Cucurbit[n]uril (n = 6, 7) Based Carbon-Gold Hybrids with Peroxidase-Like Activity.

ABSTRACT: Despite the combination of molecular recognition and local electric field enhancement endowing cucurbit[n]uril-capped metallic nanoparticles, indicating great potential in a variety of areas, prior work has paid little attention to carbonizing cucurbit[n]uril on the surface of gold nanoparticles, which may propose new carbon-gold hybrid materials with interesting applications. In this work, we developed a simple and cost-effective method to prepare carbon-gold hybrids by carbonizing cucurbit[n]uril modified gold nanoparticles. The as-prepared cucurbit[n]uril based carbon and carbon-gold hybrid materials have shown to possess peroxidase-like activity. All cucurbit[n]uril based nanomaterials exhibited high catalytic activity over a pH range 2?6 and more tolerant to high temperature (up to 60 °C) when compared to natural horseradish peroxidase.

Cucurbit[7]uril-based high-performance catalytic microreactors.

Project description:Catalytic microreactors manufactured using microfluidic devices have received significant research interest in recent years. However, little attention has been paid to immobilising metallic nanoparticles (NPs) onto microchannel walls for high efficiency catalytic reactions. We demonstrate a facile preparation of cucurbit[7]uril-based catalytic microreactors, where metallic NPs are immobilised onto microchannels via supramolecular complexation with methyl viologen@cucurbit[7]uril (CB[7]). These microreactors exhibit a remarkable catalytic activity owing to the substantially high surface area to volume ratio of the microchannels and metallic NPs. Superior to most conventional heterogeneous catalytic reactions, separation post reaction and complicated recycling steps of the catalysts are not required. Moreover, CB[7] can complex a variety of metallic NPs to its portal, providing a multifunctional high-performance in situ catalytic platform.

Cucurbit[10]uril.

Project description:Melamine diamine 1 is able to displace CB[5] from the CB[10].CB[5] complex resulting in CB[10].12 and precipitated CB[5].1. We were able to isolate free CB[10] by treatment of CB[10].1 with acetic anhydride followed by washing with MeOH, DMSO, and water. The spacious cavity of CB[10] is able to complex large guests, including a cationic calix[4]arene derivative in its 1,3-alternate form (CB[10].1,3-alt-3). The addition of adamantane carboxylic acid (4) to CB[10].3 triggers a conformational change during the formation of termolecular complex CB[10].cone-3.4.

Supramolecular Controlled Cargo Release via Near Infrared Tunable Cucurbit[7]uril-Gold Nanostars.

Project description:The near infrared (NIR) absorption and average particle size of gold nanostars (GNSs) can be precisely controlled by varying the molar ratios of cucurbit[7]urils (CB[7]) and GNSs in aqueous solution. GNSs modified with CB[7] achieved high cargo loading with thermally activated release upon the NIR laser irradiation.

Binding Studies of Cucurbit[7]uril with Gold Nanoparticles Bearing Different Surface Functionalities.

Project description:Host-guest interactions between a synthetic receptor, cucurbit[7]uril (CB[7]), and gold nanoparticles (AuNPs) have been quantified using isothermal titration calorimetry. AuNPs were functionalized with ligands containing tertiary or quaternary benzylamine derivatives, with electron donating or withdrawing groups at the para position of the benzene ring. Analysis of binding interactions reveals that functional groups at the para position have no significant effect on binding constant. However, headgroups bearing a permanent positive charge increased the binding of AuNPs to CB[7] ten-fold compared to monomethyl counterparts.

4-Sulfocalix[4]arene/Cucurbit[7]uril-Based Supramolecular Assemblies through the Outer Surface Interactions of Cucurbit[<i>n</i>]uril.

Project description:Upon mixing of aqueous solutions of the freely soluble building blocks cucurbit[7]uril (Q[7]) and 4-sulfocalix[4]arene (SC[4]A), white microcrystals instantly separate in near-quantitative yield. The driving force for this assembly is suggested to be the outer-surface interaction of the Q[n]. Dynamic light scattering, scanning electron microscopy, and NMR (diffusion-ordered NMR spectroscopy) analyses have confirmed the supramolecular aggregation of Q[7] and SC[4]A. Titration 1H NMR spectroscopy and isothermal titration calorimetry have shown that the interaction ratio of Q[7] and SC[4]A is close to 3:1. Moreover, the Q[7]/SC[4]A-based supramolecular assembly can accommodate molecules of some volatile compounds or luminescent dyes. Thus, this work offers a simple and highly efficient means of preparing adsorbent or solid fluorescent materials.

Acyclic Cucurbit[n]uril Dendrimers.

Project description:The synthesis of acyclic cucurbit[n]uril dendrimers G1-G3 that bear four dendrons on their aromatic sidewalls via thiolate S(N)2 chemistry is reported. G1-G3 are polycationic and can bind to pEGFP plasmid DNA as shown by dynamic light scattering (DLS), gel electrophoresis, and scanning electron microscopy (SEM). The gene delivery ability of G1-G3 is presented.

