E. kansui were investigated. A new tetracyclic triterpenoid, euphane-3b,20-dihydroxy-24-ene, in addition to five known triterpenoids with euphane skeletons were isolated. Their structures were elucidated on the basis of physical and spectral techniques (1D-, 2D-NMR and MS, respectively). Furthermore, these compounds 1-6 exhibited strong inhibitory activity against human 11b-hydroxysteroid dehydrogenase type 1 (11b-HSD1), with IC(50) values of 34.86 nM, 1.115 mM, 16.08 nM, 2.815 nM, 26.47 nM, 15.99 nM, and 41.86 nM, respectively. The docking results show that the ring part of compounds can insert into the hydrophobic core of h11b-HSD1 and the alkane chain orientates toward the outside. The results presented herein provide a scientific explanation for the usage of the E. kansui in clinical treatment of diabetes.

Project description:Three new myrsinol diterpenes were isolated from the roots of Euphorbia prolifera. Their structures were elucidated as 2?-O-isobutyryl-3?,5?,7?,10,15?-penta-O-acetyl-14?-O-benzoyl-10,18-dihydromyrsinol (1), 2?-O-isobutyryl-3?-O-propion-yl-5?,7?,10,15?-tetra-O-acetyl-10,18-dihydromyrsinol (2), and 2?,14?-di-O-benzoyl-3?,5?,7?,10,15?-penta-O-acetyl-10,18-dihydromyrsinol (3) on the basis of spectroscopic data analyses (IR, ESI-MS, HR-ESI-MS, and 1D and 2D NMR). Their neuroprotective activities were evaluated and compounds 1 and 2 showed neuroprotective effects against MPP+ -induced neuronal cell death in SH-SY5Y cells.

Rhodoterpenoids A?C, Three New Rearranged Triterpenoids from Rhododendron latoucheae by HPLC?MS?SPE?NMR.

Project description:Rhodoterpenoids A?C (1?3), three new rearranged triterpenoids, together with one new biogenetically related compound, rhodoterpenoid D (4), were isolated and efficiently elucidated from Rhododendron latoucheae by high-performance liquid chromatography-mass spectrometry-solid-phase extraction-nuclear magnetic resonance (HPLC?MS?SPE?NMR). Compounds 1 and 2 possess an unprecedented skeleton with a 5/7/6/6/6-fused pentacyclic ring system, while compound 3 contains a unique 6/7/6/6/6-fused pentacyclic carbon backbone. Their structures were determined by extensive spectroscopic methods and electronic circular dichroism (ECD) analyses. Plausible biogenetic pathways for 1?4 were proposed. Compounds 1 and 4 showed potential activity against herpes simplex virus 1 (HSV-1) with IC50 values of 8.62 and 6.87??M, respectively.

Exploring the Chemical Diversity of Algerian Plants: Three New Pentacyclic Triterpenoids from Launaea acanthoclada Roots.

Project description:The chemical study of Launaea acanthoclada from South-East Algeria led to the isolation of twelve oxygenated terpenoid compounds, including three new pentacyclic triterpenoids 1-3 with either lupane or ursane rearranged skeletons. The structure and the stereochemistry of these compounds were established by spectroscopic methods, including NMR techniques. The chemical pattern of L. acanthoclada is in accordance with the triterpenoid scenario of the genus Launaea embracing to date lupane, oleane, ursane and taraxastane skeletons. However, the carbon frameworks exhibited by new compounds 1-3 have never been reported from Launaea species.

Their structures were elucidated by the combination of mass spectrometry (MS), one and two-dimensional NMR experiments.

Isolation, Structural Elucidation of Three New Triterpenoids from the Stems and Leaves of Schisandra chinensis (Turcz) Baill.

Project description:Schisandra chinensis (Turcz) Baill. is sufficiently well known as a medicinal plant worldwide, which modern research shows has many pharmacological activities such as hepatoprotective, anti-inflammatory effect, potent anti-HIV-1 activity, anti-tumor effect, and activity on the central nervous system. With considerable chemical investigation, three new triterpenoids (1⁻3), together with four known triterpenoids were isolated from the S. chinensis (Turcz) Baill. Their structures were elucidated by 1D- and 2D-NMR spectroscopic analyses, single-crystal X-ray diffraction and high-resolution mass spectroscopy, which were identified as Schisanlactone I (1), Schinalactone D, (2), Schisanlactone J, (3) Kadsuphilactone B (4), Schisanlactone C (5), Schisphendilactone B (6), and Schinchinenlactone A (7). The cytotoxicity of those compounds (1⁻7) was tested against Hep-G2 cell lines, but no apparent antitumor activity was observed at 50 µg/mL using MTT method.

Euphosantianane E-G: Three New Premyrsinane Type Diterpenoids from Euphorbia sanctae-catharinae with Contribution to Chemotaxonomy.

Project description:Euphorbia species were widely used in traditional medicines for the treatment of several diseases. From the aerial parts of Egyptian endemic plant, Euphorbia sanctae-catharinae, three new premyrsinane diterpenoids, namely, euphosantianane E-G (1-3), alongside four known triterpenes, 9,19-cyclolanostane-3β,24S-diol (4), 25-methoxycycloartane-3β,24S-diol (5), 25-methylenecycloartan-3β,24R-diol (6), and 25-methylenecycloartan-3β,24S-diol (7), were isolated and identified. The chemical structures were proven depending upon spectroscopic analysis, including FTIR, HRFABMS, and 1D/2D-NMR. The chemotaxonomic significance of the isolated compounds, especially diterpenes from E. sanctae-catharinae compared to those documented from different Euphorbia species was also studied via agglomerative hierarchical clustering (AHC). The Egyptian endemic Euphorbia sanctae-catharina was grouped with E. bupleuroides, E. fidjiana, E. fischeriana, E. pithyusa subsp. cupanii, E. prolifera, and E. seguieriana, where myrsinol diterpenoids were the characteristic compounds.
