Project description:Background:The future supply of platinum group metals (PGM) is at risk because of their scarcity combined with a high demand. Thus recovery of platinum (Pt) from waste is an option worthy of study to help alleviate future shortages. This research explored the microbial reduction of platinum (Pt). The ability of anaerobic granular sludge to reduce Pt(IV) ions under different physiological conditions was studied. Results:X-Ray diffraction (XRD) and transmission electron microscope (TEM) analyses demonstrated the capacity of the microbial mixed culture to reduce Pt(IV) to Pt(0) nanoparticles, which were deposited on the cell-surface and in the periplasmic space. Ethanol supported the biologically catalyzed Pt(IV) reduction, meanwhile other electron donors; hydrogen (H2) and formate, promoted the chemical reduction of Pt(IV) with some additional biological stimulation in the case of H2. A hypothesis is proposed in which H2 formed from the acetogenesis of ethanol is implicated in subsequent abiotic reduction of Pt(IV) indicating an integrated bio-chemical process. Endogenous controls also resulted in slow Pt(IV) removal from aqueous solution. Selected redox mediators, exemplified by riboflavin, enhanced the Pt(IV) reduction rate. Conclusion:This study reported for the first time the ability of an anaerobic granular sludge to reduce Pt(IV) to elemental Pt(0) nanoparticles.

Project description:In the title compound, [Pt(CH(3))(3)I(C(12)H(12)N(2))], the Pt(IV) atom is six-coordinated in a slightly distorted octa-hedral configuration with one CH(3) group and the I atom forming a near perpendicular axis relative to the square plane formed by the bipyridine ligand and the two remaining CH(3) groups. The CH(3) group trans to the I atom has a slightly elongated bond to Pt compared to the other CH(3) groups, indicating a difference in trans influence between iodine and the bipyridine ligand.

The main geometric parameters are Pt-N = 1.960?(5)?Å, and Pt-Cl = 2.3177?(12) and 2.3196?(12)?Å. The N C bond is a typical triple bond [1.137?(7)?Å]. The Pt-N C-C unit is almost linear, with Pt-N-C and N-C-C angles of 174.6?(4) and 177.1?(6)°, respectively.

Project description:Platinum compounds are a vital part of our anti-cancer arsenal, and determining the location and speciation of platinum compounds is crucial. We have synthesised a lanthanide complex bearing a salicylic group (Ln = Gd, Eu) which demonstrates excellent cellular accumulation and minimal cytotoxicity. Derivatisation enabled access to bimetallic lanthanide-platinum(ii) and lanthanide-platinum(iv) complexes. Luminescence from the europium-platinum(iv) system was quenched, and reduction to platinum(ii) with ascorbic acid resulted in a "switch-on" luminescence enhancement. We used diffusion-based 1H NMR spectroscopic methods to quantify cellular accumulation. The gadolinium-platinum(ii) and gadolinium-platinum(iv) complexes demonstrated appreciable cytotoxicity. A longer delay following incubation before cytotoxicity was observed for the gadolinium-platinum(iv) compared to the gadolinium-platinum(ii) complex. Functionalisation with octanoate ligands resulted in enhanced cellular accumulation and an even greater latency in cytotoxicity.

Project description:The high mortality rate of lung cancer patients and the frequent occurrence of side effects during cancer therapy demonstrate the need for more selective and targeted drugs. An important and well-established target for lung cancer treatment is the occasionally mutated epidermal growth factor receptor (EGFR). As platinum(II) drugs are still the most important therapeutics against lung cancer, we synthesized in this study the first platinum(IV) complexes coupled to the EGFR-targeting peptide LARLLT (and the shuffled RTALLL as reference). Notably, HPLC-MS measurements revealed two different peaks with the same molecular mass, which turned out to be a transcyclization reaction in the linker between maleimide and the coupled cysteine moiety. With regard to the EGFR specificity, subsequent biological investigations (3-day viability, 14-day clonogenic assays and platinum uptake) on four different cell lines with different verified EGFR expression levels were performed. Unexpectedly, the results showed neither an enhanced activity nor an EGFR expression-dependent uptake of our new compounds. Consequently, fluorophore-coupled peptides were synthesized to re-evaluate the targeting ability of LARLLT itself. However, also with these molecules, flow cytometry measurements showed no correlation of drug uptake with the EGFR expression levels. Taken together, we successfully synthesized the first platinum(IV) complexes coupled to an EGFR-targeting peptide; however, the biological investigations revealed that LARLLT is not an appropriate peptide for enhancing the specific uptake of small-molecule drugs into EGFR-overexpressing cancer cells.

