Data for the lipase catalyzed synthesis of cyano-containing multi-substituted indoles.

ABSTRACT: The data presented here are related to the research paper entitled "Efficient Synthesis of Cyano-containing Multi-substituted Indoles Catalyzed by Lipase" [1]. In this data article, the lipase catalyzed synthetic procedures for the preparation of multi-substituted indoles and their derivatives were described. In total, 11 compounds were obtained and the optimum pH, reaction time and substrate ratio were screened through this study.

The reaction proceeds with up to 87% yield, 99:1 er, and >20:1 dr. Evaluation of several of the compounds revealed promising anticancer activity against MCF-7 cells.

Regioselective Mercury(I)/Palladium(II)-Catalyzed Single-Step Approach for the Synthesis of Imines and 2-Substituted Indoles.

Project description:An efficient synthesis of ketimines was achieved through a regioselective Hg(I)-catalyzed hydroamination of terminal acetylenes in the presence of anilines. The Pd(II)-catalyzed cyclization of these imines into the 2-substituted indoles was satisfactorily carried out by a C-H activation. In a single-step approach, a variety of 2-substituted indoles were also generated via a Hg(I)/Pd(II)-catalyzed, one-pot, two-step process, starting from anilines and terminal acetylenes. The arylacetylenes proved to be more effective than the alkyl derivatives.

Multicomponent assembly of highly substituted indoles by dual palladium-catalyzed coupling reactions.

Project description:Highly substituted indoles were synthesized by a palladium-catalyzed reaction involving three independent components in a one-pot reaction. Two distinct palladium-catalyzed coupling reactions occur with a single catalytic system: a Buchwald-Hartwig reaction and an arene-alkene coupling. Quantum chemical computations provide insight into the mechanism of the latter coupling step.

A palladium-catalyzed Barluenga cross-coupling - reductive cyclization sequence to substituted indoles.

Project description:A short and flexible synthesis of substituted indoles employing two palladium-catalyzed reactions, a Barluenga cross-coupling of p-tosylhydrazones with 2-nitroarylhalides followed by a palladium-catalyzed, carbon monoxide-mediated reductive cyclization has been developed. A one-pot, two-step methodology was further developed, eliminating isolation and purification of the cross-coupling product. This was accomplished by utilizing the initially added 0.025 equivalents of bis(triphenylphosphine)palladium dichloride, thus serving a dual role in the cross-coupling and the reductive cyclization. It was found that addition of 1,3-bis(diphenylphosphino)propane and carbon monoxide after completion of the Barluenga reaction afforded, in most cases, significantly better overall yields.

Synthesis of Cyano-Substituted Conjugated Polymers for Photovoltaic Applications.

Project description:Three conjugated polymers, in which the electron-donating (D) 5-alkylthiophene-2-yl-substitued benzodithiophene was linked to three different electron-accepting (A) moieties, i.e., benzothiadiazole (BT), diphenylquinoxaline (DPQ), and dibenzophenazine (DBP) derivative via thiophene bridge, were synthesized using the Stille coupling reaction. In particular, the strong electron-withdrawing cyano (CN) group was incorporated into the A units BT, DPQ, and DBP to afford three D-A type target polymers PB-BTCN, PB-DPQCN, and PB-DBPCN, respectively. Owing to the significant contribution of the CN-substituent, these polymers exhibit not only low-lying energy levels of both the highest occupied molecular orbital and the lowest unoccupied molecular orbital, but also reduced bandgaps. Furthermore, to investigate the photovoltaic properties of polymers, inverted-type devices with the structure of ITO/ZnO/Polymer:PC71BM/MoO3/Ag were fabricated and analyzed. All the polymer solar cells based on the three cyano-substituted conjugated polymers showed high open-circuit voltages (Voc) greater than 0.89 V, and the highest power conversion efficiency of 4.59% was obtained from the device based on PB-BtCN with a Voc of 0.93 V, short-circuit current of 7.36 mA cm-2, and fill factor of 67.1%.

Palladium-catalyzed enantioselective C-3 allylation of 3-substituted-1H-indoles using trialkylboranes.

Project description:We have developed a new enantioselective C-3 allylation of 3-substituted indoles using allyl alcohol and trialkylboranes. Asymmetric syntheses of 3,3-disubstituted indolines and indolenines in enantiomeric excesses up to 90% have been achieved using the bulky borane 9-BBN-C6H13 as the promoter of the reaction. The dependence of the selectivity on the nature of the borane suggests that the boron reagent has a role beyond promoting ionization of the allyl alcohol. A protocol for oxidation of indolenines to oxindoles has also been developed and led to a formal synthesis of (-)-phenserine.

Project description:A convergent and efficient transition-metal-free catalytic synthesis of 2-aryl-indoles has been developed. The interception of a highly reactive and transient aza-ortho-quinone methide by an acyl anion equivalent generated through N-hetereocyclic carbene catalysis is central to this successful strategy. High yields and a wide scope as well as the streamlined synthesis of a kinase inhibitor are reported.

Project description:A stereospecific palladium-catalyzed N-vinylation of azaheterocycles with vinyl triflates is described. Cyclic and acyclic vinyl triflates along with nonnucleophilic azaheterocycles were found to be substrates for this palladium-catalyzed synthesis of N-vinyl pyrrole and indole derivatives.

Lipase catalyzed synthesis of antimicrobial andrographolide derivatives.

Project description:In this data article we describe screening of various lipases for the regioselective acylation of Andrographolide. Each lipase was screened with seven acyl donors. Amano lipase AK from Pseudomonas fluorescens was used for the synthesis of two new acylated andrographolide derivatives. Two new compounds, andrographolide-14-propionate and andrographolide-14-caproate were characterized by various spectral studies. These two derivatives showed more antimicrobial activity than andrographolide.

One-Pot Modified Madelung Synthesis of 3-Tosyl- and 3-Cyano-1,2-disubstituted Indoles.

Project description:A One-pot, two-step procedure for the synthesis of 1,2-disubstituted-3-tosyl and 1,2-disubstituted-3-cyanoindoles from the corresponding N-(o-tolyl)benzamides is reported. The developed procedure is operationally simple, does not utilize any transition metals, and provides variably substituted indoles in good yields from readily available starting materials.
