To demonstrate the potential of the heterocyclic moiety for diversity oriented synthesis, a series of (-)-julocrotine analogues was synthesized by employing the heterocyclic precursor as an amino input in Ugi four-component reactions (Ugi-4CR) [1].

Investigation of metal mobility in gold and silver mine tailings by single-step and sequential extractions.

Project description:Metal leachate from mine tailings has the potential to release toxic metals into the surrounding environment. A single-step extraction procedure mimicking rainwater and a three-step BCR sequential extraction procedure (acid, reducing and oxidizing conditions) were applied to gold (GMT) and silver (SMT) mine tailings. Major (Al, Ca, Fe, Mg, and Mn) and trace metals were monitored to better understand the mobility and geochemistry of these metals when exposed to various environmental leaching conditions. Rainwater extraction released only small quantities of metals, while the three-step BCR extraction was more effective in mobilizing metals from the tailings. Under the acidic conditions of BCR step 1, Ca, Mg, Cd, Cu, and Mn were released in high concentrations. The dissolution of Fe, Ca, and Mg were dominant along with Pb in step 2 (reducing conditions). In step 3 (oxidizing conditions), Fe was the most dominant species together with Co, Cu, Ni, and Se. A high fraction of Al, Be, Cr, Li, Mo, Sb, Tl, and V remained in the residue. From SMT, larger quantities of As, Ca, Cd, and Zn were released compared to GMT. The BCR extraction could be applied to tailings to predict the potential release of toxic metals from mine wastes; however, excessive amounts of Ca and Fe in the tailings could cause carry-overs and incomplete extraction and carry-overs, resulting in a misinterpretation of results.

Diverse Isoquinoline Scaffolds by Ugi/Pomeranz-Fritsch and Ugi/Schlittler-Muller Reactions.

Project description:The Pomeranz-Fritsch reaction and its Schlittler-Müller modification were successfully applied in the Ugi postcyclization strategy by using orthogonally protected aminoacetaldehyde diethyl acetal and complementary electron rich building blocks. Several scaffolds, including isoquinolines, carboline, alkaloid-like tetrazole-fused tetracyclic compounds, and benzo[ d]azepinone scaffolds, were synthesized in generally moderate to good yield. All our syntheses provide a short MCR-based sequence to novel or otherwise difficult to access scaffolds. Hence, we foresee multiple applications of these synthesis technologies.

Raising the Diversity of Ugi Reactions Through Selective Alkylations and Allylations of Ugi Adducts.

Project description:We report here selective Tsuji-Trost type allylation of Ugi adducts using a strategy based on the enhanced nucleophilicity of amide dianions. Ugi adducts derived from aromatic aldehydes were easily allylated at their peptidyl position with allyl acetate in the presence of palladium catalysts. These substitutions were compared to more classical transition metal free allylations using allyl bromides.

An application of this extension of the Ugi reaction to the synthesis of ?,?-divinyl amino acids that may be cyclized via ring-closing metathesis to provide highly substituted pyrrolidines is described.

Synthesis of acylhydrazino-peptomers, a new class of peptidomimetics, by consecutive Ugi and hydrazino-Ugi reactions.

Project description:Herein we describe a versatile approach for the synthesis of acylhydrazino-peptomers, a new class of peptidomimetics. The key idea in this approach is based on a simple route using a one-pot hydrazino-Ugi four-component reaction followed by a hydrazinolysis or hydrolysis reaction and subsequent hydrazino-Ugi reaction or classical Ugi reaction for the construction of acyclic acylhydrazino-peptomers. The consecutive multicomponent reactions produced a variety of acylhydrazino-peptomers in moderate to excellent yields (47-90%). These compounds are multifunctional intermediates that can be further functionalized to obtain new peptidomimetics with potential biological activity.

Highly Stereoselective Ugi/Pictet-Spengler Sequence.

Project description:Discovering novel synthetic routes for rigid nitrogen-containing polyheterocycles using sustainable, atom-economical, and efficient (= short) synthetic pathways is of high interest in organic chemistry. Here, we describe an operationally simple and short synthesis of the privileged scaffold dihydropyrrolo[1,2-a]pyrazine-dione from readily accessible starting materials. The alkaloid-type polycyclic scaffold with potential bioactivity was achieved by a multicomponent reaction (MCR)-based protocol via a Ugi four-component reaction and Pictet-Spengler sequence under different conditions, yielding a diverse library of products.

Artificial Macrocycles by Ugi Reaction and Passerini Ring Closure.

Project description:Artificial macrocycles can be convergently synthesized by a sequence of an Ugi multicomponent reaction (MCR) followed by an intramolecular Passerini MCR used to close the macrocycle. Significantly, in this work, the first intramolecular macrocyclization through a Passerini reaction is described. We describe 21 macrocycles of a size of 15-20. The resulting macrocyclic depsipeptides are model compounds for natural products and could find applications in drug discovery.

Unimolecular Exciplexes by Ugi Four-Component Reaction.

Project description:Exciplex or excited complex emission is an excited state process, arising from considerable charge transfer of an excited energy donor to an acceptor, which can be identified by the occurrence of a redshifted emission band that is absent in the individual constituents. Particularly interesting are exciplexes that are formed by intramolecular excited state interaction, which are inherently concentration independent. Based upon our previous experience in the Ugi-4CR syntheses of donor-acceptor conjugates capable of photo-induced intramolecular electron transfer (PIET), that is, generation of light-induced charge separation, we now disclose the diversity-oriented approach on unimolecular exciplex emitters and their reference systems by Ugi-4CR. The photophysics is studied by absorption and emission spectroscopy and accompanied by density functional theory (DFT) and time-dependent density functional theory (TDDFT) calculations.
