Project description:In vitro cell cultures are frequently used to define the molecular background of drug resistance. In this study our major aim was to compare the gene expression signature of 2D and 3D cultured BRAFV600E mutant melanoma cell lines. We successfully developed BRAF-drug resistant cell lines from paired primary/metastatic melanoma cell lines in both 2D and 3D in vitro cultures. Using Affymetrix Human Gene 1.0 ST arrays, we determined the gene expression pattern of all cell lines. Our study highlights gene expression alterations that might help to understand the development of acquired resistance in melanoma cells in tumour tissue.

Project description:Differential gene expression analysis of parental and resistant sub-lines of melanoma cell lines treated or untreated with PLX4032 Using microarray we sought to obtain a genome-wide profile of differentially expressed genes in parental melanoma cell lines and resistant sub-lines in response to PLX4032 vs DMSO control treatment. Representative parental melanoma cell lines and resistant sub-lines were treated with PLX4032 at 1 uM or DMSO control for 6h. Total RNA was extracted and cDNAs were generated and hybridized onto GeneChip Human Gene 1.0 ST Arrays (Affymetrix).

Project description:The ACP02 cell line was stablished from a primary diffuse adenocarcinoma (T3N2M0) and AGP01 cell was established from ascitic fluid cells from intestinal gastric adenocarcinoma (T3N2M1). mRNA samples from cell lines were amplified, labeled, and hybridized to the Affymetrix GeneChip Human Exon 1.0 ST array. Here, we used mouse melanoma cells which have endgeneous Angpt2 expression and invesitgated the functional role of tumor cell-derived Angpt2. Total RNA from control and Angpt2 silenced mouse melanoma cells (RET) was isolated and was further utilized for microarray analysis using Affymetrix Mouse Gene 2.0 ST array.
