Recent Electrochemical/Electrical Microfabricated Sensor Devices for Ionic and Polyionic Analytes.

ABSTRACT: The recent technological advances combined with the development of new concepts and strategies have revolutionized the field of sensor devices, allowing access to increasingly sophisticated device structures associated with high sensitivities and selectivities. Among them, electrochemical and electrical sensors have gained the most interest because they offer unique intrinsic characteristics and meet the requirements to be integrated in more sophisticated devices including microfluidics or lab-on-chips, opening access to multiplex and all-in-one detection devices. In the present article, we outline and provide a short and concise overview on the most recent achievements in the field of electrical detection of ionic species as they display versatile roles in many important biological events and are ubiquitous in environment.

Photochemical functionalization of polymer surfaces for microfabricated devices.

Project description:Herein we report the topochemical modification of polymer surfaces with perfluorinated aromatic azides. The aryl azides, which have quaternary amine or aldehyde functional groups, were linked to the surface of the polymer by UV irradiation. The polymer substrates used in this study were cyclic olefin copolymer and poly(methyl methacrylate). These substrates were characterized before and after modification using reflection-absorption infrared spectroscopy, sessile water contact angle measurements, and X-ray photoelectron spectroscopy. Analysis of the surface confirmed the presence of aromatic groups with aldehyde or quaternary amine functionality. Enzyme immobilization and patterning onto polymer surfaces were studied using confocal microscopy. Enzymatic digests of protein were carried out on modified probes manufactured from thermoplastic substrates, and the resulting peptide analysis was completed using matrix-assisted laser desorption/ionization mass spectrometry. The use of functionalized perfluorinated aromatic azides allows the surface chemistry of thermoplastics to be tailored for specific lab-on-a-chip applications.

An Electrochemical Amperometric Ethylene Sensor with Solid Polymer Electrolyte Based on Ionic Liquid.

Project description:An electrochemical amperometric ethylene sensor with solid polymer electrolyte (SPE) and semi-planar three electrode topology involving a working, pseudoreference, and counter electrode is presented. The polymer electrolyte is based on the ionic liquid 1-butyl 3-methylimidazolium bis(trifluoromethylsulfonyl)imide [BMIM][NTf2] immobilized in a poly(vinylidene fluoride) matrix. An innovative aerosol-jet printing technique was used to deposit the gold working electrode (WE) on the solid polymer electrolyte layer to make a unique electrochemical active SPE/WE interface. The analyte, gaseous ethylene, was detected by oxidation at 800 mV vs. the platinum pseudoreference electrode. The sensor parameters such as sensitivity, response/recovery time, repeatability, hysteresis, and limits of detection and quantification were determined and their relation to the morphology and microstructure of the SPE/WE interface examined. The use of additive printing techniques for sensor preparation demonstrates the potential of polymer electrolytes with respect to the mass production of printed electrochemical gas sensors.

Methane-oxygen electrochemical coupling in an ionic liquid: a robust sensor for simultaneous quantification.

Project description:Current sensor devices for the detection of methane or natural gas emission are either expensive and have high power requirements or fail to provide a rapid response. This report describes an electrochemical methane sensor utilizing a non-volatile and conductive pyrrolidinium-based ionic liquid (IL) electrolyte and an innovative internal standard method for methane and oxygen dual-gas detection with high sensitivity, selectivity, and stability. At a platinum electrode in bis(trifluoromethylsulfonyl)imide (NTf2)-based ILs, methane is electro-oxidized to produce CO2 and water when an oxygen reduction process is included. The in situ generated CO2 arising from methane oxidation was shown to provide an excellent internal standard for quantification of the electrochemical oxygen sensor signal. The simultaneous quantification of both methane and oxygen in real time strengthens the reliability of the measurements by cross-validation of two ambient gases occurring within a single sample matrix and allows for the elimination of several types of random and systematic errors in the detection. We have also validated this IL-based methane sensor employing both conventional solid macroelectrodes and flexible microfabricated electrodes using single- and double-potential step chronoamperometry.

Electrical, electrochemical and structural studies of a chlorine-derived ionic liquid-based polymer gel electrolyte.

Project description:In the present article, an ionic liquid-based polymer gel electrolyte was synthesized by using poly(vinylidene fluoride-co-hexafluoropropylene) (PVdF-HFP) as a host polymer. The electrolyte films were synthesized by using the solution casting technique. The as-prepared films were free-standing and transparent with good dimensional stability. Optimized electrolyte films exhibit a maximum room-temperature ionic conductivity of σ = 8.9 × 10-3 S·cm-1. The temperature dependence of the prepared polymer gel electrolytes follows the thermally activated behavior of the Vogel-Tammann-Fulcher equation. The total ionic transference number was ≈0.91 with a wider electrochemical potential window of 4.0 V for the prepared electrolyte film which contains 30 wt % of the ionic liquid. The optimized films have good potential to be used as electrolyte materials for energy storage applications.

Hydrodynamic Characterization of Phase Separation in Devices with Microfabricated Capillaries.

