Randomized controlled study of fast track surgery of colorectal cancer

ABSTRACT: Interventions: A:Without mental intervention, bowel preparation routinely, fasting 12 hours before operation, no thermal insulation during operation, without controlling of infusion volume during operation, placing abdominal drainage tube routinely, using analgesia pump after operation, getting out of bed according to patient’s situation, pulling out the gastric tube and Ingestion of water and food after recovery of intestinal function, pulling out the catheter 3 to 4 days, use antibiotics 3-4days;B:Mental intervention, without bowel preparation, drinking 1000 mL of 10% glucose 12 hours before operation, drinking 400 mL of 10% glucose 3 hours before operation, controlling of infusion volume during operation, placing abdominal drainage tube occasionally, abdominal incision infiltration with 20 to 40 mL of 0.5% ropivacaine immediately and 24 hours after operation, taking 200mg of celecoxib orally two times per day for 3 days, getting out of bed for 1 to 2 hours at night on the operation day, Primary outcome(s): Establish FTS working mode of colorectal cancer Study Design: Randomized parallel controlled trial

Divergent transcriptional response to thermal stress by Anopheles gambiae larvae carrying alternative arrangements of inversion 2La

Project description:shock treatment<br>Differential gene expression between the alternate 2La arrangements was examined for heat shock treated and control fourth instar larvae at three discrete time points to examine temporal variation in thermal stress response. One week after egg hatching, 72 fourth instar larvae of each 2La inversion polmorphism were randomly selected from multiple pans and placed into individual 13x100 mm glass culture tubes containing 2 mL of water. Heat shock treatment was carried out by placing tube racks containing larvae into a 38?C water bath for 60 min. After heat shock, larvae were placed back into 27?C water until collection, which took place 0.25 h, 2 h, and 8 h later by pooling 24 individuals into a 1.5 mL microcentrifuge tube and snap-freezing them in liquid nitrogen. Samples were stored at -80 ?C until RNA isolation. Control groups were treated identically for each time point with the only difference being that they were not exposed to the 60 min heat shock but rather placed into 27?C water<br>

A cell atlas of the adult Drosophila midgut

Project description:Drosophila guts were dissociated to single cells as previously described (Dutta et al., 2015) with a few modifications. Guts were dissected from 7-day old adult esg-sfGFP/+, pros- Gal4>RFP/+ females. After pulling out the gut, the crop and midgut/hindgut junction (where the Malpighian tubules branch out of the gut) and Malpighian tubules were removed. Five flies at a time were dissected and immediately transferred into cold PBS containing 1% BSA to avoid exposing the midgut tissue to room temperature for a long period of time. Once 40 guts were dissected, they were transferred to a dissection plate and chopped into small pieces using a razor blade. These small fragments were immediately transferred to an eppendorf tube containing 400 ul of 1 mg/ml elastase/PBS solution (Sigma E0258) and incubated on a shaker at 27 ̊C for 30 min. 1% of BSA in PBS (final concentration) was used to stop the digestion reaction and prevent cells from aggregating. The cell suspension was passed through 100 um and then a 40 um cell strainer and loaded on the top of Optiprep with density gradient 1.12 g/ml. Viable cells were isolated from the top layer of the sample after centrifugation at 800xg for 20 min. Cell viability and number were assessed by 0.4% trypan blue and hemocytometer.