Injectable Cucurbit[8]uril-Based Supramolecular Gelatin Hydrogels for Cell Encapsulation.

Project description:Recent efforts to develop hydrogel biomaterials have focused on better recapitulating the dynamic properties of the native extracellular matrix. In hydrogel biomaterials, binding thermodynamics and cross-link kinetics directly affect numerous bulk dynamic properties such as strength, stress relaxation, and material clearance. However, despite the broad range of bulk dynamic properties observed in biological tissues, present strategies to incorporate dynamic linkages in cell-encapsulating hydrogels rely on a relatively small number of dynamic covalent chemical reactions and host-guest interactions. To expand this toolkit, we report the preparation of supramolecular gelatin hydrogels with cucurbit[8]uril (CB[8])-based cross-links that form on demand via thiol-ene reactions between preassembled CB[8]·FGGC peptide ternary complexes and grafted norbornenes. Human fibroblast cells encapsulated within these optically transparent, shear thinning, injectable hydrogels remained highly viable and exhibited a well-spread morphology in culture. These CB[8]-based gelatin hydrogels are anticipated to be useful in applications ranging from bioprinting to cell and drug delivery.

Solubilization of Pyridone-Based Fluorescent Tag by Complexation in Cucurbit[7]uril.

Project description:Aimed at further exploring the hosting properties of cucurbit[7]uril (CB7), we have exploited the spectroscopic and photophysical properties of a known fluorescent label as the guest molecule, namely, 3-cyano-6-(2-thienyl)-4-trifluoromethyl pyridine (TFP), in neat solvents. The formation of an inclusion host-guest complex with CB7 was checked by UV-vis absorption spectroscopy, and the value of binding constant (9.7 × 105 M-1) was extracted from the spectrophotometric data. The modulation of keto-enol equilibrium in TFP by the local environment is governed by the interplay between dimerization through intermolecular hydrogen bonding between individual solute molecules, favoring the enol form, and intermolecular hydrogen bonding between TFP and the surrounding solvents, favoring the keto form. Time-resolved fluorescence results established that the macromolecular CB7 host stabilizes preferentially the neutral enol form over the keto form of TFP. Unprecedentedly, our results reveal a linear dependence of the amplitudes of the extracted decay-associated spectra from the time-resolved fluorescence spectra of TFP on the sum of polarity/polarizability and hydrogen bonding parameters of the local environment, confirming that TFP at micromolar concentration in the CB7 complexes is experiencing a methanol-like environment. The results rationalized the 42-fold enhancement in the solubility of TFP in water media by complexation in CB7.

Supramolecular Fluorescence Probe Based on Twisted Cucurbit[14]uril for Sensing Fungicide Flusilazole.

Project description:The host-guest complex of the common dye, thioflavin T (ThT), and twisted cucurbit[14]uril (tQ[14]) was selected as a fluorescent probe to determine non-fluorescent triazole fungicides, including flusilazole, azaconazole, triadimefon, tebuconazole, tricyclazole, flutriafol, penconazole, and triadimenol isomer A, in an aqueous solution. The experimental results reveal that the ThT@tQ[14] probe selectively responded to flusilazole with significant fluorescence quenching and a detection limit of 1.27 × 10-8 mol/L. In addition, the response mechanism involves not only a cooperation interaction-ThT occupies a side-cavity of the tQ[14] host and the triazole fungicide occupies another side-cavity of the tQ[14] host-but also a competition interaction in which both ThT and the triazole fungicide occupy the side-cavities of the tQ[14] host.

Enhanced Energy Conversion of Z907-Based Solar Cells by Cucurbit[7]uril Macrocycles.

Project description:A dye-sensitized solar cell was constructed on the basis of encapsulating the ruthenium polypyridyl photosensitizer Z907 in the macrocycle cucurbit[7]uril (CB7). The work focuses on the photophysical properties of the new host-guest complexes in acetonitrile and water (volume ratio 1:9) and on the top of nanocrystalline titanium dioxide (TiO2) electrode prior to the addition of poly(3-hexylthiophene) polymer and gold electrode. Complexation to CB7 in aqueous solutions has decreased the emission intensity and excited-state lifetime for metal-to-ligand charge transfer (MLCT) state at 650 nm by twofold because of collisional quenching, which opens a non-radiative deactivation channel. Similarly, a twofold decrease in the emission intensity and excited-state lifetime of MLCT at 750 nm on the top of TiO2 electrodes was observed with the addition of CB7. Encapsulation of Z907 dye to CB7 host has, also, led to fourfold enhancement in the short circuit current and power conversion efficiency of the final solar cell. The results support the premise that host-guest complexation of CB7 facilitates faster electron injection from Z907 dye into the conduction band of TiO2 electrodes.