Project description:Capillary microseparators have been gaining interest in downstream unit operations, especially for pharmaceutical, space, and nuclear applications, offering efficient separation of two-phase flows. In this work, a detailed analysis of the dynamics of gas?liquid separation at the single meniscus level helped to formulate a model to map the operability region of microseparation devices. A water?nitrogen segmented flow was separated in a microfabricated silicon-glass device, with a main channel (width, W = 600 ?m; height, H = 120 ?m) leading into an array of 276 capillaries (100 ?m long; width = 5 ?m facing the main channel and 25 ?m facing the liquid outlet), on both sides of the channel. At optimal pressure differences, the wetting phase (water) flowed through the capillaries into the liquid outlet, whereas the nonwetting phase (nitrogen) flowed past the capillaries into the gas outlet. A high-speed imaging methodology aided by computational analysis was used to quantify the length of the liquid slugs and their positions in the separation zone. It was observed that during stable separation, the position of the leading edge of the liquid slugs (advancing meniscus), which became stationary in the separation zone, was dependent only on the outlet pressure difference. The trailing edge of the liquid slugs (receding meniscus) approached the advancing meniscus at a constant speed, thus leading to a linear decrease of the liquid slug length. Close to the liquid-to-gas breakthrough point, that is, when water exited through the gas outlet, the advancing meniscus was no longer stationary, and the slug lengths decreased exponentially. The rates of decrease of the liquid slug length during separation were accurately estimated by the model, and the calculated liquid-to-gas breakthrough pressures agreed with experimental measurements.

Project description:We review the recent progress in the emerging area of devices and circuits operating on the basis of ionic currents. These devices operate at the intersection of electrochemistry, electronics, and microfluidics, and their potential applications are inspired by essential biological processes such as neural transmission. Ionic current rectification has been demonstrated in diode-like devices containing electrolyte solutions, hydrogel, or hydrated nanofilms. More complex functions have been realized in ionic current based transistors, solar cells, and switching memory devices. Microfluidic channels and networks-an intrinsic component of the ionic devices-could play the role of wires and circuits in conventional electronics.

Ionic liquid stationary phase coating optimization for semi-packed microfabricated columns.

Project description:This work highlights the effect of the stationary phase coating process on the separation efficiency of gas chromatography microcolumns. The stationary phase coating quality was characterized by three different bis(trifluoromethylsulfonyl)imide (NTf2) anion based ionic liquids. The ionic liquids containing NTf2 anion are used for gas chromatography due to their high temperature stability. In this work, the chemical and physical approaches of column deactivation as well as the temperature treatment were evaluated by separating a mixture of 20 organic components and saturated alkanes. The results show that higher oven temperature treatment provides higher efficiency while losing a bit of peak symmetry. The thermal treated 1-butylpyridinum bis(trifluoromethylsulfonyl) imide [BPY][NTf2] stationary phase at 240°C demonstrated as high as 8300 plates per meter for naphthalene. This was a 5-fold increase in separation efficiency in comparison to those of the columns treated at 200°C. Albeit being within acceptable ranges, the peak tailing degraded from 1.17 to 1.46 for naphthalene when the processing temperature for coating increased. Both chemical and physical deactivation process increased separation efficiencies and peak resolution.

Microfabricated valveless devices for thermal bioreactions based on diffusion-limited evaporation.

Project description:Microfluidic devices that reduce evaporative loss during thermal bioreactions such as PCR without microvalves have been developed by relying on the principle of diffusion-limited evaporation. Both theoretical and experimental results demonstrate that the sample evaporative loss can be reduced by more than 20 times using long narrow diffusion channels on both sides of the reaction region. In order to further suppress the evaporation, the driving force for liquid evaporation is reduced by two additional techniques: decreasing the interfacial temperature using thermal isolation and reducing the vapor concentration gradient by replenishing water vapor in the diffusion channels. Both thermal isolation and vapor replenishment techniques can limit the sample evaporative loss to approximately 1% of the reaction content.

Pt-Decorated MWCNTs-Ionic Liquid Composite-Based Hydrogen Peroxide Sensor To Study Microbial Metabolism Using Scanning Electrochemical Microscopy.

Project description:Hydrogen peroxide (H2O2) is a highly relevant metabolite in many biological processes, including the oral microbiome. To study this metabolite, we developed a 25 ?m diameter, highly sensitive, nonenzymatic H2O2 sensor with a detection limit of 250 nM and a broad linear range of 250 nM to 7 mM. The sensor used the synergistic activity of the catalytically active Pt nanoparticles on a high surface area multiwalled carbon nanotube and conducting ionic liquid matrix to achieve high sensitivity (2.4 ± 0.24 mA cm-2 mM-1) for H2O2 oxidation. The unique composite allowed us to miniaturize the sensor and couple it with a Pt electrode (25 ?m diameter each) for use as a dual scanning electrochemical microscopy probe. We could detect 65 ± 10 ?M H2O2 produced by Streptococcus gordonii (Sg) in a simulated biofilm at 50 ?m above its surface in the presence of 1 mM glucose and artificial saliva solution (pH 7.2 at 37 °C). Because of its high stability and low detection limit, the sensor showed a promising chemical image of H2O2 produced by Sg biofilms. We were also able to detect 30 ?M H2O2 at 50 ?m above the biofilm in the presence of the H2O2-decomposing salivary lactoperoxidase and thiocyanate, which would not otherwise be possible using an existing H2O2 assay. Thus, this sensor can potentially find applications in the study of other important biological processes in a complex matrix where circumstances demand a low detection limit in a compact space.

Microfabricated polyacrylamide devices for the controlled culture of growing cells and developing organisms.

Project description:The ability to spatially confine living cells or small organisms while dynamically controlling their aqueous environment is important for a host of microscopy applications. Here, we show how polyacrylamide layers can be patterned to construct simple microfluidic devices for this purpose. We find that polyacrylamide gels can be molded like PDMS into micron-scale structures that can enclose organisms, while being permeable to liquids, and transparent to allow for microscopic observation. We present a range of chemostat-like devices to observe bacterial and yeast growth, and C. elegans nematode development. The devices can integrate PDMS layers and allow for temporal control of nutrient conditions and the presence of drugs on a minute timescale. We show how spatial confinement of motile C. elegans enables for time-lapse microscopy in a parallel fashion.