Project description:In the title complex, [PtCl(4)(C(12)H(8)N(2))]·H(2)O, the Pt(4+) ion is six-coordinated in a distorted octa-hedral environment by two N atoms of a 1,10-phenanthroline ligand and by four Cl atoms. As a result of the different trans effects of the N and Cl atoms, the Pt-Cl bonds trans to the N atom are slightly shorter than those trans to the Cl atom. The compound displays inter-molecular ?-? inter-actions between the six-membered rings, with a centroid-centroid distance of 3.834?Å. There are also weak intra-molecular C-H?Cl hydrogen bonds. According to the IR spectrum, solvent water was present in the crystal, but owing to the high thermal motion of the uncoordinated O atom, the H atoms could not be detected.

Project description:The synthesis, characterization, and cytotoxicity of eight new platinum(IV) complexes having the general formula cis,cis,trans-[Pt(NH(3))(2)Cl(2)(O(2)CNHR)(2)] are reported, where R = tert-butyl (4), cyclopentyl (5), cyclohexyl (6), phenyl (7), p-tolyl (8), p-anisole (9), 4-fluorophenyl (10), or 1-naphthyl (11). These compounds were synthesized by reacting organic isocyanates with the platinum(IV) complex cis,cis,trans-[Pt(NH(3))(2)Cl(2)(OH)(2)]. The electrochemistry of the compounds was investigated by cyclic voltammetry. The aryl carbamate complexes 7-11 exhibit reduction peak potentials near -720 mV vs Ag/AgCl, whereas the alkyl carbamate complexes display reduction peak potentials between -820 and -850 mV vs Ag/AgCl. The cyclic voltammograms of cis,cis,trans-[Pt(NH(3))(2)Cl(2)(O(2)CCH(3))(2)] (1), cis,cis,trans-[Pt(NH(3))(2)Cl(2)(O(2)CCF(3))(2)] (2), and cis-[Pt(NH(3))(2)Cl(4)] (3) were measured for comparison. Density functional theory studies were undertaken to investigate the electronic structures of 1-11 and to determine their adiabatic electron affinities. A linear correlation (R(2) = 0.887) between computed adiabatic electron affinities and measured reduction peak potentials was discovered. The biological activity of 4-11 and, for comparison, cisplatin was evaluated in human lung cancer A549 and normal MRC-5 cells by the MTT assay. The compounds exhibit comparable or slightly better activity than cisplatin against the A549 cells. In MRC-5 cells, all are equally or slightly less cytotoxic than cisplatin, except for 4 and 5, which are more toxic.

Project description:Mono-axial functionalised octahedral diazido Pt(iv) complexes trans, trans, trans-[Pt(py)2(N3)2(OR1)(OR2)] (OR1 = OH and OR2 = anticancer agent coumarin-3 carboxylate (cou, 2a), pyruvate dehydrogenase kinase (PDK) inhibitors 4-phenylbutyrate (PhB, 2b) or dichloroacetate (DCA, 2c)), and their di-axial functionalised analogues with OR1 = DCA and OR2 = cou (3a), PhB (3b), or DCA (3c) have been synthesised and characterised, including the X-ray crystal structures of complexes 2a, 3a, 3b and 3c. These complexes exhibit dark stability and have the potential to generate cytotoxic Pt(ii) species and free radicals selectively in cancer cells when irradiated. Mono-functionalised complexes 2a-2c showed higher aqueous solubility and more negative reduction potentials. Mono- and di-functionalised complexes displayed higher photocytotoxicity with blue light (1 h, 465 nm, 4.8 mW cm-2) than the parent dihydroxido complex 1 (OR1 = OR2 = OH) in A2780 human ovarian (IC50 0.9-2.9 μM for 2a-2c; 0.11-0.39 μM for 3a-3c) and A549 human lung cancer cells (5.4-7.8 μM for 2a-2c; 1.2-2.6 μM for 3a-3c) with satisfactory dark stability. Notably, no apparent dark cytotoxicity was observed in healthy lung MRC-5 fibroblasts for all complexes (IC50 > 20 μM). Significantly higher platinum cellular accumulation and photo-generated ROS levels were observed for the di-functionalised complexes compared with their mono-functionalised analogues when cancer cells were treated under the same concentrations.

Project description:Cisplatin is one of the most widely used anticancer drugs. Its side effects, however, have motivated researchers to search for equally effective analogs that are better tolerated. Selectively targeting cancer tissue is one promising strategy. For this purpose, a platinum(IV) complex was conjugated to the cancer-targeting peptide chlorotoxin (CTX, TM601) in order to deliver cisplatin selectively to cancer cells. The 1:1 Pt-CTX conjugate was characterized by mass spectrometry and gel electrophoresis. Like most platinum(IV) derivatives, the cytotoxicity of the conjugate was lower in cell culture than that of cisplatin, but greater than those of its Pt(IV) precursor and CTX in several cancer cell lines.
