Complete mitochondrial genome of Stictochironomus akizukii (Tokunaga) (Chironomidae, Diptera) assembled from next-generation sequencing data.

ABSTRACT: The complete mitochondrial genome of Stictochironomus akizukii (Tokunaga) was sequenced. The circular mitochondrial genome is 15,052 bp and consists of 13 protein-coding, 3 ribosomal RNAs, and 22 transfer RNA genes (GenBank accession no. MT185679). Results of maximum likelihood analysis showed that this species clustered with other species of the family Chironomidae. This study will contribute to the phylogenetics of genus Stictochironomus and the other genera of Chironomidae.

The circularized mitochondrial genome spans 15,306 bp with a high A + T content of 74.7% and consists of 13 protein-coding genes, 22 tRNA genes, and 2 rRNA genes. Its gene composition and order were similar to Anax imperator. The overall base composition is 36.6% for A, 34.8% for T, 16.8% for C, and 11.8% for G. A phylogeny of 31 dragonfly species clustered this species within Odonata.

The Bayesian comparative analysis of molecular evolution rates revealed no acceleration of the mitochondrial DNA (mtDNA) evolution of P. magnifica. Results of maximum likelihood analysis showed that this species clustered with other species of the phylum Bryozoa.

Complete mitochondrial genome of Limnophyes minimus (Diptera: Chironomidae).

Project description:The complete mitochondrial genome of Limnophyes minimus (Meigen 1818) was sequenced and annotated, and its general features and base composition were analyzed. The phylogenetic relationships of the families Chironomidae, Simuliidae, Sciaridae and Culicidae based on 25 metagenomes were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that L. minimus belongs family Chironomidae, which is consistent with the traditional morphological classification.

Complete mitochondrial genome of Dacus trimacula (Diptera: Tephritidae) using next-generation sequencing from China.

Project description:The complete mitochondrial genome (mitogenome) of the fruit fly species Dacus trimacula (Diptera: Tephritidae: Dacinae) are sequenced and annotated. The mitochondrial genome is 15,851?bp (GenBank No. MK940811) has an A?+?T content of 72.8% (A 39.2%; C 17.0%; G 10.2%, and T 33.6%), which is the classical structure for insect mitogenome. All PCGs started with ATN and stopped with TAN. The phylogenetic tree confirms that D. trimacula clustered with D. longicornis as the sister group to Zeugodacus. This study enriches the mitogenomes of the fruit flies.

Project description:Odonata (dragonflies and damselflies) is a small order at the base of flying insects (Pterygota). Resolving family-level phylogenetic relationships within this order receives great attention. Hereby, genetic data already resulted in various changes, which are however still under discussion. Mitochondrial genomes may further enhance such phylogenies. This study presents the complete mitochondrial genome of the Neotropical damselfly Megaloprepus caerulatus based on next generation sequencing (NGS) data on total genomic DNA. The total length comprises 16,094?bp and includes the standard metazoan set of 37 genes together with a 1376?bp long A?+?T rich (control) region. Gene content, gene arrangement and base frequency are consistent with other odonate mitochondrial genomes. It further contains four intergenic spacer regions, indicating a possible family specific feature for the Coenagrionidae and its close relatives.

Project description:Propsilocerus akamusi is one of the most important genera of Chironomidae. However, the systemically classification and taxonomic studies have so far been limited. In this study, we report the complete mitochondrial genome sequence of P. akamusi. The mitogenome has 15,632 base pairs (77.58% A + T content) and made up of total of 37 genes (13 protein-coding, 22 transfer RNAs, and 2 ribosomal RNAs). This study will provide useful genetic information for future phylogenetic and taxonomic classification of Chironomidae.

The first complete mitochondrial genome of Dacus longicornis (Diptera: Tephritidae) using next-generation sequencing and mitochondrial genome phylogeny of Dacini tribe.

Project description:The genus Dacus is one of the most economically important tephritid fruit flies. The first complete mitochondrial genome (mitogenome) of Dacus species - D. longicornis was sequenced by next-generation sequencing in order to develop the mitogenome data for this genus. The circular 16,253?bp mitogenome is the typical set and arrangement of 37 genes present in the ancestral insect. The mitogenome data of D. longicornis was compared to all the published homologous sequences of other tephritid species. We discovered the subgenera Bactrocera, Daculus and Tetradacus differed from the subgenus Zeugodacus, the genera Dacus, Ceratitis and Procecidochares in the possession of TA instead of TAA stop codon for COI gene. There is a possibility that the TA stop codon in COI is the synapomorphy in Bactrocera group in the genus Bactrocera comparing with other Tephritidae species. Phylogenetic analyses based on the mitogenome data from Tephritidae were inferred by Bayesian and Maximum-likelihood methods, strongly supported the sister relationship between Zeugodacus and Dacus.

Project description:Endornaviruses have large double-stranded RNA (dsRNA) genomes that carry a single open reading frame (ORF). Here we report the complete genome of a novel endornavirus, assembled from next-generation sequence data generated from Vitis vinifera-extracted dsRNA. Two different fungal hosts have been identified for this virus, suggesting that horizontal transmission of the virus is possible.

This study is helpful to the identification of C. flaviplumus larvae, which is difficult to be identified by morphology.
