Project description:Temperature-resolved magnetic particle imaging (MPI) represents a promising tool for medical imaging applications. In this study an approach based on a single calibration measurement was applied for highlighting the potential of MPI for monitoring of temperatures during thermal ablation of liver tumors. For this purpose, liver tissue and liver tumor phantoms embedding different superparamagnetic iron oxide nanoparticles (SPION) were prepared, locally heated up to 70?°C and recorded with MPI. Optimal temperature MPI SPIONs and a corresponding linear model for temperature calculation were determined. The temporal and spatial temperature distributions were compared with infrared (IR) camera results yielding quantitative agreements with a mean absolute deviation of 1?°C despite mismatches in boundary areas.

Project description:Super-resolution microscopy has become a powerful tool for biological research. However, its spatial resolution and imaging depth are limited, largely due to background light. Interferometric temporal focusing (ITF) microscopy, which combines structured illumination microscopy and three-photon excitation fluorescence microscopy, can overcome these limitations. Here, we demonstrate ITF microscopy using three-photon excitation fluorescence, which has a spatial resolution of 106 nm at an imaging depth of 100 µm with an excitation wavelength of 1060 nm.

Project description:While the concept of focusing usually applies to the spatial domain, it is equally applicable to the time domain. Real-time imaging of temporal focusing of single ultrashort laser pulses is of great significance in exploring the physics of the space-time duality and finding diverse applications. The drastic changes in the width and intensity of an ultrashort laser pulse during temporal focusing impose a requirement for femtosecond-level exposure to capture the instantaneous light patterns generated in this exquisite phenomenon. Thus far, established ultrafast imaging techniques either struggle to reach the desired exposure time or require repeatable measurements. We have developed single-shot 10-trillion-frame-per-second compressed ultrafast photography (T-CUP), which passively captures dynamic events with 100-fs frame intervals in a single camera exposure. The synergy between compressed sensing and the Radon transformation empowers T-CUP to significantly reduce the number of projections needed for reconstructing a high-quality three-dimensional spatiotemporal datacube. As the only currently available real-time, passive imaging modality with a femtosecond exposure time, T-CUP was used to record the first-ever movie of non-repeatable temporal focusing of a single ultrashort laser pulse in a dynamic scattering medium. T-CUP's unprecedented ability to clearly reveal the complex evolution in the shape, intensity, and width of a temporally focused pulse in a single measurement paves the way for single-shot characterization of ultrashort pulses, experimental investigation of nonlinear light-matter interactions, and real-time wavefront engineering for deep-tissue light focusing.

Project description:Innate lymphoid cells (ILC) play critical roles in regulating immunity, inflammation, and tissue homeostasis in mice. However, limited access to non-diseased human tissues has hindered efforts to profile anatomically-distinct ILCs in humans. Through flow cytometric and transcriptional analyses of lymphoid, mucosal, and metabolic tissues from previously healthy human organ donors, here we have provided a map of human ILC heterogeneity across multiple anatomical sites. In contrast to mice, human ILCs are less strictly compartmentalized and tissue localization selectively impacts ILC distribution in a subset-dependent manner. Tissue-specific distinctions are particularly apparent for ILC1 populations, whose distribution was markedly altered in obesity or aging. Furthermore, the degree of ILC1 population heterogeneity differed substantially in lymphoid versus mucosal sites. Together, these analyses comprise a comprehensive characterization of the spatial and temporal dynamics regulating the anatomical distribution, subset heterogeneity, and functional potential of ILCs in non-diseased human tissues.

Project description:Objectives4D (3D + time) indices of tissue impedance using power Doppler (PD) ultrasound (US) can be measured with spatial-temporal image correlation (STIC) imaging. We wished to evaluate their repeatability and their influence under changes to US machine settings and regional differences within the placenta.MethodsA total of 46 healthy women were recruited at 20-34 weeks of gestation. A total of 9940 3D frames from 644 4D data sets were analysed providing both 3D and 4D indices of vascularity. 4D vs. 3D indices were compared with different machine settings, across the cardiac cycle and in the different placenta regions to assess regional variability.Results3D and 4D indices significantly decreased as wall motion filter (WMF) was increased (P < 0.001). Repeatability decreased as WMF increased (ICC; low1 = 0.80; high1 = 0.60). Indices were significantly lower at the maternal aspect (P = 0.002-0.009) of the placenta and showed less repeatability (ICC; 0.42-0.79) than the fetal aspect (ICC 0.49-0.88). 4D repeatability was good in the central region (ICC 0.80-0.81) but poor in the periphery (ICC 0.45-0.59), while 3D indices were good and comparable between regions (ICC; 0.80 central; 0.81 peripheral).ConclusionsThis study supports the future use of WMF 'low1' and PD to generate more reliable 4D indices values. For 3D indices, HD Flow may improve Doppler signal sensitivity. Regarding placental regional variability, the fetal plate and the central region demonstrated more repeatable 4D indices. 4D PD indices have potential to overcome the limitations of VOCAL™ indices and provide an internally standardised measure of localised impedance in vascular beds.

Project description:Optical methods capable of manipulating neural activity with cellular resolution and millisecond precision in three dimensions will accelerate the pace of neuroscience research. Existing approaches for targeting individual neurons, however, fall short of these requirements. Here we present a new multiphoton photo-excitation method, termed three-dimensional scanless holographic optogenetics with temporal focusing (3D-SHOT), which allows precise, simultaneous photo-activation of arbitrary sets of neurons anywhere within the addressable volume of a microscope. This technique uses point-cloud holography to place multiple copies of a temporally focused disc matching the dimensions of a neuron's cell body. Experiments in cultured cells, brain slices, and in living mice demonstrate single-neuron spatial resolution even when optically targeting randomly distributed groups of neurons in 3D. This approach opens new avenues for mapping and manipulating neural circuits, allowing a real-time, cellular resolution interface to the brain.

Project description:Attention is a multifaceted phenomenon, which operates on features (e.g., colour or motion) and over space. A fundamental question is whether the attentional selection of features is confined to the spatially-attended location or operates independently across the entire visual field (global feature-based attention, GFBA). Studies providing evidence for GFBA often employ feature probes presented at spatially unattended locations, which elicit enhanced brain responses when they match a currently-attended target feature. However, the validity of this interpretation relies on consistent spatial focusing onto the target. If the probe were to temporarily attract spatial attention, the reported effects could reflect transient spatial selection processes, rather than GFBA. Here, using magnetoencephalographic recordings (MEG) in humans, we manipulate the strength and consistency of spatial focusing to the target by increasing the target discrimination difficulty (Experiment 1), and by demarcating the upcoming target's location with a placeholder (Experiment 2), to see if GFBA effects are preserved. We observe that motivating stronger spatial focusing to the target did not diminish the effects of GFBA. Instead, aiding spatial pre-focusing with a placeholder enhanced the feature response at unattended locations. Our findings confirm that feature selection effects measured with spatially-unattended probes reflect a true location-independent neural bias.

Project description:Mental imagery behaviors of various modalities include visual, auditory, and motor behaviors. Their alterations are pathologically involved in various psychiatric disorders. Results of earlier studies suggest that imagery behaviors are correlated with the modulated activities of the respective modality-specific regions and the additional activities of supramodal imagery-related regions. Additionally, despite the availability of complexity analysis in the neuroimaging field, it has not been used for neural decoding approaches. Therefore, we sought to characterize neural oscillation related to multimodal imagery through complexity-based neural decoding. For this study, we modified existing complexity measures to characterize the time evolution of temporal complexity. We took magnetoencephalography (MEG) data of eight healthy subjects as they performed multimodal imagery and non-imagery tasks. The MEG data were decomposed into amplitude and phase of sub-band frequencies by Hilbert-Huang transform. Subsequently, we calculated the complexity values of each reconstructed time series, along with raw data and band power for comparison, and applied these results as inputs to decode visual perception (VP), visual imagery (VI), motor execution (ME), and motor imagery (MI) functions. Consequently, intra-subject decoding with the complexity yielded a characteristic sensitivity map for each task with high decoding accuracy. The map is inverted in the occipital regions between VP and VI and in the central regions between ME and MI. Additionally, replacement of the labels into two classes as imagery and non-imagery also yielded better classification performance and characteristic sensitivity with the complexity. It is particularly interesting that some subjects showed characteristic sensitivities not only in modality-specific regions, but also in supramodal regions. These analyses indicate that two-class and four-class classifications each provided better performance when using complexity than when using raw data or band power as input. When inter-subject decoding was used with the same model, characteristic sensitivity maps were also obtained, although their decoding performance was lower. Results of this study underscore the availability of complexity measures in neural decoding approaches and suggest the possibility of a modality-independent imagery-related mechanism. The use of time evolution of temporal complexity in neural decoding might extend our knowledge of the neural bases of hierarchical functions in the human brain.

Project description:The ability to manipulate acoustic and elastic waveforms in continuous media has attracted significant research interest and is crucial for practical applications ranging from biological imaging to material characterization. Although several spatial focusing techniques have been developed, these systems require sophisticated resonant structures with narrow bandwidth, which limit their practical applications. Here we demonstrate temporal pulse manipulation in a dispersive one-dimensional phononic crystal waveguide, which enables the temporal control of ultrasonic wave propagation. On-chip pulse focusing is realized at a desired time and position with chirped input pulses that agree perfectly with the theoretical prediction. Moreover, traveling four-wave mixing experiments are implemented, providing a platform on which to realize novel nonlinear phenomena in the system. Incorporating this dispersive pulse engineering scheme into nonlinear phononic crystal architecture opens up the possibility of investigating novel phenomena such as phononic solitons.

Project description:Digital optical phase conjugation (DOPC) is a new technique employed in wavefront shaping and phase conjugation for focusing light through or within scattering media such as biological tissues. DOPC is particularly attractive as it intrinsically achieves a high fluence reflectivity in comparison to nonlinear optical approaches. However, the slow refresh rate of liquid crystal spatial light modulators and limitations imposed by computer data transfer speeds have thus far made it difficult for DOPC to achieve a playback latency of shorter than ~200 ms and, therefore, prevented DOPC from being practically applied to thick living samples. In this paper, we report a novel DOPC system that is capable of 5.3 ms playback latency. This speed improvement of almost 2 orders of magnitude is achieved by using a digital micromirror device, field programmable gate array (FPGA) processing, and a single-shot binary phase retrieval technique. With this system, we are able to focus through 2.3 mm living mouse skin with blood flowing through it (decorrelation time ~30 ms) and demonstrate that the focus can be maintained indefinitely-an important technological milestone that has not been previously reported, to the best of our knowledge.
